Broad-range PCR-TTGE for the first-line detection of bacterial pathogen DNA in ticks Inra
Halos, L.; Mavris, M.; Vourc'h, G.; Maillard, R.; Barnouin, J.; Boulouis, H.J.; Vayssier-Taussat, M..
Ticks are known or suspected vectors for a wide range of bacterial pathogens. One of the first steps for tick-borne risk assessment is the detection of these pathogens in their vectors. In the present study, a broad-range PCR amplification of the eubacterial gene encoding the 16S rRNA gene combined with Temporal Temperature Gradient gel Electrophoresis (TTGE) was evaluated as a method allowing the one-step detection of bacterial pathogen DNA in ticks. Firstly, DNA extracts from bacteria known to be tick-borne pathogens, i.e., Borrelia burgdorferi lato sensu, Anaplasma phagocytophilum, Spotted Fever Group (SFG) Rickettsia spp., were used to establish a TTGE pathogen DNA reference marker. Secondly, we used broad-range PCR-TTGE to detect the presence of DNA...

Monoterpene effect on feeding choice by deer Inra
Vourc'h, G.; de Garine-Wichatitsky, M.; Labbé, A.; Rosoloswki, D.; Martin, J.L.; Fritz, H..
A previous study showed that Sitka black-tailed deer (Odocoileus hemionus sitkensis) consumption was negatively correlated with monoterpene content in western redcedar (Thuja plicata). To test whether these monoterpenes were deterrent to Sitka black-tailed deer, we performed feeding choice experiments with four hydrocarbon (sabinene, myrcene, α-pinene, and d + l-limonene) and one oxygenated (α,β-thujone) monoterpene solution at their highest natural concentration in western redcedar foliage. To test whether deer response was species specific, we ran similar experiments on European roe deer (Capreolus capreolus) and rusa deer (Cervus timorensis russa). In all experiments, monoterpenes were repellent. Solutions with α,β-thujone, the major monoterpene in...
