Comparative analysis of sulfated galactans from red algae by reductive hydrolysis and mild methanolysis coupled to two different HPLC techniques Inra

The sugar determination of the sulfated galactans, agars and carrageenans of various red algae was performed using two different techniques of depolymerisation with subsequent HPLC analysis: 1) reductive hydrolysis/ HPAEC-PAD; 2) mild methanolysis/ RPLC-DR. Both techniques were optimized to release quantitatively the composite sugars (galactose, 6-O-methyl-galactose, the labile 3,6-anhydrogalactose and 2-O-methyl-3,6-anhydrogalactose residues) and precise relative response factors of authentic 3,6-anhydrogalactose were determined. The methanolysate neutralisation step, performed subsequently to methanolysis depolymerisation, was demonstrated as a key step for the quantitative recovery of the anhydrogalactose residues. Effect of some plant flavonoids on in vitro behaviour of an arbuscular mycorrhizal fungus Inra

Deux isoflavonoïdes (la glycéolline I et le coumestrol) et un flavonoïde (la quercétine) ont été éprouvés vis-à-vis de la germination in vitro de Gigaspora margarita. Les expériences ont été faites en utilisant les concentrations de 0, 0,05, 0,5, 5 et 50 μmol.l-1 dans l’eau gélosée contenant 0,5% d’éthanol pour les premiers et de 0, 0,1, 1,et 10 μmol.l-1 dans l’eau gélosée pure pour la quercétine. Les mesures, réalisées après 5 et 7 j de croissance ont montré que le taux de germination n’a été affecté par aucun des composés, que le nombre de tubes germinatifs a été légèrement augmenté par la glycéolline I et que la longueur du mycélium par spore germée a été augmentée par la glycéolline I aux faibles concentrations alors qu’elle a été significativement...

Soybean β-conglycinin α subunit is phosphorylated on two distinct serines by protein kinase CK2 in vitro Inra

Protein kinase CK2 purified from the yeast Yarrowia lipolytica was used to phosphorylate soybean β-conglycinin agr subunit. CK2 is known to phosphorylate serines and threonines in the consensus sequence Ser/Thr-X-X-Glu/Asp/SerP/TyrP. β-Conglycinin α subunit (68 kDa) presents seven consensus sequences, but only 0.5–1 mol P/mol α subunit was incorporated by CK2. [32P]Phosphorylated β-conglycinin α subunit was cleaved either by cyanogen bromide or by trypsin. 32P was incorporated into the largest cyanogen bromide fragment only (50 kDa, N-terminal) and only two radiolabeled zones were detected after HPLC of the trypsic digest. The corresponding phosphorylated zones were collected and further analyzed by RP-HPLC coupled to electrospray ionization mass...
