Accelerated ageing test and behaviour investigation of isoenzymes in sesame seeds Agronomy

The seeds of a sesame crop contain approximately 50% oil content, one of the main reasons for their cultivation. However, information regarding methodologies for the assessment of quality seeds of this culture are scarce. Therefore, the objective of this study was to adapt the methods of accelerated ageing tests for sesame seeds and investigate changes in the behaviour of isoenzymes. The physiological characterization of the seeds and electrophoretic analysis of the isoenzymes superoxide dismutase, esterase, catalase, alcohol dehydrogenase, malate dehydrogenase and isocitrate lyase were also performed. Image analysis of coffee seeds submitted to the LERCAFE test Agronomy

The aim of this experiment was to quantify the stained areas of coffee seeds submitted to the LERCAFE test using image analysis. The seeds used were of the Catuaí Vermelho IAC 99 and Paradise cultivars. The physiological quality of the lots was assessed using germination tests, moisture content and a germination speed index. The LERCAFE test was conducted using seeds without parchment immersed in a solution of 3% sodium hypochlorite for 3 hours. Color photographs (RGB) with a resolution of 5 MPx were taken of each seed. The seeds were visually evaluated, and the functions generated from the analyses of the stained and non-stained regions were quantified by the Matlab R2009b program. In vitro germination and seed cryopreservation of paineira-rosa Sci. Agrar.Paran. / SAP

The paineira-rosa (Chorisia speciosa St. Hil) is an arboreal species of great importance in restoring degraded ecosystems and riparian forests, however, the availability of seeds of this species is low. The objective was to establish in vitro germination protocols and cryopreservation of paineira-rosa seeds. For the in vitro germination, MS culture medium and WPM was tested three concentrations of sucrose (0, 15, 30 g L-1) and three pH levels (4.8, 5.8 and 6.8). For cryopreservation, the seeds were dried on silica gel or laminar flow for different times (0, 1, 2, 3 and 4 h) and then were stored in liquid nitrogen (-196 °C) for 24 h, defrosted in a water bath (38 °C) for four minutes and they were inserted into test tubes containing WPM medium cultivation....
