Characterization of resistance breaking Meloidogyne incognita - like populations using lectins, monoclonal antibodies and spores of Pasteuria penetrans IRD

Six clones de #Meloidogyne$, provenant de populations récoltées en Côte d'Ivoire et qui possèdent un phénotype estérasique particulier (codé pVI) ainsi que la faculté de briser la résistance conférée par le gène #Mi$, ont été comparés à #M. incognita$ à l'aide de différentes sondes. Le marquage à l'aide de lectines des exsudats amphidiaux de juvéniles de second stade ne permet pas de distinguer ces lignées de #M. incognita$. Des souches de #Pasteuria penetrans$ permettent de les distinguer et mettent aussi en évidence une certaine variabilité au sein de ce groupe. Deux anticorps monoclonaux, spécifiques de #M. incognita$ indiquent que les clones de phénotype pVI partagent certains caractères avec #M. incognita$ mais aussi qu'ils s'en distinguent. (Résumé...

Differentiation of peanut clump virus serotypes by monoclonal antibodies IRD

Des anticorps monoclonaux dirigés contre le virus du "clump" de l'arachide ont permis de caractériser 5 sérotypes du virus. La comparaison de 4 types de tests immunoenzymatiques ELISA a permis de sélectionner celui qui est le mieux adapté au diagnostic de routine et à la différenciation entre sérotypes du virus. La plupart des anticorps monoclonaux conservent leur activité après adsorption sur la phase solide en ELISA, et le même anticorps peut être utilisé comme capteur et comme anticorps biotinylé. Etant donné que les anticorps obtenus sont capables de reconnaître des changements mineurs de conformation de l'antigène viral, il est important de sélectionner avec soin le format du test ELISA utilisé dans les comparaisons entre différents isolats du virus....

Etude immunologique du virus de la mosaïque de l'igname IRD

L'utilisation d'anticorps monoclonaux permet de préciser la diversité biologique de divers isolats du virus de la Mosaïque de l'Igname et de les distinguer en fonction de leur origine. Ces anticorps monoclonaux ont par ailleurs permis de distinguer nettement le virus de la Mosaïque de l'Igname d'autres potyvirus africains. Cette étude permet d'envisager la mise au point d'un kit de diagnostic performant de ce virus. (Résumé d'auteur)

 The detection of Beet necrotic yellow vein virus (BNYVV) instored sugar beets by means of monoclonal antibodies or antibody singlechain fragments (scFv) often poses problems, because the immunodominantC-terminal epitope of the viral coat protein is readily lost due toproteolysis. Clones which produce scFv specific for protease-stableBNYVV epitopes were selected from two naive phage display libraries.Fusion proteins of the scFv with a human IgG kappa chain (expressed fromthe newly designed vector pCL) or with alkaline phosphatase,respectively, allow the ELISA detection of BNYVV even in stored sugarbeets with a sensitivity which was comparable or often higher than thatachieved with polyclonal antibodies

Six monoclonal antibodies specific for the major capsid protein of rotavirus, VP6, previously characterized, were tested in a biological assay for their capacity to block the transcriptase activity associated with the single-shelled particles. The results showed that two MAbs (RV-50 and RV-133), specific for distinct antigenic sites, were able to block the transcription when they were incubated with a purified baculovirus-expressed group A VP6, prior to the reconstitution of the single-shelled particles from the cores, suggesting that at least two domains are involved in active single-shelled particle reconstitution. La mosaïque de l'igname : aspects épidémiologiques au Burkina Faso et variabilité du virus IRD

L'igname, culture importante de régions tropicales est sensible au virus de la mosaïque de l'igname (yam mosaic virus, YMV, potyvirus), propagé par tubercules et transmis par pucerons. Au Burkina Faso, des isolats de potyvirus biologiquement semblables au virus de la mosaïque de l'igname ont été identifiés dans les plantes d'igname (#D. cayenensis-rotundata$ et #D. alata$) par la présence de symptômes de mosaïque et de surcoloration nervaire, et ont été détectés par des anticorps polyclonaux dirigés contre ce virus. The chemokine stromal cell-derived factor 1 (SDF-1) is the natural ligand for CXC chemokine receptor 4 (CXCR4). SDF-1 inhibits infection of CD4+ cells by X4 (CXCR4-dependent) human immunodeficiency virus (HIV) strains. We previously showed that SDF-1α interacts specifically with heparin or heparan sulfates (HSs). Herein, we delimited the boundaries of the HS-binding domain located in the first β-strand of SDF-1α as the critical residues. We also provide evidence that binding to cell surface heparan sulfate proteoglycans (HSPGs) determines the capacity of SDF-1α to prevent the fusogenic activity of HIV-1 X4 isolates in leukocytes. Reciprocal assistance for aphid transmission between non-transmissible strains of zucchini yellow mosaic potyvirus in mixed infections Inra

 Several aphid non-transmissible strains of zucchini yellow mosaic potyvirus (ZYMV) have been described, in relation with mutations in the two viral proteins required for transmissibility, coat protein and helper component. Assistance mechanisms were previously shown to allow transmission of such strains when functional proteins are provided in trans. In this paper, we used monoclonal antibodies to allow a specific detection of two aphid non-transmissible strains of ZYMV and we revealed that reciprocal assistance mechanisms can mediate simultaneous aphid transmission of these deficient strains. Potential epidemiological and evolutive consequences of such assistance mechanisms between variants in complex virus populations are discussed.

Use of monoclonal antibodies for the identification of different antigenic domains in apple chlorotic leaf spot virus Inra

Thirteen monoclonal antibodies (Mabs) specific for apple chlorotic leaf spot virus (ACLSV), produced by the somatic cell hybridization technique, were used to investigate the antigenic structure of the virus. Epitope specificity studies showed that these Mabs defined in ACLSV particles seven independent antigenic domains, representing at least eight distinct epitopes. One of them was present only in virions and not in dissociated subunits. It appeared that the interaction between a Mab and the virus could, in some cases, induce conformational changes in the viral particles which enhanced the binding of others.
