Agar is the most widely used gelling agent in the preparation of semi-solid culture media in plant micropropagation. However, because it is one of the most costly components, alternative gelling agents have been prospecting. The...

Castro,Gabriela Fernandes Pepe da Silva de; Rizzo,Roberta Ferreira; Passos,Thaís Souza; Santos,Beatriz Nascimento Corrêa dos; Dias,Daiana da Silva; Domingues,Josiane Roberto; Araújo,Kátia Gomes de Lima.
In biotechnological processes, the culture media components are responsible for high costs and exert a strong influence on the cyanobacteria behavior. The objective of this study was to evaluate the Arthrospira platensis growth potential for biomass production under different cultivation conditions using an experimental design. Three factors that are important for cyanobacteria growth were evaluated: sodium bicarbonate (9 to 18 g/l), sodium nitrate (1.25 to 2.5 g/l), and irradiance (20 to 120 µmol photons/m2.s–1). The results showed that the concentration of NaNO3 in the A. platensis medium can be reduced, resulting in increased concentrations of biomass produced. There was a higher biomass production due to the increase in the concentration of NaHCO3 and...

Rezende,Juliana Costa de; Carvalho,Carlos Henrique Siqueira de; Pasqual,Moacir; Santos,Ana Carolina Ramia; Carvalho,Stephan Malfitano de.
Three experiments were carried out with the objective of achieving high effectiveness in calli induction from high heterozygosis leaf explants of Coffea arabica through indirect somatic embryogenesis. A randomized-block design in a 2x5 factorial arrangement made up of two media [BOXTEL & BERTHOULY (1996) and TEIXEIRA et al. (2004)] and five C. arabica genotypes were used in the first experiment. In the second experiment the embryogenic calli production potential was evaluated in ten genotypes. Each of them was considered as a treatment. In the third experiment the variations in both 2.4-D (2.5 e 20µM) and 2-iP (2.5 e 20µM) concentrations in TEIXEIRA et al. (2004) medium and secondary media were evaluated. Crops were kept in a growth room under...

Chlorophyll a fluorescence of sweet potato plants cultivated in vitro and during ex vitro acclimatization Braz. J. Plant Physiol.
Cassana,Francine F.; Falqueto,Antelmo R.; Braga,Eugenia J. B.; Peters,Jose A.; Bacarin,Marcos A..
Sweet potato (Ipomoea batatas L.) plants were cultivated in vitro in Murashige and Skoog (MS) medium with 20 and 40 g L-1 of sucrose under two different photon flux densities (21 and 60 µmol m-2 s-1). Leaves developed in vitro mostly showed high variable to maximum fluorescence ratio (F V/F M) , indicating good development of photosynthetic apparatus. This ratio decreased during leaf aging, especially in the plants cultivated in vitro on medium with higher sucrose concentration and higher photon flux. Leaves developed ex vitro showed high F V/F M ratio during whole experiment. The effective photochemical efficiency (F V'/F M') was maximum at 15th day after emergence of leaves. Photosynthetic potential rate was higher in leaves developed in vitro than in...

Deleterious effect of TRIS buffer on growth rates and pigment content of Gracilaria birdiae Plastino & E.C. Oliveira (Gracilariales, Rhodophyta) Acta Botanica
Ursi,Suzana; Guimarães,Melina; Plastino,Estela Maria.
This work evaluated the effects of Tris (hydroxymethyl)-aminomethane (TRIS) buffer and its interaction with nutrient concentration on the development of Gracilaria birdie, a common species on the Brazilian coast that has been exploited for agar production. Responses to different conditions were assessed through growth rates and pigment content (chlorophyll a, phycoerythrin, phycocyanin and allophycocyanin). Provasoli's nutrient solution with and without TRIS addition was tested at concentrations of 12.5, 25 and 50%. The pH was also monitored. G. birdiae grew better in the absence of TRIS and at low nutrient concentrations, 12.5 and 25% (growth rates of 10.8-11.3%.day-1). Higher contents of phycoerythrin and chlorophyll a were observed without TRIS at 12.5...

Elaboration and evaluation of a new screening medium for detection and presumptive identification of extended-spectrum beta-lactamase-producing organisms (ESBL) BJM
Silva,Carlos Henrique Pessôa de Menezes e.
The new ß-lactamases have arisen largely as a consequence of heavy use of new expanded spectrum ß-lactam antibiotics. Health professionals need to be aware of the resistance problems caused by the new enzymes, and know the correct procedures to detect, prevent and control such problems. In this study, a new screening medium called Ceftazidime-Inositol-Vancomycin-Amphotericin B Agar (CIVA) was elaborated and the microbiological performance was evaluated for the detection and presumptive identification of ESBL-producing members of Enterobacteriaceae. It was performed in 126 stool samples from hospitalized patients at Santa Monica Hospital (Vila Velha, ES, Brazil), who had been heavily exposed to broad-spectrum antibiotic combinations. The bacteria were...

Filtration enrichment method for isolation of auxotrophic mutants of Trichoderma harzianum Rifai. Repositório Alice
CASSIOLATO, A. M. R.; MELO, I. S. de.
The isolation of genetic markers, like drug resistance and auxotrophy, is a laborious but important step in genetic research. The isolation of auxotrophic mutants of Trichoderma harzianum using the filtration enrichment technique was more effective than using the total isolation technique. Most of 12 auxotrophic mutants exhibited similar growth rate and higher sporulation when compared with the wild type, but only two mutants (TWS-410 and TW5-523) could grow in 500ug/L of benomyl.

In vitro seed germination and seedling development of Annona crassiflora Mart. Scientia Agricola
Ribeiro,Márcia de Nazaré Oliveira; Pasqual,Moacir; Villa,Fabíola; Pio,Leila Aparecida Salles; Hilhorst,Henk William Maria.
Annona crassiflora Mart known as 'araticum', 'marolo' or 'field araticum' is a typical fruit from the Cerrado biome of Brazil with socio-economic and medicinal importance. Normally, Annona crassiflora is propagated through seeds. However, due to a deep dormancy that the seeds display at dispersion and the difficulty to obtain uniform plants in a short time period, micropropagation may be a feasible alternative. Concentrations of gibberellic acid (GA3) and naphthalene-acetic acid (NAA) and their interactive effects on in vitro seed germination and seedling development of Annona crassiflora were studied. Mature fruits of Annona crassiflora were depulped and the seeds washed in clear water and dried at room temperature. Seed coat was removed and the seeds...
