Analysis of Clinical Ostreid Herpesvirus 1 (Malacoherpesviridae) Specimens by Sequencing Amplified Fragments from Three Virus Genome Areas ArchiMer
Renault, Tristan; Moreau, Pierrick; Faury, Nicole; Pepin, Jean-francois; Segarra, Amelie; Webb, Stephen.
Although there are a number of ostreid herpesvirus 1 (OsHV-1) variants, it is expected that the true diversity of this virus will be known only after the analysis of significantly more data. To this end, we analyzed 72 OsHV-1 "specimens" collected mainly in France over an 18-year period, from 1993 to 2010. Additional samples were also collected in Ireland, the United States, China, Japan, and New Zealand. Three virus genome regions (open reading frame 4 [ORF4], ORF35, -36, -37, and -38, and ORF42 and -43) were selected for PCR analysis and sequencing. Although ORF4 appeared to be the most polymorphic genome area, distinguishing several genogroups, ORF35, -36, -37, and -38 and ORF42 and -43 also showed variations useful in grouping subpopulations of this...

Antiviral immunity in the Pacific oyster, Crassostrea gigas: last development and perspective ArchiMer
Renault, Tristan; Faury, Nicole; Barbosa, Valerie; Moreau, Kevin; Haffner, Philippe; Pepin, Jean-francois.
Herpes-like viruses have been reported last decades in various marine mollusc species in association with mortality outbreaks throughout the world (Renault & Novoa 2004). One of these viruses isolated during French outbreaks has been characterised as an unassigned member of the Herpesviridae family and named ostreid herpes virus 1 (OsHV-1) or Pacific oyster herpes virus (Davison et al. 2005). Studying anti-viral non-specific defence mechanisms (innate immunity) is of great interest, because they constitute the only one in molluscs. Therefore, innate immunity has be investigated in the Pacific cupped oyster, Crassostrea gigas. The main aim of the work was focused on the identification and the characterisation of genes induced by OsHV-1 in the Pacific...

Detection and description of a particular Ostreid herpesvirus 1 genotype associated with massive mortality outbreaks of Pacific oysters, Crassostrea gigas, in France in 2008 ArchiMer
Segarra, Amelie; Pepin, Jean-francois; Arzul, Isabelle; Morga, Benjamin; Faury, Nicole; Renault, Tristan.
Ostreid herpesvirus 1 (OsHV-1) infections have been reported around the world and are associated with high mortalities of the Pacific oyster (Crassostrea gigas). In the summer 2008, abnormal mortality rates ranging from 80% to 100% were reported in France and affected only Pacific oysters. Analyses of oyster samples collected during mortality outbreaks demonstrated a significant detection of OsHV-1 (75% of analysed batches), which appeared stronger than previous years. DNA sequencing based on C and IA regions was carried out on 28 batches of OsHV-1 infected Pacific oysters collected in 2008. Polymorphisms were described in both the C and IA regions and characterized a genotype of OsHV-1 not already reported and termed OsHV-1 μVar. A microsatellite zone...

Detection of ostreid herpesvirus 1 DNA by PCR in bivalve molluscs: A critical review ArchiMer
Batista, Frederico; Arzul, Isabelle; Pepin, Jean-francois; Ruano, Francisco; Friedman, Carolyn S.; Boudry, Pierre; Renault, Tristan.
Herpes-like viral infections have been reported in different bivalve mollusc species throughout the world. High mortalities among hatchery-reared larvae and juveniles of different bivalve species have been associated often with such infections. The diagnosis of herpes-like viruses in bivalve molluscs has been performed traditionally by light and transmission electron microscopy. The genome sequencing of one of these viruses, oyster herpesvirus 1 (OsHV-1), allowed the development of DNA-based diagnostic techniques. The polymerase chain reaction (PCR) has been used for the detection of OsHV-1 DNA in bivalve molluscs at different development stages. In addition, the PCR used for detection of OsHV-1 has also allowed the amplification of DNA from an OsHV-1...

Development of a new diagnostic tool (Mini-Array) for ostreid Herpesvrus 1 (OsHV-1) detection in the Pacific oyster Crassostre gigas ArchiMer
Varsamos, S.; Pepin, Jean-francois; Sauvage, Christopher; Renault, Tristan.
OsHV-1 was the first herpesvirus to be identified in an invertebrate host and is associated to mortalities in the Pacific oyster (Crassostrea gigas) and other economically important bivalve species. Infections due to OsHV-1 are frequently reported and cause high mortality rates in larvae and spat of C. gigas. Detection of this pathogen is usually performed by PCR and/or in situ hybridization. This presentation will focus on the development of a new molecular diagnostic tool for OsHV-1 (and OsHV-1 variant 1) detection: the mini-array. This detection system, which allows multiple pathogens detection, combines viral DNA amplification by PCR and a specific hybridisation on a nylon membrane, revealed by a colorimetric reaction (DNA chip). Multiplex PCR involves...

Disinfectants and antiseptics used in mollusc hatcheries and nurseries ArchiMer
Francois, Cyrille; Pepin, Jean-francois.
Increasing development of aquaculture business has thrown up new requirements in terms of prevention and control of mollusc diseases. In closed structures, one way to reduce infectious risk is to make use of anti-infectives . Specific drugs like antibiotics should be used only for therapeutic purpose against identified agents. Less specific anti-infectives like disinfectants could be employed for preventive and also for curative goals against causal pathogens and opportunist agents. A critical analysis synthesizes the bibliography available on disinfection in mollusc hatcheries and nurseries. Physical and chemical tools are reviewed then selected depending on four criteria: (1) efficiency against most important pathogens of molluscs, (2) safety for...

Background: Massive mortality outbreaks affecting Pacific oyster (Crassostrea gigas) spat in various countries have been associated with the detection of a herpesvirus called ostreid herpesvirus type 1 (OsHV-1). However, few studies have been performed to understand and follow viral gene expression, as it has been done in vertebrate herpesviruses. In this work, experimental infection trials of C. gigas spat with OsHV-1 were conducted in order to test the susceptibility of several bi-parental oyster families to this virus and to analyze host-pathogen interactions using in vivo transcriptomic approaches. Results: The divergent response of these oyster families in terms of mortality confirmed that susceptibility to OsHV-1 infection has a significant...

In France, today, more than 80% of farmed blue mussel seed collection is based on larvae fixation on collector ropes. Mussels are traditionally grown on bouchot, which are rows of wooden poles placed perpendicularly to the shore, stuck in the ground within the gently sloping intertidal strip. Mussels are transferred either as seed attached on collecting ropes or as juveniles placed in net sockings and then wound up around the poles or suspended to longlines, where they will fatten and grow (Garen et al., 2004). In order to optimize seed collection and to limit unwanted biofouling on ropes, mussel growers local organizations identify best timing of seed collector ropes deployment by monitoring the number and size of planktonic mussel larvae from water...

Experimental infection of Pacific oyster Crassostrea gigas spat by ostreid herpesvirus 1: demonstration of oyster spat susceptibility ArchiMer
Schikorski, David; Renault, Tristan; Saulnier, Denis; Faury, Nicole; Moreau, Pierrick; Pepin, Jean-francois.
In 2008 and 2009, acute mortalities occurred in France among Pacific cupped oyster, Crassostrea gigas, spat. Different hypothesis including the implication of environmental factors, toxic algae and/or pathogens have been explored. Diagnostic tests indicated that OsHV-1 including a particular genotype, termed OsHV-1 mu Var, was detected in most of samples and especially in moribund oysters with the highlighting of virus particles looking like herpes viruses by TEM examination. In this study, an experimental protocol to reproduce OsHV-1 infection in laboratory conditions was developed. This protocol was based on the intramuscular injection of filtered (0.22 mu m) tissue homogenates prepared from naturally OsHV-1 infected spat collected on French coasts...

Experimental ostreid herpesvirus 1 infection of the Pacific oyster Crassostrea gigas: Kinetics of virus DNA detection by q-PCR in seawater and in oyster samples ArchiMer
Schikorski, David; Faury, Nicole; Pepin, Jean-francois; Saulnier, Denis; Tourbiez, Delphine; Renault, Tristan.
Herpes- and herpes-like viruses are known to infect a wide range of bivalve mollusc species throughout the world. Abnormal summer mortalities associated to the detection of ostreid herpesvirus 1 (OsHV-1) have been currently reported in France among larvae and spat of the Pacific cupped oyster Crassostrea gigas. In the present work, we have developed an experimental protocol of horizontal transmission based on the cohabitation between healthy and experimentally infected oysters. Through a cohabitation trial, the kinetics of OsHV-1 detection in different oyster organs and seawater samples were investigated and characterized for the first time using real time quantitative PCR.
