Brain glutamic acid decarboxylase and synthesis of amino-butyric acid in vertebrate and invertebrate species National Institute of Agronomic Research

An isotopic method is described for the simultaneous determination of the CO2 and GABA production from glutamic acid in nervous tissue preparations. Tissue from different species has been examined under a variety of conditions. Discrepancies between the production of GABA and of CO2 have been demonstrated in two instances-at the higher of two pH optima and in subcellular fractions. A quantitative analysis of the free amino acids of the honeybee brain is appended.

Cerebral glutamate metabolism In parkinson's disease: An in vivo dynamic C-13 nms study In the rat Inra

The aim of this work was to explore in vivo the metabolism of the basal ganglia in a rat model of Parkinson's disease. C-13 NMR spectroscopy was used to monitor the synthesis of glutamate/glutamine from [2-C-13] sodium acetate. C-13 label incorporation in glutamate at the carbon C4 was measured in the brain of rats in different physiopathological states and after antiparkinsonian treatment. Studies were performed in control rats (n = 6) and parkinsonian rats (n = 5) in a stable state and after acute levodopa administration (50 mg/kg iv). C-13 NMR spectra recorded using a H-1/C-13 surface probe were acquired in the injured cerebral hemisphere. In certain brain parts of queen bees, 2 and 3 years old, a defective tissue is found that, in bilateral structures also occurs symmetrically. The cavities can reach 0.0015 mm3 of volume, this size being found almost exclusively in the nuclear parts. Very seldom there are comparatively small lacunas in the fibrillar tissue. In these investigations the corpora pedunculata (mushroom bodies) - the place of associative functions - showed no signs of degeneration in the cup sphere. This divergent behaviour of the superior centres is in accordance with already know statements.

Electron microscope observations on the calyces of the insect brain National Institute of Agronomic Research

The pedunculate bodies consist of three fibrillar regions: calyces, stalk, and roots. A group of small association neurons (the globule cells) lies within and around the calyces. The synaptic region of the calyces shows, under the phase contrast microscope, small, dense circumscribed areas of neuropil corresponding to the so-called “glomeruli” of light microscopists. The electron microscope demonstrated that each glomerulus consists of a large central nerve ending and several thin peripheral fibers. The central ending is filled with small vesicles. The increase of plasma arginin-vasopressin (AVP) release, which translates hypothalamic AVP neuron activation in response to immune challenge, appears to occur independently of plasma osmolality or blood pressure changes. Histological evidence is presented for a caste difference in endocrine activity during the critical period of determination in the larval honeybee. Until the Last larval instar, neurosecretory cells in the brain of the honeybee larva remain undifferentiated. During this undifferentiated period there is a quantitative difference in apparent activity, as evidenced by the frequency of occurrence of cytoplasmic particles in certain large neuroblasts. Whereas the histological criteria for neurosecretory cells cannot be fulfilled, the brain is presumably exercising a regulatory role in growth and development. The present experiment was designed to study changes in behavior following immunolesioning of the basal forebrain cholinergic system. Rats were lesioned at 3 months of age by injection of the 192 IgG-saporin immunotoxin into the medial septum area and the nucleus basalis magnocellularis, and then tested at different times after surgery (from days 7-500) on a range of behavioral tests, administered in the following order: a nonmatching-to-position task in a T-maze, an object-recognition task, an object-location task, and an open-field activity test. The results revealed a two-way interaction between post-lesion behavioral testing time and memory demands.
