A new tool for induced spawning : the use of 17alpha-hydroxy-20beta-dihydroprogesterone to spawn carp at low temperature Animal Physiology

The efficiency of different hormonal treatments to induce ovulation of carp at low temperatures (13-15°C) was tested. "Priming" with a low dose (0.6 mg/kg) of carp pituitary extract was found necessary for a subsequent successful treatment with 17alpha-hydroxy-20beta-dihydroprogesterone (17alpha-20beta P) (2 mg/kg) 1 day later. The eggs produced by this method showed satisfactory fertilization (75-96%) and hatching success (70%). On the other hand, normal hypophysation (5.4 mg/kg) following priming resulted only in partial ovulation and oocyte resorption at this temperature. Priming was shown to increase the gonadotropin level in plasma from about 2 to 35 ng/ml and to induce germinal vesicle migration toward the periphery of oocytes. Neither...

Action prédominante d'un dérivé hydroxylé de la progestérone sur la maturation in vitro des ovocytes de truite arc-en-ciel, Salmo gairdnerii Animal Physiology

La 17alpha-hydroxy-20 beta-dihydroprogestérone (pregn-4-ene-17 alpha, 20 beta-diol-3-one) induit la maturation in vitro d'ovocytes de truites arc-en-ciel, Salmo gairdnerii, entourés de leur follicule. Avec 97 % de maturations vitellines à la concentration de 22 ng/ml, il représente le stéroïde le plus efficace éprouvé jusqu'à présent. « Progestagène » connu chez les salmonidés, son rôle physiologique possible est discuté.

Androgen and estrogen treatments alter steady state messengers RNA (mRNA) levels of testicular steroidogenic enzymes in the rainbow trout, Oncorhynchus mykiss Animal Physiology

Recent investigations have shown that estrogens have profound inhibitory effects on steroidogenic enzyme gene expressions before and after testicular differentiation in the rainbow trout, Oncorhynchus mykiss. This present study bring new data on juvenile rainbow trout treated with estrogens and androgens. Following a 8 days oral treatment of juvenile male with 17-ethynyl-estradiol (EE2, 20 mg/kg diet) or 11-hydroxyandrostenedione (11OH4, 10 mg/kg diet), we observed a fast and marked decrease of steady-state mRNA levels for 3HSD, P450scc, P450c17, and P450c11 enzymes in the testis. After completion of these treatments, mRNA levels of these enzymes remained low in EE2 treated males whereas in 11OH4 treated males they recovered their initial levels in 8 days....

Les possibilités d'un contrôle de la reproduction des poissons sont basées sur une bonne connaissance de la gamétogenèse, de son contrôle endocrinien et de son déroulement au cours du cycle reproducteur annuel. On examinera le rôle clé joué par le complexe hypothalamo-hypophysaire qui intègre les facteurs de l'environnement et répond en élaborant et sécrétant une ou plusieurs hormones impliquées dans le développement des gonades. Binding of steroids by plasma of a teleost: the rainbow trout, Salmo gairdnerii Animal Physiology

Steroid binding in female trout plasma has been studied. Two thermolabile binding systems are detected by saturation analysis and competitive assays. One is a "sex binding protein" with high affinity for testosterone (K4°C = 3.8 x 10exp8 M-1) and oestradiol-17beta (K4°C = 1.8 x 10exp8 M-1). The second is a "transcortin type': system binding testosterone (K4°C = 3.7 x 10exp7 M-1), progesterone (K4°C = 2.2 x 10exp7 M-1), oestradiol-17beta (K4°C = 1.4 x, 10exp7 M-1) and corticosteroids. Association constants and capacities determined by gel filtration or by equilibrium dialysis are compared. As in other fish, measured capacities are higher than for mammalian binding systems. Characterization of rainbow trout egg quality: a case study using four different breeding protocols, with emphasis on the incidence of embryonic malformations Animal Physiology

The aim of this study was to set up a methodology to accurately evaluate the effects of various husbandry practices on a fish broodstock based on the developmental potential of the egg. For that purpose, long-short photoperiod manipulations (tested twice, PM1 and PM2 groups), spawning induction by injection of a GnRH analog (SI group), and a 16-day post-ovulatory ageing of eggs (POA group) were used in rainbow trout (Oncorhynchus mykiss). Un dosage radioimmunologique spécifique a été utilisé pour suivre la libération de 17 alpha hydroxy - 20 beta dihydroprogestérone par des follicules ovariens entiers de 2 truites arc-en-ciel, au cours de la maturation ovocytaire, induite in vitro par la gonadotropine glycoproteique maturante de salmonidé (s-GtH). Sans stimulation gonadotrope, la production de ce progestagène reste très faible. Elle s'accroît fortement avec l'augmentation des niveaux de s-GtH maintenus dans le milieu. A chacun de ces niveaux, elle est plus importante pour des follicules provenant de la femelle donneuse la plus proche de sa date d'ovulation supposée. Effect of some repetitive factors on turbot stress response Inra

The stress response of the turbot, Scopthalmus maximus (L.), to repetitive factors including netting, air exposure, blood sampling and hand-stripping, were tested using two different tank sizes as well as two blood sampling techniques. Exposure of juvenile fish to air for 1-4 min had no immediate effect on plasma cortisol concentrations or haematocrit values. Similarly, the serial netting of immature fish from tanks did not significantly modify plasma cortisol concentrations, haematocrit or osmolarity values. Hand-stripping of mature males was more disturbing than air exposure. The development of a new sex determination test of Siberian sturgeon based on vitellogenin (VTG) detection in plasma led to identify this complex factor in plasma from male, female and immature fish of this species. The hypothesis of an estrogenic contamination from an environmental origin was confirmed by a test based on the feeding of fish on different diets. In control fish fed with a casein-based diet free of any estrogenic compound, VTG disappeared from the plasma within 15 weeks. With a fish meal-based diet as well as with a soy-based diet, the plasma VTG levels raised significantly. Most of the fish living in the temperate zone exhibit an annual reproductive cycle. Reproduction, or more precisely hatching, occurs when food is available for the fry in the wild. L'évolution des concentrations plasmatiques en hormone gonadotrope glycoprotéique (GTH) et en 17alpha hydroxy-20beta dihydroprogestérone a été suivie au cours de l'ovulation chez 6 truites arc-en-ciel, à l’aide de dosages radio-immunologiques de prélèvements effectués tous les 2 jours. II apparaît un premier accroissement progressif et modéré des niveaux de la GTH avant et pendant l'ovulation (10 à 20 ng/ml), accompagné d'une rapide et forte montée des niveaux de la 17 alpha 20 beta OHP (plusieurs centaines de nanogrammes par millilitre). Changes in gonadal structure and serum levels of sex steroids were investigated during natural sex inversion from female to male in reared populations of the protogynous Mediterranean red porgy, Pagrus pagrus. Four developmental phases were identified by histological observation: female, early transitional (ETr), late transitional (LTr) and male phases. At female phase, a few nests of spermatogonia were observed at the posterior-ventral part of the gonad mainly in females out of the breeding season. Identification and quantification of steroids in the serum of rainbow trout during spermiation and oocyte maturation Animal Physiology

17alpha-Hydroxy-20beta-dihydroprogesterone and 17alpha-hydroxyprogesterone were found in higher concentrations in serum from female Salmo gairdneri undergoing final oocyte maturation immediately before ovulation than in serum from spermiating male trout. Other steroids (11-deoxycorticosterone, 11-deoxycortisol and progesterone) which have been implicated in oocyte maturation and/or ovulation in lower vertebrates were not identified at such high concentrations and the differences between the serum of both sexes were not so great.
