The objective of the present study was to evaluate growth and development of corpus luteum (CL) using protein, DNA analysis and PCNA labeling index throughout the estrous cycle in goats. Non pregnant goats (n=20), 12 months of age were assigned for estrus synchronization. Goats CL were obtained from days 3, 8, 13 and 18 of estrous cycle using bilateral ovariectomy and blood samples were taken for plasma progesterone (P4) concentration. The CL was weighed, and samples of each were fixed in Carnoy’s solution. Additional samples of each CL were frozen and stored at -70 °C until analyzed for tissue DNA and protein. Luteal fresh weight and DNA contents increased (P<0.01) from day 3 to day 8, were similar between days 8 and 13, and then decreased (P<0.01)...

The objectives of this study were to determined the variation of quality and shelf life color of pork carcass as affected by leg shackling. Six Largewhite barrows (130 kg) were slaugtered and one side of hind leg was shackled before exsanguination. The carcasses were splitted to two sides (shackled vs non-shackled) and carcass pH at loin and ham were recorded every 1 h for 4 h postmortem. Pork carcasses were aged for 9 days and loin samples were collected to determine for shear force and shelf life color everyday for 7 days. The result showed that pH at loin (Longissimus dorsi muscle) and Ham (Gluteus medius muscle) were similar between shackled and non-shackled side (P>0.05). The shear force of loin samples were similar between treatments. The color...

The study was conducted to determine effect of EB or CS supplements in LAMP reaction on greater accuracy of sexing cattle embryos. Bovine ovaries were collected from the slaughterhouse, oocytes were aspirated from follicles then were evaluated morphology. Oocytes were cultured in TCM-199 at 38.5 ºC 24 h. For in vitro fertilization (IVF), oocytes were co-incubated with spermatozoa at 38.5 ºC 24 h. Fertilization rate was evaluated and then the embryos were in vitro cultured until morula stage. Then, embryo samples were frozen and stored at -196 ºC. Frozen embryos (n=35) were randomly extracted for total cellular DNA and amplified by PCR method. DNA samples were stored at -20 ºC. Sexing was determined by modification of LAMP method with either 1mM EB or 1M CS...

Objective of this study was to determine the effect of stair-step feeding regimen during estrous cycle on follicular development and oocytes quality. Sixteen Thai-native non pregnant goats were used with the average age and body weight of 8 months and 17±0.6 kg, respectively. The does exhibited at least 2 normal estrous cycle and were housed in 1 m2 steel cages. Heat detection was daily recorded using a vasectomized buck. The does were randomly assigned into two groups: Control and Stair-step feeding group. In the control group, does were fed 100% of nutrients requirement for growth in puberty periods. Does consumed Ruzi grass hay, DM = 500 g/head/d and concentrate supplemented at 1% of body weight. Total feed intake was 670 g/head/d, (ME = 1.48 Mcal/kg/d,...
