A novel natural mutation in HC-Pro responsible for mild symptomatology of Zucchini yellow mosaic virus (ZYMV, Potyvirus) in cucurbits Inra

A natural mild isolate of Zucchini yellow mosaic virus was found to contain a mutation in the helper component (HC-Pro) within a conserved motif, “CDNQLD”, located 12 residues downstream from the “FRNK” motif involved in symptom severity. Introducing the mutation in an infectious cDNA clone of ZYMV resulted in an almost complete absence of symptoms, although viral accumulation was only partially reduced. The FRNK(X)12CDNQLD sequence might be part of a larger motif that is conserved in potyviruses and plays a role in symptomatology and/or silencing inhibition

Deux fractions élicitrices, issues de Phytophthora capsici et partiellement purifiées, G4 et G5 sont appliquées sur organes foliaires en survie de Capsicum annuum et de Cucumis melo. Elles ont pour effet non seulement de réduire l’infection fongique par P. capsici (tabl. 1) ou Colletotrichum lagenarium (tabl. 2), mais encore d’augmenter de façon significative le nombre de lésions locales produites par le virus de la mosaïque du concombre (tabl. 1) ou celui de la criblure du melon (tabl. 2). Sur cotylédons de C. annuum, Lycopersicon esculentum et C. melo en survie, la fraction G4 est très efficace si elle est déposée au site de contamination ou appliquée sur une blessure (tabl. 3). Asymmetrical over-infection as a process of plant virus emergence Inra

Disentangling the role of epidemiological factors in plant pathogen emergences is a prerequisite to identify the most likely future invaders. An example of emergence was recently observed in France: in 10 years, “classic” (CL) strains of Watermelon mosaic virus (WMV) were displaced at a regional scale by newly introduced “emerging” (EM) strains. Here we analyse a 3 years dataset describing the co-dynamics of CL and EM strains at field scale using state-space models estimating jointly: (i) probabilities of primary and secondary infection and (ii) probabilities of over-infecting with a CL [EM] strain a plant already infected with an EM [CL] strain. Results especially indicate that it is more than 3 times less probable for a CL strain to over-infect an EM...

Biological characterization and complete nucleotide sequence of a Tunisian isolate of Moroccan watermelon mosaic virus Inra

During a survey conducted in October 2005, cucurbit leaf samples showing virus-like symptoms were collected from the major cucurbit-growing areas in Tunisia. DAS-ELISA showed the presence of Moroccan watermelon mosaic virus (MWMV, Potyvirus), detected for the first time in Tunisia, in samples from the region of Cap Bon (Northern Tunisia). MWMV isolate TN05-76 (MWMV-Tn) was characterized biologically and its full-length genome sequence was established. MWMV-Tn was found to have biological properties similar to those reported for the MWMV type strain from Morocco. Phylogenetic analysis including the comparison of complete amino-acid sequences of 42 potyviruses confirmed that MWMV-Tn is related (65% amino-acid sequence identity) to Papaya ringspot virus...

Characterisation of a new potyvirus species infecting meadow saffron (Colchicum autumnale) Inra

The alkaloids contained in Colchicum autumnale seeds are used in numerous medicines. Good quality seeds are difficult to obtain from this undomesticated plant. Therefore, a research program was set up aiming to cultivate C. autumnale in order to improve alkaloid contents and seed yields. In this context, a collection was established in 1999 by transplanting corms from twelve different locations in Eastern France. However, serious symptoms of necrosis and decay have appeared in this collection since 2001. Electron microscopic observations of plants showing symptoms revealed the presence of filamentous particles and pinwheel-like structures characteristic of the Potyviridae family. Le dosage immunoenzymatique du virus de la mosaïque du concombre (CMV) dans le melon (Cucumis melo L.) est utilisé pour étudier comparativement la multiplication du virus dans une variété sensible (« Vedrantais ») et une variété résistante (« SC »). Le test ELISA (méthode « sandwich ») est réalisé sur des extraits obtenus par broyage des tissus infectés, dans une solution de citrate trisodique 0,5 M, clarifiés à volume égal par le chloroforme et stabilisés avec 0,4 p. 100 de formaldéhyde. Evidence for multiple intraspecific recombinants in natural populations of Watermelon mosaic virus (WMV, Potyvirus ) Inra

The full-length sequences of 13 isolates of Watermelon mosaic virus (WMV, genus Potyvirus) belonging to the three main molecular groups defined at the capsid level were obtained and compared to 4 sequences available in databases. Many isolates presented evidence for intraspecific recombination, particularly between molecular groups 1 and 2. Most isolates had different recombination breakpoints suggestive of multiple independent recombination events. The breakpoints were for the most part located between the C-terminal half of the HC-Pro coding region and the N-terminal part of the CI coding region. There was no evidence for positive selection in the WMV genome

Identification et caractérisation d’un potyvirus provoquant la maladie du rabougrissement jaune du melon Inra

Le virus du rabougrissement jaune du melon (muskmelon yellow stunt virus - MYSV) a été isolé dans le Sud de la France de melons (Cucumis melo L.) présentant de graves symptômes sur le feuillage et sur les fruits. Ce virus, transmissible mécaniquement, infecte de nombreuses espèces de Cucurbitacées (dont le concombre, la courgette et la pastèque) chez lesquelles il provoque des symptômes graves ; par contre, peu d’espèces sont sensibles à l’extérieur de cette famille : 3 espèces sur 36 éprouvées ont développé des infections généralisées. Sur Cucurbitacées (concombre, courgette, melon, pastèque), il existe 3 oïdiums : Sphaerotheca fuliginea, Erysiphe cichoracearum, Leveillula taurica. En fait, seuls les 2 premiers sont importants dans les conditions de la pratique agricole en France. Différents critères d’identification concernant la forme conidienne sont passés en revue : forme des spores, disposition du tube germinatif, présence de grains de fibrosine, aspect de l’appressorium, etc... Les conditions sèches sont plus favorables au développement et à la sporulation d’E. cichoracearum qu’à ceux de S. fuliginea qui semble davantage inféodé aux cultures sous abri.
