Assessment of biochemical methods to detect enzymatic depolymerization of polysaccharides ArchiMer

To find biocatalyst allowing depolymerization of new polysaccharides, one needs to get a sensitive and well adapted method to a screening program. This led us to compare biochemical methods of detection of the depolymerization. Currently used methods such as reducing sugars assays, double bond monitoring or molecular weight determination were tested to follow the kinetic of depolymerization with different enzyme / polysaccharide couples. The range of concentrations of assorted enzymes allowed us to identify the most sensitive and appropriate method to detect polysaccharide degradation. Determination of microalgae cellular composition after phycoremediation of swine wastewaters. Repositório Alice

Phycoremediation of swine wastewaters has been widely reported as an attractive tertiary treatment system, that effectively removes the excessive nutrient loadswhilst offering a valuable source of feedstock biomass. Digestate from an upflow anaerobic sludge blanket (UASB, 6%v/v) and a nitrification reactor (NR; 50% v/v) were used as culturing media to microalgae. Experiments were carried out in lab scale photobioreactors (PBRs) using a consortia of Chlorella and Scenedesmus. Ammonia (44 to 90%) and phosphorus (77%) were efficiently removed from both effluents tested after 4 days. Effect of sediment nearness on the metabolic enzyme activity and energy state of the oyster Crassostrea gigas ArchiMer

This study was designed to assess in situ the effects of sediment nearness on Crassostrea gigas metabolism. One year-old oysters were reared for 5 months (April-August) in plastic bags on metallic frames at 10 and 60 cm off the sediment at the beginning of April. The management of the energetic resources (storage, consumption) was estimated with respect to biochemical changes of proteins, lipids and carbohydrates. Enzyme-assisted extraction (EAE) for the production of antiviral and antioxidant extracts from the green seaweed Ulva armoricana (Ulvales, Ulvophyceae) ArchiMer

In order to develop innovative methods for the production of bioactive extracts with potential health benefits, enzyme-assisted extraction has been applied for the bioconversion of Ulva armoricana (Ulvales, Ulvophyceae). The extraction yields, chemical composition, the antioxidant and antiviral activities were determined to assess the efficiency of six commercial enzymatic preparations. Endo-protease treatments significantly increased the extraction yields compared to the control. The organic matter, neutral sugar and protein contents were increased in all extracts compared to an extraction with water, up to 2.0-fold, 2.7-fold and 1.75-fold, respectively. This review considers the importance of compartmentation in the regulation of carbohydrate metabolism in leaves. We draw particular attention to the role of the vacuole as a site for storage of soluble sugars based on sucrose, and discuss briefly their characteristic metabolism. We also point out inconsistencies between the observed properties of vacuoles and the behaviour in vitro of the enzymes of fructan biosynthesis that do not support the hypothesis that the vacuole is the site of synthesis as well as of storage. Mimosa tenuiflora (Mimosaceae) or “jurema-preta” is well distributed in the northeast Brazil, being popularly used to treat skin lesions, burns and inflammation. The healing effect of the alcoholic extract prepared with its barks corroborates the popular use. This study aimed to evaluate the inflammatory response of polysaccharides extracted from M. tenuiflora barks (EP-Mt) by methanol/NaOH and ethanol precipitation. Inflammatory activity was assessed in rat models of acute inflammation (paw edema and peritonitis), by the following parameters: edema, vascular permeability, leukocyte migration, myeloperoxidase activity and pharmacological modulation of nitric oxide and prostaglandins. Proteolytic release and partial characterization of human sperm-surface glycopeptides BJMBR
Sperm-surface glycopeptides were obtained from intact sperm membranes after proteolytic release by different enzymatic treatments such as autoproteolysis, trypsin, papain and pronase. Glycopeptides were isolated, their properties and composition were examined, and their monosaccharide and amino acid constituents were characterized. The monosaccharides identified were fucose, mannose, galactose, N-acetylglucosamine, and N-acetylgalactosamine, which form part of more than one type of oligosaccharide units. Estimates of carbohydrate assimilation within the mesozooplankton compartment are a fundamental concern in oceanography because they give a general expression of the biologically-regulated incorporation of metabolic fuel produced from inorganic nutrients by phytoplankton in the sea. Root carbohydrate storage in young saplings of an Amazonian tidal várzea forest before the onset of the wet season Acta Botanica

Root starch and glucose content were measured for young saplings of 18 Amazonian tidal várzea tree species during a dry season. The pattern of carbohydrate storage depended on the type of plant involved and soil topography which is directly linked to flood regime. Most plants showed high root carbohydrate content at this point in the dry season, however, several typically flood-tolerant species (particularly palm trees) presented a low root carbohydrate content, suggesting a strategy of acquiring reserves during the wet season to survive the dry season, when depletion occurs. Plant survival in these flood-prone forests seems to be the result of more than only one adaptive mechanism.

Soluble sugars and starch accumulation on cotton plant leaves affected by cotton leafroll dwarf virus. Repositório Alice

The cotton blue disease, a main cotton pest in Brazil, is caused by a luteovirus, named Cotton leafroll dwarf virus (CLRDV) or Cotton vein mosaic virus (CVMV). Symptoms include shortening of internodes and stunting. Luteoviruses have been reported to cause carbohydrates accumulation in infected plants. The aim of this research was to verify if soluble sugars and starch accumulation occurs in leaf of cotton plants infected by the virus. Leaf petioles of the cotton plants with symptoms of cotton blue disease presented greater brix values than the leaves of healthy plants. Aim of this research was to investigate physical characters and some chemical characters of northeast local rice grain of Thailand. Physical characters were studied in term of weight, width and size of seed and dehusked seed including elongation rate and water absorption rate of 15 rice cultivars (cv.). Chemical test were checked by protein content, amylose content, lipid content, carbohydrate content, vitamin B1, vitamin B2, vitamin B3 and vitamin B6 content.
