Somatic embryogenesis (SE) in conifers is at the base of nearly allbiotechnology products that potentially might be developedincluding transgenic trees. SE is a powerful tool for clonalpropagation that has opened avenues for deployment of superiorclonally replicated planting stock in forest plantations. Althoughthe SE process can be managed for high efficiency there are stillseveral problems that require better understanding of underlyingcauses. Pinus pinaster is the main industrial conifer in South-western Europe, especially France. Advanced forestry and conventional genetic breeding for 50 years resulted in a 3-fold increase in wood productivity. Up to 15% increase in growth and straightness were effective with first-generation varieties but genetic redundancy is expected to lower forthcoming genetic gain. Maximizing genetic gain for historical and new selection criteria remains challenging owing to long generation time of Maritime pine and low ability for conventional clonal propagation. Clonal plant production from self- and cross-pollinated seed families of Pinus sylvestris (L.) through somatic embryogenesis Inra

Several factors affecting somatic embryogenesis (SE) in Pinus sylvestris from self- and cross-pollinated seed families were studied with the aim of producing large quantities of clonal plants. Somatic embryogenesis initiation from zygotic embryos was improved on a medium with lower than standard concentrations of 2,4-dichlorophenoxyacetic acid (2.2 vs. 9.5 mu M) and 6-benzyladenine (2.2 vs. 4.5 mu M). On this medium, initiation rates of four controlled crosses, including one self-cross, varied from 3% to 25%. Among the maturation factors tested, the concentration of abscisic acid (ABA 80, 120 mu M) had no significant effect on the production of mature somatic embryos when the medium contained 0.1 M sucrose. Expression patterns of LmAP2L1 and LmAP2L2 encoding two-APETALA2 domain proteins during somatic embryogenesis and germination of hybrid larch (Larix x marschlinsii). Inra

Two APETALA2 domain transcription factors were firstly characterized in angiosperms and recently in several gymnoperms. These proteins are involved in several processes from flowering to embryogenesis in Arabidopsis thaliana and we extrapolated this result to hybrid larch (Larix x marschlinsii Coaz) resulting from a cross between European (Larix decidua) and Japanese (Larix kaempferi) larches. Somatic embryogenesis is a well described and controlled for this Pinaceae. We characterized two-AP2 domain genes named LmAP2L1 and LmAP2L2. Abstract Germins and germin-like proteins (GLPs) are members of a superfamily of proteins widely distributed in plants. Their localization within the extracellular matrixand in some cases their hydrogen peroxide-producing activity suggests that these proteins are involved in cell wall metabolism during stress responses and developmental processes. Somatic embryogenesis represents an important clonal propagation method for conifers (Lelu-Walter and Pâques 2009). Desiccation treatments applied to hybrid larch somatic embryos promote plantlet development (Lelu et al. 1995). Water availability could also be modulated via the gelling agents of the culture medium. We investigated the effect of high gellan gum concentration in the medium during somatic embryo maturation of hydrid larch. High gellan gum concentration of 8 gL-1 was compared to the normal concentration of 4 gL-1 at different stages of maturation. An integrated physiological and proteomic approach was used to investigate the effects of high gellan gum concentration in the medium during maturation of somatic embryos of hybrid larch, by comparing embryos incubated in media with a high gellan gum concentration (8 gL-1) and the conventional concentration (4 gL-1) after 1, 3, 6 and 8 weeks of maturation. Due to the reduced availability of water in the 8 gL-1 medium, the embryos cultivated on it had lower osmotic potential and water contents, but higher dry weights, at 8 weeks than embryos cultivated on the standard medium. Des cultures embryogènes de mélèze hybride, d'épicéa de sitka et de chêne ont été soumises à différentes conditions de culture et leur croissance étudiée en fonction de l'ajout de L-proline au milieu de culture (1 mM, 10 mM, et 100 mM). Si des températures basses (4 °C) inhibent totalement la croissance des cultures, celle-ci redevient partiellement normale en présence de proline. Nos résultats montrent que les cultures non seulement survivent à de basses températures mais sont aussi capables de croître activement au cours de la durée d'application du froid. L’embryogenèse somatique des conifères : une filière pour produire du matériel forestier de reboisement. Inra

Parmi les techniques de culture in vitro, l’embryogenèse somatique est potentiellement la méthode de régénération la plus performante pour la propagation clonale d’arbres forestiers du fait de son taux de multiplication élevé. Chez les conifères, l’intérêt de cette technologie pour d’amélioration génétique est indéniable. La cryoconservation des cultures embryogènes (dans l’azote liquide à –196°C) laisse entrevoir la possibilité de disposer à volonté de matériel juvénile et la conservation des ressources génétiques. L’embryogenèse somatique représente aussi un outil de développement en foresterie. De fait, cette technologie est déjà utilisée à l’échelle industrielle pour les épicéas et différents pins au Canada, aux USA et en Nouvelle Zélande.

Overproduction of proline in transgenic hybrid larch (Larix x leptoeuropaea (Dengler) cultures renders them tolerant to cold, salt and frost Inra

Cold, salt and frost are important environmental stresses in forest trees and may significantly reduce productivity. Elevated levels of proline are associated with these stresses and may help alleviate their effects. Transgenic hybrid larch Larix leptoeuropaea has been produced expressing a Vigna aconitifolia gene for pyrroline 5-carboxylate synthase, the rate-limiting step in proline synthesis. Embryogenic masses of hybrid larch were co-cultivated with Agrobacterium tumefaciens harbouring a binary vector expressing the gene. The integration of the gene into the plant genome was confirmed by Southern blot and by proline content analysis. Simplified and improved somatic embryogenesis for clonal propagation of Pinus pinaster (Ait.) Inra

In this study, several improvements and simplifications of SE protocols in Pinus pinaster (Ait.), a species of economic importance in the regions of Western Europe,are described. These improvements pertained to all stages of SE including high initiation frequencies in eight control pollinated seed families, relatively high somatic embryomaturation yield when cells were coated with particles of activated charcoal and a rapid production of plants directly in a shade house.
