Approaches towards improvement of microbial strains used in solid state fermentation IRD

The low water activity, the complex nature of the support and/or substrate, the concentation of substrates used and products formed, and the heterogeneity within the system, influence the growth and physiology of the microbial strains used in solid state fermentation (SSF). As a result, the quality and quantity of the products formed from SSF processes significantly differ from those of submerged fermentation (SmF). Some strains that performed well in SmF processes do not perform efficiently in SSF process. Therefore, it is necessary to maximize the potential of the organism through genetic manipulation to suit the SSF process. Molecular techniques applied to fungal strain upgradation capability related to SSF cultures IRD

Mutant strains derived from #A. niger$ C28B25 and belonging to series AW99 [producing maximum pectinase level in submerged fermentation (SmF)] and series AW96 [producing maximum pectinase level in solid state fermentation (SSF)] were compared concerning pectinase properties and thermal stability. Pectinases produced by AW96 exhibited increased thermal stability compared to AW99. Results support the idea that not only the quantity but also the quality of the enzymes can be modified when SSF or SmF techniques are used. The use of differential display polymerase chain reaction may enable the isolation of solid state fermentation-specific genes. (Résumé d'auteur)

Mutation by UV light resulted in the isolation of #A. niger$ strains geared towards high pectinase production in either SSF or SFM but never in both. Water activity plays an essential role on the levels of activity of specific enzymes. Mutated strains showed growth and sporulation patterns distinguishable by morphometric techniques, when cultured on defined media. (Résumé d'auteur)

Pectinase-hyperproducing mutants of Aspergillus niger C28B25 for solid-state fermentation of coffee pulp IRD

The aim of this study was to improve mold strains for the production of pectinases by solid-state fermentation (SSF) of coffee pulp. A screening of 248 strains, isolated in Mexico's coffee-growing areas, permitted us to select a wild strain of #Aspergillus niger$ which in 72 h attains a peak production of 27.7 U ml-1(138 U g-1 dry pulp) of pectinase measured by viscosimetry. Though the use of a selective culture medium with low water activity (aw = 0.954) with 2-deoxy-glucose (2DG) it was possible to isolate pectinase-hyperproductive mutant strains for SSF(44.5 U ml-1, 228 U g-1 dry pulp). Derepressed mutant strains that hyperproduced pectinase by submerged fermentation (SmF) were also obtained using a classic selective medium with high water activity...

Solid substrate cultures of moulds behave in quite a different way to conventional submerged cultures. Catabolic repression often observed in submerged fermentation can easily be overcome by using solid state fermentation. #A. niger$ strains can be grown by SSF technique with the following advantages : enzyme titres and productivity are usually higher ; there is a tendency to early enzyme excretion as compared to SmF technique.
