A Feedback Mechanism to Control Apoptosis Occurs in the Digestive Gland of the Oyster Crassostrea gigas Exposed to the Paralytic Shellfish Toxins Producer Alexandrium catenella ArchiMer

To better understand the effect of Paralytic Shellfish Toxins (PSTs) accumulation in the digestive gland of the Pacific oyster, Crassostrea gigas, we experimentally exposed individual oysters for 48 h to a PSTs producer, the dinoflagellate Alexandrium catenella. In comparison to the effect of the non-toxic Alexandrium tamarense, on the eight apoptotic related genes tested, Bax and BI.1 were significantly upregulated in oysters exposed 48 h to A. catenella. Effect of Dichlorvos on the acetylcholinesterase from tambaqui (Colossoma macropomum) Brain. Repositório Alice

Dichlorvos is an acutely toxic organophosphorous pesticide that is known as a classical acetylcholinesterase (AChE; EC 3.1.1.7) inhibitor. Here, the brain AChE from the important Amazonian fish tambaqui (Colossoma macropomum) was assayed in the presence of this insecticide and also of deltamethrin, a classical sodium and potassium channel inhibitor (negative control). Four tissue homogenates were analyzed in triplicate for AChE activity using acetylthiocholine as the substrate and 5,5'-dithiobis(2-nitrobenzoic) acid (DTNB) as the color-developing agent. Evaluation of biomarkers in oyster larvae in natural and polluted conditions ArchiMer

Crassostrea gigas D-shaped larvae were subjected to different conditions of temperature and salinity for 24 h and four biomarkers (acetylcholinesterase (AChE) activity, thiobarbituric acid reactive substances (TBARS) levels, glutathione S-transferase (GST) and catalase (CAT) activities) were measured. AChE activity decreased when salinity increased from 25 to 30 and 35 psu at 20 and 25 degreesC. Temperature did not seem to have an influence on AChE activity. TBARS levels increased as a function of salinity when the temperature was maintained at 20 degreesC, whereas at 25 degreesC no effect of salinity could be observed. Evaluation of the impacts of land-based contaminants on the benthic faunas of Jakarta Bay, Indonesia ArchiMer

To evaluate the impact of land-based contaminants on benthic faunas, as part of the Land-Ocean Contamination Study (LOCS), a study transect was established in Jakarta Bay, Indonesia during the 1996 southeasterly monsoon. The transect extended 72 km between the city's main port, Tanjung Priok, and the Pilau Seribu Island chain in the Java Sea. The dissolved concentrations of Pb, Cu, Zn, Cr and Ni in seawater, in suspended particulate matter (SPM) and surficial sea bed sediments were measured along the transect at nine sites. Genotoxicity and activation of cellular defenses in transplanted zebra mussels Dreissena polymorpha along the Seine river ArchiMer

The aim of the present study was to confirm the relevance of studying DNA adduct formation in a field study. In that context, freshwater mussels Dreissena polymorpha, collected in a reference station, were transplanted in different sites with a pollution gradient. After one and two months, mussels were collected and DNA adduct formation was analyzed using the 32P post labelling technique on both gills and digestive glands. In addition, the expression of genes involved in the detoxification system (catalase (CAT), superoxide dismutase (SOD), glutathione S-transferase (GST), HSP70, aryl hydrocarbon receptor (AHR), P glycoprotein (PgP), metallothionein (MT)) was assessed by RT-PCR. A 49-month-old Holstein cow with anorexia, tachypnea, enlarged peripheral lymph nodes, and difficulty standing up was suspected of bovine leukosis. Hematological examination revealed lymphocytosis with the presence of neoplastic cells. Increased total lactate dehydrogenase (LDH) activity, isozymes of LDH-2 and LDH-3 activities and thymidine kinase activity were observed. Cytological findings of fine needle aspiration of subiliac lymph nodes indicated lymphosarcoma. Histopathology and antibody analysis confirmed the diagnosis of enzootic bovine leukosis, a B-cell bovine lymphoma caused by bovine leukemia virus. Rapid metabolic adaptation in European sea bass (Dicentrarchus labrax) juveniles fed different carbohydrate sources after heat shock stress ArchiMer

A study was conducted to evaluate the effect of two dietary carbohydrate sources (waxy maize starch and glucose) on the metabolic adaptation of sea bass juveniles (initial weight: 24 g) to a heat shock treatment (temperature rise from 18 degrees C to 25 degrees C within 24 h). Two isonitrogenous and isolipidic diets were fon-nulated to contain 20% waxy maize starch (WS diet) or 20% glucose (GLU diet). Triplicate groups of fish were fed to near satiation for 4 weeks at both temperatures (18 degrees C and 25 degrees C). Responses of the European flounder (Platichthys flesus) to a mixture of PAHs and PCBs in experimental conditions ArchiMer

A multibiomarker approach was developed to evaluate the juvenile European flounder responses to a complex mixture of 9 polycyclic aromatic hydrocarbons (PAHs) and 12 polychlorinated biphenyls (PCBs). The variations of catalase activity, which is a biochemical biomarker of exposure to an oxidative stress, were followed month by month (from May 1998 to May 1999) in Mediterranean clams (Ruditapes decussatus) collected at three stations (J, A and F), and mussels (Mytilus galloprovincialis) collected at two stations (J and C) in Lake Bizerte (Tunisia). Several oceanographic parameters were measured together with catalase activity in the animals to evaluate the quality of waters in the lake. Compared to traditional systems, recirculating systems allow to reduce replacement water and waste volumes. Use of clearance rate in Choromytilus chorus (Bivalvia: Mytilidae) as a non-destructive biomarker of aquatic pollution RChHN

Reduction in clearance rate was observed in the mussel Choromytilus chorus in relation to the degree of environmental pollution. Three sampling sites included the polluted San Vicente Bay, middling polluted Corral Bay, and weakly polluted Yaldad Bay in southern Chile. Pollution levels were estimated by quantitative analysis of polynuclear aromatic hydrocarbons (PAHs) in mussel tissue from the three sites. Mussels from San Vicente showed lowest clearance rates and highest PAHs levels in both spring 1998 and summer 1999. Heavy metals (Cd, Cu, Hg and Zn) and metallothionein levels were determined in oysters Crassostrea gigas from two sites: the Gironde Estuary (contaminated site) and Bourgneuf Bay (uncontaminated site). Metallothionein levels led us to suspect the presence of cadmium in the environment when quantified in the gills. Soluble cadmium and metallothioneins were positively correlated in the digestive gland of oysters from Gironde (contaminated site), but metallothionein levels in this organ were not significantly different from levels observed in the digestive gland of oysters from Bourgneuf Bay (uncontaminated site).
