Caractérisation partielle d'un facteur hypothalamique de libération des hormones gonadotropes chez la carpe (Cyprinus carpio L.). Etude in vitro Animal Physiology

L'influence de différents facteurs sur la secrétion in vitro d'hormone gonadotrope par des hypophyses de Carpe a été étudiée grâce au choix d'un milieu d'incubation favorable. Après filtration sur Séphadex G 25 d'extraits acides réalisés, soit à partir d'hypothalamus, soit à partir de neurohypophyses, une activité de type "releasing factor" a été mise en évidence dans des fractions correspondant à un poids moléculaire inférieur à 5,000. Cette activité se distingue d'une part de celle de l'ocytocine et de la vasopressine et d'autre part de celle de neuroamines: adrénaline, noradrénaline, sérotonine et dopamine, qui ne montrent pas d'activité stimulante propre sur la secrétion gonadotrope in vitro. Effects of changes of photoperiod on gametogenesis in the rainbow trout (Salmo gairdneri) Animal Physiology

After their first reproductive period, adult male and female rainbow trout were put under three different photoperiodic regimes between January and June : (1) short daylength (6 h of light/day), (2) decreasing daylength (16L : 8D -> 10L : 14D), (3) skeleton photoperiod consisting of short day-length (6L : 18D) with a one-hour flash at various times during the dark cycle in a decreasing daylength fashion (between 16L and 10L), i.e. at the end of the daylight period of the preceding group. The rearing temperature was 15°C. After the fish were killed in June, it was found that decreasing daylength (group 2) had stimulated vitellogenesis. Effects of photoperiod and temperature on plasma gonadotropin and spermatogenesis in the rainbow trout Salmo Gairdnerii Richardson Animal Physiology

Two-year old male rainbow trout, Salmo gairdnerii, were exposed to the following photoperiods at 8° or 16°C from February 24 to June 19 : constant short photoperiod (8 L + 16 D), constant long photoperiod (16 L + 8 D), and gradually decreasing photoperiod (from 16 L to 8 L). Control fish were exposed to natural photoperiod and temperature. Plasma and pituitary gonadotropin levels were determined by radioimmunoassay and correlated with spermatogenic response in testes. Control fish did not show advancement of spermatogenesis, whereas those exposed to decreasing photoperiod at 8° or 16°C showed complete induction of spermatogenesis. La réponse hypophysaire à l'injection d'un extrait hypothalamique homologue et de LH-RH a été étudiée au cours du cycle reproducteur chez la Carpe Cyprinus carpio L. Elle est maximale pendant la mois d'été au moment de la période de reproduction. Elle est nulle ou faible en hiver pendant les mois de repos sexuel et apparait en avril-mai au moment du démarrage de la gamétogenèse et de l'augmentation de la température de l'eau.

At implantation the endometrium undergoes modifications necessary for its physical interactions with the trophoblast as well as the development of the conceptus. We aim to identify endometrial factors and pathways essential for a successful implantation in the caruncular (C) and the intercaruncular (IC) areas in cattle. Using a 13,257-element bovine oligonucleotide array, we established expression profiles at day 20 of the estrous cycle or pregnancy (implantation), revealing 446 and 1,295 differentially expressed genes (DEG) in C and IC areas, respectively (false discovery rate = 0.08). Induction de décharges gonadotropes hypophysaires chez la carpe (Cyprinus carpio L.) à l'aide du citrate de cisclomiphene Animal Physiology

L'injection de citrate de clomiphène (doses de 0,1, 1, et 10 mg/kg en une seule fois) à des Carpes mâles ou femelles, provoque une importante décharge d'hormone gonadotrope plasmatique. La latence et l'amplitude de la réponse croissent avec la dose utilisée. Les résultats les plus constants sont obtenus à la dose de 1 mg/kg, dose à laquelle tous les animaux, quels que soient leur état de maturité et leur sexe, présentent un pic de secrétion gonadotrope supérieur à 50 ng/ml et d'une durée de 12 hr environ, 56 à 60 hr après l'injection du produit. Cette décharge est comparable à la décharge gonadotrope ovulante observée chez le Carassin et parait donc susceptible d'entraîner l'ovulation ou la spermiation chez des poissons choisis au stade de maturité...

L'expérimentation a consisté : 1) à diluer avant insémination le sperme ou les ovules dans des dilueurs (dNa-K) constitués de quantités variables de KCl et NaCl (Tableau 1) ou 2) à pratiquer I'insémination directement dans ces mêmes dilueurs. Après insémination et avant transfert en eau douce, les gamètes peuvent être laissés simplement dans le dNa-K (technique de simple dilution). On peut aussi ajouter le dilueur d'insémination standard (dNa : dilueur no 1, tableau 1) (technique de double dilution). Pituitary response to LH-RH has been studied in the male and female rainbow trout (Salmo gairdneri) at different stages of gametogenesis. The male was hardly sensitive at the onset of spermatogenesis, but a response occured at the spermatid stage and continued during spermatogenesis and spermiation. We particularly studied the end of the female cycle. Response was low in immature fishes and in the early stages of oocyte maturation, but became stronger at vitelline maturation. The relationships of these responses to plasma and pituitary t-GTH levels are discussed. Plasma estradiol-17beta and gonadotropin during ovulation in rainbow trout (Salmo gairdneri R.) Animal Physiology

A radioimmunoassay (RIA) for estradiol-17beta (E2-17beta) with a chromatographic step has been tested on rainbow trout plasma. Overloading and dilution tests gave satisfactory results. Intracardiac injection of E2-17beta (10 microg/kg) in July gave a peak in the first 30 min after injection ; return to the basic level occurred within the next 24 hrs. Using RIA, trout-gonadotropin (t-GTH) and E2-17beta were followed in trout plasma during the very last stages of the reproductive cycle. Oocyte maturity was determined simultaneously. t-GTH increase during maturation and ovulation was confirmed while E2-17beta dropped before oocyte maturation to reach a low level when t-GTH was high. Precocious induction of oocyte maturation and ovulation in rainbow trout (Salmo gairdneri) : problems when using 17 alpha-hydroxy-20 beta-dihydroprogesterone Animal Physiology

The efficiency of 17 alpha-hydroxy-20 beta-dihydroprogesterone (17 alpha-20 beta P) administered alone or after pituitary priming, was investigated in vivo before peripheral migration of the germinal vesicle 4 to 8 weeks prior to natural ovulation. Treatment with 17 alpha-20 beta P alone (2 injections of 3 mg/kg at a 2-day interval) induced oocyte maturation in 94 p. 100 of the fish, but only 25 p. 100 ovulated. Treatment with 17 alpha-20 beta P (3 mg/kg once) 2 days after pituitary priming (1 ml/kg of trout pituitary extract, TPE, containing 3.25 x 10exp-3 mg/ml of trout gonadotropin, t-GTH) induced maturation in all fish, 59 p. 100 of which ovulated. Purification of gonadotropin from rainbow trout (Salmo gairdnerii Richardson) pituitary glands Animal Physiology

A method of purifying trout gonadotropic hormone is discussed. The technique consists of the following stages : alcoholic extraction, two successive gel filtrations on Sephadex G-50 and G-100, chromatography on DEAE-cellulose and ultrogel ACA54. The biological activity of fractions obtained is measured using as assay the maturation in vitro of trout oocytes. The preparation obtained has a molecular weight of about 27500, determined by behavior on Sephadex-G 100 and analytical ultracentrifugation for a KS20w of 2.45 Polyacrylamide gel electrophoresis shows two strips of electrophoretic mobility, 0.50 and 0.58. Sexual differences in salmon gonadotropin Animal Physiology

Salmon (Oncorhynchus tschawytscha) GTH's were purified from a mixture of pituitaries of both sexes or from separate male and female glands. Some of their biochemical, immunological and biological properties have been compared. These hormones differed essentially in amino acid composition, especially lysine, histidine and half cystine residues, and by the presence of alpha-amino (or alpha-aminoiso) butyric acid in male and GTH II preparations. The same differences were found between GTH I-II and GTH male-female. A testicular feed back on the hypothalamo-pituitary system was studied in male rainbow trout (Salmo gairdneri) after bilateral castration and steroid or testicular supplementation. Experiments were carried out at various periods of the spermatogenetic cycle ; in March-April (resting stage), June (initiation of spermatogenesis), October (end of spermatogenesis and before spermiation), and December (running stage). In March and April, plasma t-GTH is low in intact males (< 2 ng/ml) and castration induced a threefold rise of GTH. Plasma immunoreactive t-GTH and androgens, sperm production and biochemical changes in seminal fluid were studied during the spermiation period in rainbow trout. The volume of sperm collected by hand-stripping was very low (< 0.1 ml) at the onset of spermiation, increased slowly during the next 4 weeks and sharply thereafter. Plasma t-GTH was high (6 ng/ml) at the onset and thin decreased ; the sharp rise in sperm production started when circulating androgens had reached maximum values.
