Aislamiento y caracterización de Streptococcus bovis y estudio inicial para el aislamiento de su bacteriófago Colegio de Postgraduados

El objetivo del presente estudio fue obtener un cultivo puro de la bacteria ruminal Streptococcus bovis a partir de líquido ruminal de ovinos alimentados con una dieta alta en sorgo (70 % en base seca). Además, se analizó su actividad fermentativa (producción de AGV y lactato) y el proceso de liofilizado como método de conservación, y se realizaron ensayos para expresar su bacteriófago. Para el aislamiento se usó un medio comercial para Streptococcus spp. (Merck) preparado bajo anaerobiosis. Para la identificación se usó el sistema API 20 Strep, reacción catalasa, morfología celular y tinción de Gram. Control of Listeria monocytogenes growth in soft cheeses by bacteriophage P100 BJM

The purpose of this study was to determine the effect of bacteriophage P100 on strains of Listeria monocytogenes in artificially inoculated soft cheeses. A mix of L. monocytogenes 1/2a and Scott A was inoculated in Minas Frescal and Coalho cheeses (approximately 10(5) cfu/g) with the bacteriophage added thereafter (8.3 x 10(7) PFU/g). Samples were analyzed immediately, and then stored at 10 ºC for seven days. At time zero, 30 min post-infection, the bacteriophage P100 reduced L. monocytogenes counts by 2.3 log units in Minas Frescal cheese and by 2.1 log units in Coalho cheese, compared to controls without bacteriophage. Between 1997 and 2007, 18% of the analyses carried out in dry weather recorded more than 230 Escherichia coli / 100 g of bivalves (Ifremer French microbial monitoring network), leading the administration to restrict the shellfish harvest area, including suspension of production. One of the most economically important bacterial pathogens of plants and plant products is Dickeya dadantii. This bacterium causes soft rot disease in tubers and other parts of the potato and other plants of the Solanaceae family. The application of restricted host range bacteriophages as biocontrol agents has recently gained widespread interest. This study purposed to isolate the infectious agent of the potato and evaluate its biocontrol by bacteriophages. Lactococcus bacteriophages isolated from whey and their effects on commercial lactic starters BABT

The incidence of phages of lactic acid bacteria in milk industry and their effects on acidification ability of commercial lactic acid starters were studied. Cheese whey samples (33 samples) were collected from 17 factories. A total of 16 bacteriophages were isolated (12 specific for Lactococcus lactis, 3 for L. diacetylactis and one capable of lysing both species). The results showed that 10% reduction in acidification tests was not good indication of phage in the sample. The majority of samples showed reduction higher than 10%, although only 65% were phage positive. An inter-laboratory study of the accuracy of microbial source tracking (MST) methods was conducted using challenge fecal and sewage samples that were spiked into artificial freshwater and provided as unknowns (blind test samples) to the laboratories. The results of the Source Identification Protocol Project (SIPP) are presented in a series of papers that cover 41 MST methods. This contribution details the results of the virus and bacteriophage methods targeting human fecal or sewage contamination. Human viruses used as source identifiers included adenoviruses (HAdV), enteroviruses (EV), norovirus Groups I and II (NoVI and NoVII), and polyomaviruses (HPyVs). Phage amplification assay as rapid method for Salmonella detection BJM

The application of rapid methods is crucial for the HACCP program implantation in food industry. In this context, Phage Amplification Assay is a good candidate because is based on the interactions of phage and their host bacteria. This method using phage P22 was applied with to detect Salmonella cells in chicken breast. Samples of 25 g of chicken breast were diluted and the appropriate dilutions were used in phage amplification assay for Salmonella detection. After 3-4 h of incubation, it was observed a phage titre of approximately 10(4) pfu mL-1, indicating that there were Salmonella cells which were naturally present in the meat. Background: Ideally, bacteriophages of pathogenic bacterial hosts should be polyvalent to be able to replicate in an alternative nonpathogenic bacterium. Thus, accidental infection by the original host can be avoided when bacteriophage lysates are used in biocontrol protocols. Results: From 15 wastewater samples, collected at different sites in the V Region in Chile, we selected three bacteriophages (FC, FP, and FQ) capable of productively infecting Salmonella enterica serovar Choleraesuis. Stability inside hen eggs of a Salmonella enterica serovar Enteritidis bacteriophage Electron. J. Biotechnol.

Background: Bacteriophages are viruses that infect bacteria and therefore are widespread in nature. Those that lyse the pathogen Salmonella enterica serovar Enteritidis (SE) should be expected in niches in which this bacterium thrives, among others the avian egg. Furthermore, bacteriophages could remain viable in this milieu. Results: Upon artificially infecting hen eggs with the SE bacteriophage f18 we found that the bacteriophage titer remains stable at least for up to 144 hrs post-infection , both in yolk and albumen at 25ºC. Conclusion: Bacteriophage f18 withstands the physico-chemical conditions of the egg inner milieu and could be considered for SE-controlling measures in the poultry industry.
