Combined effects of dietary HUFA level and temperature on sea bass (Dicentrarchus labrax) larvae development ArchiMer
Vagner, Marie; Robin, Jean; Zambonino, Jose-luis; Person, Jeannine.
The purpose of this study was to investigate the combined effect of the incorporation of vegetable products in diet and temperature on enzymatic pathways for high unsaturated fatty acids (HUFA) desaturation in sea bass larvae. Four replicated groups were fed a low (LH; 0.8% EPA+DHA) or a high (HH; 2.2% EPA+DHA) n-3 HUFA microparticulated diet from mouth opening, six days post-hatching and were reared at 16 or 22 degrees C. The four experimental conditions (LH16, HH16, LH22 and HH22) were tested for 45 days. At the end of the experiment, body weight, total length and biomass were affected by temperature (P<0.001), while biomass as well as fresh body weight was also influenced by diet (P<0.05 and P<0.001 respectively). This always lead to the same...

Depletion of Essential Fatty Acids in the Food Source Affects Aerobic Capacities of the Golden Grey Mullet Liza aurata in a Warming Seawater Context ArchiMer
Vagner, Marie; Lacoue-labarthe, Thomas; Infante, Jose-luis Zambonino; Mazurais, David; Dubillot, Emmanuel; Le Delliou, Herve; Quazuguel, Patrick; Lefrancois, Christel.
The objective of this study was to evaluate the combined effects of thermal acclimation and n-3 highly unsaturated fatty acids (n-3 HUFA) content of the food source on the aerobic capacities of fish in a thermal changing environment. The model used was the golden grey mullet Liza aurata, a species of high ecological importance in temperate coastal areas. For four months, fish were exposed to two food sources with contrasting n-3 HUFA contents (4.8% ecosapentaenoic acid EPA + docosahexaenoic acid DHA on the dry matter DM basis vs. 0.2% EPA+DHA on DM) combined with two acclimation temperatures (12 degrees C vs. 20 degrees C). The four experimental conditions were LH12, LH20, HH12 and HH20. Each group was then submitted to a thermal challenge consisting of...

Is it possible to influence European sea bass (Dicentrarchus labrax) juvenile metabolism by a nutritional conditioning during larval stage? ArchiMer
Vagner, Marie; Zambonino, Jose-luis; Robin, Jean; Person, Jeannine.
The purpose of this study was to check if it is possible to influence sea bass juvenile metabolism by a conditioning of larvae from day 6 post hatching to day 45 to a low or a high HUFA compound diet (LH, 0.8% EPA + DHA and HH, 2.2% EPA + DHA) when reared at 16 or 22 °C. Following a 3-month intermediate period (at 19 °C using a commercial diet), the adaptability of the 4 initial larval groups to a HUFA experimental deprived diet (0.5% EPA + DHA) were tested at 19 °C in a 60 day-experiment (d-151–211). The four experimental duplicated conditions were ex-LH16 and ex-HH16 for the 2 groups previously reared at 16 °C (initial weight, 7.3 ± 0.5 g) and ex-LH22 and ex-HH22 for the 2 groups previously reared at 22 °C (initial weight, 11.1 ± 0.5 g). Survival was...

Ocean warming combined with lower omega-3 nutritional availability impairs the cardio-respiratory function of a marine fish ArchiMer
Vagner, Marie; Pante, Eric; Viricel, Amelia; Lacoue-labarthe, Thomas; Zambonino-infante, Jose-luis; Quazuguel, Patrick; Dubillot, Emmanuel; Huet, Valerie; Le Delliou, Herve; Lefrançois, Christel; Imbert-auvray, Nathalie.
Highly unsaturated fatty acids of the omega-3 series (HUFA) are major constituents of cell membranes, yet poorly synthesised de novo by consumers. Their production, mainly supported by aquatic microalgae, has been decreasing with global change. Understanding the consequences of such reductions is essential for ectotherm consumers, since temperature tightly regulates the HUFA content in cell membranes, maintaining their functionality. Integrating individual, tissue and molecular approaches, we examined the consequences of the combined effects of temperature and HUFA depletion on the key cardio-respiratory functions of the golden grey mullet, an ectotherm grazer of high ecological importance. For four months, fish were exposed to two contrasting HUFA diets...

Ontogenic effects of early feeding of sea bass (Dicentrarchus labrax) larvae with a range of dietary n-3 highly unsaturated fatty acid levels on the functioning of polyunsaturated fatty acid desaturation pathways ArchiMer
Vagner, Marie; Robin, Jean H.; Zambonino, Jose-luis; Tocher, Douglas R.; Person, Jeannine.
Four replicated groups of sea bass (Dicentrarchus labrax) larvae were fed diets containing an extra-high level of highly unsaturated fatty acids (HUFA) (XH; 3.7% EPA + DHA), a high level of HUFA (HH; 1.7%), a low level of HUFA (LH: 0.7%) or an extra-low level of HUFA (XLH; 0.5%) from day 6 to day 45 (experiment 1; XH1, HH1, LH1, XLH1). After a subsequent I-month period feeding a commercial diet (2.7% EPA + DRA), the capacity of the four initial groups to adapt to an n-3 HUFA-restricted diet (0-3% EPA + DHA; R-groups: XH2(R), HH2(R), LH2R, XLH2(R)) was tested for 35 d. Larval dietary treatments had no effect on larval and juvenile survival rates. The wet weight of day 45 larvae was higher in XH1 and HH1 (P<0.001), but the R-juvenile mass gains were...

Orientation des processus métaboliques chez les juvéniles de bar européen (Dicentrarchus labrax) par un conditionnement au stade larvaire ArchiMer
Vagner, Marie.
Worldwide supplies of fish oils and meals have reached their sustainable limits, forcing industries to look for alternative lipid sources for use in marine fish diets. As terrestrial animal products are prohibited, there is great interest in aquaculture to produce fish better able to utilise vegetable feedstuffs. Vegetable products are rich in 18 carbon fatty acids (C18 FA) but do not contain C20-22 n-3 highly unsaturated fatty acids (n-3 HUFA), such as ecosapentaenoic (EPA; 20:5n-3) and docosahexaenoic acids (DHA; 22:6n-3). The aim of this study was to know if it would be possible to apply a metabolic programming using nutritional conditioning during early larval stages, as already described in mammals, in order to stimulate the FA desaturation pathways...

Reduced n-3 highly unsaturated fatty acids dietary content expected with global change reduces the metabolic capacity of the golden grey mullet ArchiMer
Vagner, Marie; Zambonino-infante, Jose-luis; Mazurais, David; Imbert Auvray, Nathalie; Ouillon, Natascha; Dubillot, Emmanuel; Le Delliou, Herve; Akbar, David; Lefrancois, Christel.
In this study, we hypothesised that a reduction in n-3 HUFA availability for higher consumers, as expected with global change, would negatively impact the physiological performances of fish. The aim was to experimentally evaluate the effect of n-3 HUFA dietary content on cardio-respiratory performances of the golden grey mullet (Liza aurata), a microalgae grazer of high ecological importance in European coastal areas. These performances were evaluated in terms of critical swimming speed U (crit), associated oxygen consumption MO2, post-exercise oxygen consumption and calcium fluxes in cardiomyocytes. Two replicated groups of fish were fed on a rich (standard diet, SD diet: 1.2 % n-3 HUFA on dry matter basis, DMB) or a poor n-3 HUFA (low n-3 HUFA diet, LD...
