A comparison of growth and biochemical composition of Mytilus galloprovincialis (Lmk.) and Mytilus edulis (L.) on the West coast of Cotentin, Normandy, France 1999-2000 ArchiMer

The blue mussel, Mytilus edulis has been reared along the Normandy coast line since the 1960s. The gonadal cycle of this mussel species shows a sharp decrease in meat quality during the winter period after spawning. This decline in meat quality is so severe that sales have to be suspended from December to July. Another species of mussel, Mytilus galloprovincialis, which is reared in the lagoons along the French shore of the Mediterranean Sea has a different spawning cycle. L'étude historique de l'ostréiculture française monte que cette activité est caractérisée par des périodes de prospérités et de pénurie. Des épizooties ont été à l'origine de ces alternances. A chacune de ces période, on a pu constater des changements d'espèces : Ostrea edulis fut remplacée par Crassostrea angulata, de même celle-ci fut remplacée par Crassostrea gigas dans les années 1970. Les objectifs pour l'année 1996 étaient les suivants: Poursuivre les premières reproductions des espèces pour lesquelles nous disposons de géniteurs GO. L'objectif est toujours de limiter le stockage d'animaux importés en produisant une descendance simultanément à des témoins C. gigas qui serviront de comparaison. Tenter des hybridations inter-spécifiques des différents taxons présents au sein du conservatoire en fonction des connaissances disponibles dans la littérature sur ce sujet (voir pour revue Gaffney et Allen, 1993). • Importer de nouvelles souches ou espèces. Élargir la gamme des espèces présentes au sein du conservatoire de souches (les capacités d'accueil de la salle de quarantaine étant Limitée).

The Pacific cupped oyster is genetically subdivided into two sister taxa, Crassostrea gigas and C. angulata, which are in contact in the north-western Pacific. The nature and origin of their genetic and taxonomic differentiation remains controversial due the lack of known reproductive barriers and the high degree of morphologic similarity. Development of four EST-SSR multiplex PCRs in the Pacific oyster (Crassostrea gigas) and their validation in parentage assignment ArchiMer

We report four highly informative multiplex PCRs developed from twelve previously described ESTSSRs in Crassostrea gigas. We evaluated and validated these multiplex PCRs in twelve full-sib families. The average allelic richness and the polymorphism information content (PIC) were 11.1 and 0.811 respectively. The combined power of exclusion was greater than 99.99% using all four multiplex assays. A hundred and forty three tests of segregation ratios revealed 11 significant departures from expected Mendelian ratios. The frequency of null alleles was estimated as 4.9% of all the alleles segregating based on a within-family analysis of Mendelian segregation patterns. Etat des stocks conchylicoles normands en 2000 ArchiMer

Crassostrea gigas and Mytilus edulis are reared in four areas along the eastern coast of Cotentin (Normandy). Every five years, a stock assessment survey, based on a stratified random sampling is carried out using aerial photographs. During September 2000, oysters bags and mussel banks random samplings have been weighed on leasing grounds and in random sampled farms in order to evaluate the overall total gross and net weights breeded (oysters 40 000 and mussels 17 000 metric tons, net weights). The European flat oyster Ostrea edulis has been considered as the traditional French oyster for a very long time and has now nearly disappeared from our farms, as a consequence of the two successive epizootic of the Marteilia refringens parasite and Bonamia ostreae. The actual quasi monoculture of Crassostrea gigas has made necessary the search for a diversification in oyster culture, including the search for a new start of the indigenous flat oyster culture. The Ifremer has been conducting parallel works since 1985 in order to better understand the biology of the parasite as well as to try and select different lines of flat oyster resistant to Bonamia ostreae.

In order to study individual reproductive success under controlled conditions, we used microsatellite markers to quantify parental contributions in in vitro crosses (5 males and 5 females) of Crassostrea gigas, the Pacific oyster. Multiplex PCR sets of novel microsatellite loci for the great scallop Pecten maximus and their application in parentage assignment ArchiMer

We report the isolation, development and multiplex optimisation of 12 new microsatellite loci for the great scallop, Pecten maximus. Diversity was moderate to high, with number of alleles ranging from 4 to 20 and observed heterozygosity between 0.28 and 0.88. Progeny produced in a commercial hatchery was used to test locus power for parentage assignment. The percentage of offspring that was unambiguously assigned to a unique pair of parents was 97% (software package CERVUS-COLONY). Parentage assignment revealed that 22% of the studied progeny resulted from unplanned crosses. Effective population size of the study progeny was also estimated. Our study illustrates the power of microsatellites for the genetic monitoring of hatchery-produced great scallops.
