SSR molecular markers SSR are tandem arranged repeats of short DNA motifs (1-6 bp in length) distributed throughout the genome that frequently exhibit variation in the number of repeats at the locus of each rice variety. The SSR markers, therefore, are suitable for detecting polymorphisms between closely related genotypes of Thai local rice varieties from the National Rice Germplasm Bank. In this research, DNA fingerprinting was conducted using 8 Thai local rice varieties. i.e., Niaw Sanpahtawng (NSPT_GB), Khao Gaw Diaw (KGD_GB), Niaw Dam (ND_GB), Khao tah Haeng 17 (KTH17_GB), Jao’ Haw (JH_GB), Pin Gaew 56 (PG56_GB), Jek Chuey Gahb Khiaw (JCGK_GB), Sang Yod Pattalung (SYPL_GB), and 5 standard varieties, i.e., Khao Dawk Mali 105 (KDML105_GB, KDML105_C),...

การตรวจวิเคราะห์ข้าวและสินค้าข้าวดัดแปรพันธุกรรมด้วยปฏิกิริยาลูกโซ่ ในสภาพเวลาจริง Thai Agricultural

Genetically modified (GM) insecticidal rice line, Bt63 (Shanyou63), has been developed in China and found unauthorized in processed rice products in many countries. This transgenic rice contains a copy of the synthetic DNA sequence with two genes: CryIA(b) and CryIA(c). These genes encode for the respective Bt toxins which is lethal to Lepidoptera. The aim of this study was to determine the contamination of Bt63 in non-labelled commercially available processed rice products from supermarkets. In this study, a real-time Polymerase Chain Reaction (PCR) was used to simultaneously determine the contamination of Bt63. The survey was conducted on a total of 21 and 42 non-labeled samples of processed rice products collected in the years 2011 and 2012,...

Currently, there are several reports on GM rice illegally available on market. To set up a defense measure in order to prevent GM contamination in Thai rice market, this activity was conducted by analyzing for CaMV 35S Promoter, NOS terminator and a transgene CryIA(b), which were carrying by the Bt63 rice. The Bt63 rice is an insect-resistant rice variety developed by the People Republic of China. The analyses were done by using a Real-Time PCR technique. DNA samples were extracted from rice products using a GeneScan extraction method and confirmed by a PCR using a primer pair KVM159/KVM160, which amplifies a rice species specific PLD gene encoding for an enzyme Phospholipase D. It was found that all samples could be amplified showing a PLD gene with a...

Development of rice varieties conferring broad-spectrum and durable resistance against brown planthopper (BPH) is one of the major challenges facing the plant breeders. Idea has been generated in this study for incorporating resistance genes of ovicidal response and feeding inhibition into rice breeding lines. Our initial experiments focused on discovering the ovicidal trait in indica rice cultivars. We found that some weedy rice lines had the ovicidal resistance, causing 60% to 100% egg mortality. The egg mortality rate depends on the rice developmental stages and is the highest at the maximum tillering stage. This is the first report of the ovicidal trait in indica rice lines. The BPH resistance in weedy rice lines is not only mediated by the ovicidal...

Bacterial blight (BB), caused by Xanthomonas oryzae pv. oryzae (Xoo), is one of the most serious diseases of rice in all major rice-growing areas of Thailand. The most effective approach to control BB is the use of resistant varieties which is economically feasible, an environmental-friendly control method and easy for farmers to handle. However, the selection pressure from the usage of resistant cultivars may result in mutation of the pathogen population to overcome the resistance genes. A feasible method to solve this problem is by pyramiding multiple resistance genes, which are board spectrum, into susceptible rice varieties. The aims of this research were to identify the effectiveness of board spectrum BB resistance genes and to better understand the...

Thai Agricultural

Most farmers in the Northeastern Thailand grow Khao Dawk Mali 105, a non-glutinous variety because of its premium cooking and eating quality and its aroma. However, KDML105 is susceptible to blast, brown planthopper and soil salinity. In order to maintain its yield stability, it is necessary to incorporate those above mentioned traits into KDML105 background. By using molecular markers to select for those plants carrying the target genes in each generation, it will help improving the selection efficiency in term of accuracy and time consumed. The experiment was conducted at Ubon Ratchathani Rice Research Center during 2007-2012 using the lines derived from the backcrossing program of KDML105; UBN03004 (IR77955-14-17*2/B.T) and UBN03007...

แผนที่กายภาพพันธุกรรมที่ควบคุมขนาดของรากและการค้นหายีนที่เกี่ยวข้องกับความสามารถชอนไชของรากข้าวบนโครโมโซมที่ 4 Thai Agricultural
Varapong Chamarerk.
Major quantitative trait loci (QTL) associated with basal root thickness (BRT) on chromosome 4 was used as the target for physical mapping. Two parental rice lines contrasting in their root characteristics, CT9993-5-10-1-M (CT) and IR62266-42-6-2 (IR), were used in this study. The objectives of this study were to fine map the major BRT QTL on chromosome 4 in rice and to isolate candidate genes related to root penetration index (RPI) trait which located in this region. The BAC library available for screening in this study was constructed from a rice cultivar Nipponbare (japonica) with average insert sizes of 130 to 150 kb and with a 10X genome equivalent. The RFLP markers located near the target BRT QTL on rice chromosome 4 were used for screening BAC...
