Adsorption capacity of phenolic compounds onto cellulose and xylan Ciênc. Tecnol. Aliment.
Costa,Telma dos Santos; Rogez,Hervé; Pena,Rosinelson da Silva.
The interaction between three phenolic compounds (catechin, caffeic acid and ferulic acid) onto two dietary fibres (cellulose and xylan) has been evaluated to inquire possible interferences on the biodisponibility of phenolic compounds. The adsorption kinetics were performed using solutions containing 100 mg/L of phenolic compounds during a contact time ranging between 10 and 120 minutes at pH 2.0, 4.5, and 7.0. After the kinetics, isotherms were obtained using phenolic compounds concentration ranging between 10 and 80 mg/L during 60 minutes, at pH 2.0 and 7.0 and temperature of 36 °C. Results indicate that adsorbed quantities mainly changed in function of pH, however the maximum adsorption was only of 0.978 mg of caffeic acid/g of xylan at pH 2 and after...

Han,Lirong; Feng,Juntao; Zhang,Shuangxi; Ma,Zhiqing; Wang,Yonghong; Zhang,Xing.
The efficiency of enzymatic hydrolysis of cellulose can be improved by various pretreatments of the substrate. In order to increase the efficiency of enzymatic saccharification of the wheat straw, we determined the effect of different pretreatments on the physical structure, chemical components and enzymatic saccharification of wheat straw. Our results showed that combination of grinding and sodium hydroxide (NaOH) treatment had high effect on the enzymatic hydrolysis of wheat straws. The optimal pretreatment condition was to grind the wheat straws into the sizes of 120 meshes followed by treatment with 1.0% NaOH for 1.5 h (121°C/15psi). Under this condition, the cellulose content of wheat straw was increased by 44.52%, while the content of hemicellulose...

Bottino,Flávia; Cunha-Santino,Marcela Bianchessi; Bianchini Jr.,Irineu.
Abstract Considering the importance of lignocellulose macrophyte-derived for the energy flux in aquatic ecosystems and the nutrient concentrations as a function of force which influences the decomposition process, this study aims to relate the enzymatic activity and lignocellulose hydrolysis in different trophic statuses. Water samples and two macrophyte species were collected from the littoral zone of a subtropical Brazilian Reservoir. A lignocellulosic matrix was obtained using aqueous extraction of dried plant material (≈40 °C). Incubations for decomposition of the lignocellulosic matrix were prepared using lignocelluloses, inoculums and filtered water simulating different trophic statuses with the same N:P ratio. The particulate organic carbon and...

Gréggio,Thiago Claudino; Assis,Luiz Carlos; Nahas,Ely.
The decomposition of soil litter contributes to maintaining agricultural sustainability, since the nutrients released by microbial activity are determinants of the net productivity of the agroecosystem. The decomposition of rubber tree (Hevea brasiliensis [Willd. ex A. Juss.] Müll. Arg.) leaves located on the surface and buried in at 10 cm depth in Forest and Savannah (Cerradão) soils (Oxisols) was studied, with emphasis on the production of CO2 and the monthly variation of the remaining amounts of litter mass, soluble substances, cellulose and lignin. To evaluate CO2 production, H. brasiliensis leaves were incubated for 30 days in soils from the 0-2 cm and 10-12 cm layers. CO2 production increased in the 0-2 cm layer in comparison to the 10-12 cm layer....

Silva,Rafael Lopes da; Araújo,Eric Portilho de; Rocha,Mariucha Karina Honório Ribeiro; Damasceno,Flavia Mota; Freitas,Jakeline Marcela Azambuja de; Sartori,Maria Márcia Pereira; Meirelles,Paulo Roberto de Lima; Barros,Margarida Maria; Pezzato,Luiz Edivaldo.
Abstract: The objective of this work was to evaluate the diet components - crude fiber, neutral detergent fiber, acid detergent fiber, hemicellulose, cellulose, and lignin - as internal indicators in the determination of the apparent digestibility coefficients of dry matter, gross energy, and of the nutrients - crude protein, phosphorus, and amino acids - in Nile tilapia (Oreochromis niloticus). Groups of ten juveniles (80.3±1 g) were randomly distributed in six tanks of 250 L and fed two practical diets, either of a plant-origin diet or of a plant- and animal-origin diet. Both diets were supplemented with 0.1% chromium (III) oxide (Cr2O3). Faeces were collected by the modified Guelph system. The apparent digestibility coefficients were determined by the...

Catani,M.; Amâncio,O.M.S.; Fagundes-Neto,U.; Morais,M.B..
The objective of the present study was to determine the effect of cellulose on intestinal iron absorption in rats during recovery from iron deficiency anemia. Twenty-one-day-old male Wistar-EPM rats were fed an iron-free ration for two weeks to induce anemia. At 5 weeks of age, the rats were divided into two groups (both groups receiving 35 mg of elemental iron per kg diet): cellulose group (N = 12), receiving a diet containing 100 g of cellulose/kg and control (N = 12), receiving a diet containing no cellulose. The fresh weight of the feces collected over a 3-day period between the 15th and 18th day of dietary treatment was 10.7 ± 3.5 g in the group receiving cellulose and 1.9 ± 1.2 g in the control group (P<0.001). Total food intake was higher in the...

Efficient expression and characterization of a cold-active endo-1, 4-β-glucanase from Citrobacter farmeri by co-expression of Myxococcus xanthus protein S Electron. J. Biotechnol.
Bai,Xi; Yuan,Xianjun; Wen,Aiyou; Li,Junfeng; Bai,Yunfeng; Shao,Tao.
Background: Cold-active endo-1, 4-β-glucanase (EglC) can decrease energy costs and prevent product denaturation in biotechnological processes. However, the nature EglC from C. farmeri A1 showed very low activity (800 U/L). In an attempt to increase its expression level, C. farmeri EglC was expressed in Escherichia coli as an N-terminal fusion to protein S (ProS) from Myxococcus xanthus. Results: A novel expression vector, pET(ProS-EglC), was successfully constructed for the expression of C. farmeri EglC in E. coli. SDS-PAGE showed that the recombinant protein (ProS-EglC) was approximately 60 kDa. The activity of ProS-EglC was 12,400 U/L, which was considerably higher than that of the nature EglC (800 U/L). ProS-EglC was active at pH 6.5-pH 8.0,...

Generation and analysis of an Eucalyptus globulus cDNA library constructed from seedlings subjected to low temperature conditions Electron. J. Biotechnol.
Rasmussen-Poblete,Susana; Valdes,Jorge; Gamboa,Maria Cecilia; Valenzuela,Pablo D.T; Krauskopf,Erwin.
Eucalyptus globulus is the most important commercial temperate hardwood in the world because of its wood properties and due to its characteristics for biofuel production. However, only a very low number of expressed sequence tags (ESTs) are publicly available for this tree species. We constructed a cDNA from E. globulus seedlings subjected to low temperature and sequenced 9,913 randomly selected clones, generating 8,737 curated ESTs. The assembly produced 1,062 contigs and 3,879 singletons forming a Eucalyptus unigene set. Based on BLASTX analysis, 89.3% of the contigs and 88.5% of the singletons had significant similarity to known genes in the non-redundant database of GenBank. The Eucalyptus unigene set generated is a valuable public resource that...

Interrelations entre des communautes mixtes de bacteries anaerobies isolees de sediments marins ArchiMer
Marty, D.
The mineralization of cellulose was examined in mono and cocultures of marine anaerobic bacteria (cellulolytic bacteria, fermentative non-cellulolytic bacteria and methane-producing bacteria). These studies displayed interactions between these groups of microorganisms: inhibition relationships (inhibition of methanogens during active growth of fermentative bacteria) and syntrophic relationships (growth of fermentative non-cellulolytic bacteria depended on metabolites produced by cellulolytic bacteria; increased in rate of cellulose hydrolysis in presence of fermentative non-cellulolytic bacteria; interspecies H sub(2) transfer reaction between fermentative bacteria and methane-producing bacteria).

Materials from renewable resources: new properties and functions Anais da ABC (AABC)
GALEMBECK,FERNANDO; BURGO,THIAGO A.L.; SILVA,DOUGLAS S. DA; SANTOS,LEANDRA P..
Abstract: Sustainable production requires increasing use of raw materials from renewable sources, processed under mild conditions with minimal effluent production. These requirements are satisfied by using materials derived from biomass, in synergy with food and energy production. The possibilities of biomass are continuously enlarged by new findings, as in the intrinsic nanocomposite properties of natural rubber and the amphiphile behavior of cellulose that translated into new functional materials, including high-performance, flexible and conductive non-metallic materials. Other findings are allowing a better understanding of electrostatic phenomena that play a positive role in electrostatic adhesion and cohesion of nanocomposites made from biomass...
