Cryobanking of fish somatic cells: Optimizations of fin explant culture and fin cell cryopreservation Animal Physiology

When gametes or embryos are not available, somatic cells should be considered for fish genome cryobanking of valuable or endangered fish. The objective of this work was to develop a method for fin explant culture with an assessed reliability, and to assess fin cells ability to cryopreservation. Anal fins from goldfish (Carassius auratus) were minced and gently loosened with collagenase before explants were plated at 20°C in L-15 medium supplemented with fetal bovine serum and pH buffering additives. Quantification of cell donor explants per fin rated the culture success. Cells were successfully obtained from every cultured anal fin (mean = 65% cell-donor explant per fin). La détection des femelles de Salmonidés en vitellogénèse par séro-diagnostic est décrite. Le test utilisé repose sur la mise en évidence de la vitellogénine plasmatique caractéristique des femelles en gamètogénèse active. Son utilisation pratique est simple, rapide et sans risque pour les poissons. Cette technique de terrain permet, entre autres, d'estimer le potentiel reproducteur des populations naturelles et de gérer en pisciculture les stocks de géniteurs. Lors d'inventaires automnaux réalisés par pêche électrique, une technique de sexage (prélèvement sanguin sans sacrifice des animaux) a permis de recenser le nombre de femelles en vitellogénèse dans la population de Truite (Salmo trutta L.) d'une rivière de Bretagne sud, le Scorff. 48 femelles ont été capturées sur une population totale (tous âges confondus) de 460 Truites. La densité de femelles en vitellogénèse par secteur varie de 0,3 à 3 ind./100m2 et apparaît peu reliée aux caractéristiques physiques des divers types d'habitat. Differential expression of two GH receptor mRNAs following temperature change in rainbow trout (Oncorhynchus mykiss) Animal Physiology

The recent genome duplication in salmonids has led to the presence of two Growth Hormone Receptors (GHR). As temperature is a determining factor in fish growth this study aims to determine whether the growth-promoting effect of temperature may be related to a change in GHR gene expression in embryo and juvenile rainbow trout. During embryonic development, using real time PCR, we showed that high temperatures (12°C versus 4°C) increased the amounts of GHR1 transcript up to the hatching stage. By contrast, whatever the stage examined, levels of GHR2 mRNA were unaffected by the incubation temperature. Does the GH/IGF system mediate the effect of water temperature on fish growth? A review Animal Physiology

In fish, as in all poikilotherms, growth is strongly dependent on water temperature. Given that the GH/IGF system regulates growth, it could mediate the effects of temperature on fish growth. Indeed, before hatching, the higher embryonic growth rate in rainbow trout at high temperatures is associated with higher expression of the IGF2 gene in the whole embryo. Furthermore, post-natal growth fluctuations depend on water temperature and are associated with variations of plasma GH and IGF1. Although seasonal parameters such as photoperiod and nutritional status can also affect GH/IGF system activity, it has been shown that an increased temperature led to a specific increase of plasma GH. Des essais réalisés sur une population de Saumon Atlantique de l'Allier, élevée entièrement en eau douce, ont démontré la possibilité d'induire artificiellement l'ovulation avec une avance d'au moins 15 jours sur la date d'ovulation naturelle. Le traitement le plus avantageux (dose minimale de gonadotropine de Saumon partiellement purifiée, G.S.P.P. de 0,1mg/kg) fournissant une réponse rapide est constitué par deux injections successives de G.S.P.P. In fish, the regulation of the growth hormone receptor has been mainly analyzed by binding studies, with some discrepancies in the results. The present work aims at determining whether circulating growth hormone (GH) levels influence the measurement of hepatic GH binding capacities. To do this, the effectiveness of the dissociation of the GH/GH receptor (GHR) complex by an MgCl2 treatment was assessed in rainbow trout and data on GH binding capacities in several physiological situations were studied using different means of expression. Les poissons-chats du genre Harttia se répartissent principalement dans l'est du continent sud-américain : dans les fleuves côtiers du Sud-Est brésilien, dans la région amazonienne et sur le bouclier guyanais. Rapid determination of the sex of puberal salmonid fish by a technique of immunoagglutination Animal Physiology

The present study describes a rapid, simple and specific method to determine the sex of salmonid fish. This method is based on the detection by immunoagglutination of the serum vitellogenin of females in which oocytes are undergoing vitellogenesis. The application of this technique demonstrated the possibility of detecting vitellogenin in female brown trout as early as April and in all female Atlantic salmon captured in Brittany during the fishing season, between March and June.
