A European epidemiological survey of Vibrio splendidus clade shows unexplored diversity and massive exchange of virulence factors ArchiMer

The Vibrio splendidus clade has previously been associated with epidemic outbreaks of various aquatic animals, as in the case of the cupped oyster, Crassostrea gigas. To investigate whether involved strains could present a clonal origin and to identify possible alternative background carriage animals or zooplankton, a large epidemiological survey was conducted on isolates of the splendidus clade. A field report on vibrio disease of seabass (Dicentrarchus labrax ) in the South of France ArchiMer

Vibrio anguillarum was isolated from diseased seabass reared in aquatic farms (hatcheries and grow-out farms) located along the French Mediterranean coast. A representative strain (V62) differs from the French V. anguillarum strain (V408) by its ability to grow on a 0% salinity medium. The pathogenicity of the representative strain for sea bass was confirmed by injections and bath challenge. The V62 strain reacted with V. anguillarum antiserum (V408) and 84% of the isolated strains from diseased sea bass agglutinated with anti V62 serum.

A 4-year bacteriological survey (2003-2007) of four molluscs cultivated in France and faced with mortality episodes was performed by the French shellfish pathology network. The more abundant bacteria isolated during 92 mortality episodes, occurring mainly in Pacific oyster Crassostrea gigas, were identified by genotyping methods. It allowed us both to confirm the representativeness of Vibrio splendidus and Vibrio aestuarianus bacterial strains and to identify both a large number of Vibrio harveyi-related strains mainly detected during 2007 oyster mortality outbreaks and to a lesser extent bacterial strains identified as Shewanella colwelliana. Oyster diseases caused by pathogenic vibrios pose a major challenge to the sustainability of oyster farming. In France since 2012 a disease affecting specifically adult oysters has been associated with the presence of Vibrio aestuarianus. Here, by combining genome comparison, phylogenetic analyses and high throughput infections of strains isolated before or during the recent outbreaks, we show that virulent strains cluster into two V. aestuarianus lineages independently of the sampling dates. The bacterial lethal dose was not different between strains isolated before or after 2012. Hence the emergence of a new highly virulent clonal strain is unlikely. Herpes-like viruses have been reported last decades in various marine mollusc species in association with mortality outbreaks throughout the world (Renault & Novoa 2004). One of these viruses isolated during French outbreaks has been characterised as an unassigned member of the Herpesviridae family and named ostreid herpes virus 1 (OsHV-1) or Pacific oyster herpes virus (Davison et al. 2005). Comparative study of bactrial infections responsible for mass mortality in penaeid shrimp hatcheries of the Pacific zone ArchiMer

Mass mortality associated with luminescence has been dbserved in sorne Thai and Ecuadorian penaeid shrimp hatcheries in recent years. Samples collected from hatcheries in these two different areas revealed luminescent sucrose-negative Vibrio strains in larVal rearing tanks where mortality occurred. Pathogenicity trials were carried out to identify the causative agents of mortality and to study the course of infection . . Development of an antiseptic procedure to improve retention rates during the grafting process of black-lip oyster, Pinctada margaritifera ArchiMer

In French Polynesia, black pearl cultivation now takes place on about 30 of the 118 atolls or islands. This activity has largely replaced copra, coconut oil and fishing as a source of income. Around 1,000 pearl farms, generally small ones of less than 5 ha, are registered. The pearl farms are mainly owned by families. In 1998, about 7,000 persons were employed in theentire pearl industry, from production to retailing. In value, this activity represents the first exportation of French Polynesian products. From 1988 to date, exports increased exponentially before leveling. Starting with less than two kilograms in 1972, exports have now stabilized at Il tons of "black" pearls.

Ecologically realistic model of infection for exploring the host damage caused by Vibrio aestuarianus ArchiMer

Although vibrios are frequently associated with marine organisms mortality outbreaks, knowledge on their ecology and pathogenicity is sparse, thus limiting disease management and prophylactic strategies. Here, we investigated V. aestuarianus infection onset and progression in the wild, taking advantage of a “claire” pond: a semi‐closed system with limited seawater renewal, theoretically more adapted to disease transmission. We showed a positive association of the bacteria with oysters, which can constitute a reservoir for the bacteria in the winter. Mass mortality outbreaks (90-100%) in both juvenile and adult blue mussels were reported in 2014 along the Pertuis Charentais area of the French Atlantic coast. A series of studies were carried out to learn more about the phenomenon through detecting pathogens, describing environmental conditions and establishing the physiological status of the animals. Bacteria identif ied as belonging to the V. splendidus species were detected in dying mussels and seem able to cause mortality in blue mussels in the laboratory. Unusual environmental conditions (large amounts of fresh water, re-suspension of sediments during a series of storms, and a high renewal rate of specific water masses in Brittany’s Pertuis area) may be partially responsible for sparking the phenomen

Etude du virus de type herpes observé chez les huîtres ArchiMer

Since 1991, sporadic, but high mortality in larvae of the Pacific oyster (Crassostrea gigas) has been observed during the summer in various French hatcheries. This mortality was concomitant with the detection of a virus by transmission electron microscopy. This virus, by virtue of its morphological characteristics and developmental cycle, seems to belong to the Herpesviridae family. The pathogenicity of this viral agent has been demonstrated in the laboratory by experimentally induced mortality on healthy larvae. Moreover, concomitant mortality has been reported in larvae of Crassostrea gigas and Ostrea edulis reared in the same tanks, with detection of a herpes-type virus.
