A rapid quantitative fluorescence-based bioassay to study allelochemical interactions from Alexandrium minutum ArchiMer

Harmful microalgal blooms are a threat to aquatic organisms, ecosystems and human health. Toxic dinoflagellates of the genus Alexandrium are known to produce paralytic shellfish toxins and to release bioactive extracellular compounds (BECs) with potent cytotoxic, hemolytic, ichtyotoxic and allelopathic activity. Negative allelochemical interactions refer to the chemicals that are released by the genus Alexandrium and that induce adverse effects on the physiology of co-occurring protists and predators. Releasing BECs gives the donor a competitive advantage that may help to form dense toxic blooms of phytoplankton. However BECs released by Alexandrium minutum are uncharacterized and it is impossible to quantify them using classical chemical methods....

Action of Ponceau 4R (E-124) food dye on root meristematic cells of Allium cepa L. Biological Sciences

This study aimed to evaluate the toxicity of Ponceau 4R food dye on the cell cycle in root meristematic cells of Allium cepa L. at three concentrations: 0.25, 0.50 and 0.75 g L-1, at exposure times of 24 and 48 hours. For each concentration, we used a set of five onion bulbs that were first rooted in distilled water and then transferred to their respective concentrations. Radicles were collected and fixed in acetic acid (3:1) for 24 hours. The slides were mounted with the crushing technique and stained with 2% acetic orcein. Activity of the aqueous extract from Polymnia sonchifolia leaves on growth and production of aflatoxin B1 by Aspergillus flavus BJM

The aqueous extract from Polymnia sonchifolia leaves (AE) was tested for inhibitory activity on aflatoxin B1(AFB1) production and growth of Aspergillus flavus. The cytotoxicity of AE on Vero cells was also performed. Suspensions of A. flavus spores were inoculated into 50 mL of YES medium together with different concentrations of the AE. The aflatoxin B1 was extracted, analyzed by thin layer chromatography and quantified by photodensitometry. All the concentrations of AE induced inhibition of AFB1 production. The aqueous extract showed in vitro cytotoxicity to Vero cells only at concentrations above 500 µg/mL.

The antiviral potency of Caulerpa cupressoides (Chlorophyta) sulfated polysaccharidic fractions (Cc-SP1→3) associated with thrombin generation (TG) assay is unknown. This study analyzed the structure of Cc-SP1 and its effects against herpes viruses (HSV-1 and HSV-2), dengue-virus (DENV-1), human-metapneumovirus (HMPV) and adenovirus (AdV) in Vero and mosquito C6/36 cell-lines and as inhibitor of TG in 60-fold diluted human plasma using continuous system. Bioactive Bromotyrosine-Derived Alkaloids from the Polynesian Sponge Suberea ianthelliformis ArchiMer

Herein, we describe the isolation and spectroscopic identification of eight new tetrabrominated tyrosine alkaloids 2–9 from the Polynesian sponge Suberea ianthelliformis, along with known major compound psammaplysene D (1), N,N-dimethyldibromotyramine, 5-hydroxy xanthenuric acid, and xanthenuric acid. Cytotoxicity and acetylcholinesterase inhibition activities were evaluated for some of the isolated metabolites. They exhibited moderate antiproliferative activity against KB cancer cell lines, but psammaplysene D (1) displayed substantial cytotoxicity as well as acetylcholinesterase inhibition with IC50 values of 0.7 μM and 1.3 μM, respectively.

We investigated the in vitro process of cell death caused by Egyptian cobra venom on primary human embryonic kidney (293T) and mouse myoblast (C2C12) cell lines. The aim of these studies was to provide further information about triggering cell death, and suggest methods for eliminating unwanted cells, such as tumour cells. Both cell lines were treated with 10, 20, and 50 m g/ml of Egyptian cobra (Naja haje) venom in serum free media (SFM) and incubated for 8 hours. Many chemotherapeutic agents with a potential against solid tumors or leukemia can cause lymphopenia. Tamoxifen (TAM) is a synthetic non-steroidal anti-estrogen drug employed in female breast cancer treatment. The present study investigated the capacity of TAM to induce cell death in human lymphocytes cultivated in vitro. Lymphocytes were obtained from young (25-30 years; n = 3) and elderly women (58-77 years; n = 3) and cultivated for 24 or 48h, with or without TAM (20 &micro;M). Different materials are used for prostheses in dental medicine, though titanium is the most frequent. In the present report, several commercial dental implants and their potential capacity to integrate to bone tissues have been analysed by means of cytotoxicity, genotoxicity and cytocompatibility techniques. Cultures from osteosarcoma-derived UMR 106 cell line were used for bioassays since they showed several osteoblast properties. Cytological and cytogenic effects induced by thiabendazole on Allium cepa root meristem JBAG

The toxicity and genotoxicity of thiabendazole (TBZ) in its commercial formulation Foldan® was studied in Allium cepa meristematic cells at concentrations ranging between 10 and 250 µg / ml. Our results show that exposure to TBZ induces a significant increase in the frequency of anaphase telophase chromosomal aberrations, micronuclei and binucleated cells at all the concentrations of TBZ assayed. The frequency of anaphase telophase aberrations was observed to reach a plateau at the concentration of 50 µg / ml of TBZ, which could be probably due to TBZ mitotic toxicity manifested at concentrations of 100 and 250 µg / ml. Mussel (Mytilus galloprovincialis) plasma contains cytotoxic activity against both vertebrate (erythrocytes and mouse tumour) and protozoan cells. Procaryotes (Escherichia coli and Vibrio alginolyticus) were not sensitive to the cytotoxicity. The activity was still present in dialyzed samples but was inhibited by heating at 45°C. Large individual variability which was not correlated with protein concentration and an increasing number of reactive specimens following injection, suggested that naive mussels were in various stages of immune response. Scorpion venom toxicity is of major concern due to its influence on human activities and public health. The cytotoxicity and apoptosis induced by scorpion L. quinquestriatus venom on two established eukaryotic cell lines (293T and C2C12) were analyzed. Both cultured cell lines were incubated with varying doses (10, 20, and 50 µg/ml) of scorpion venom in serum free medium (SFM) for 0.5, 1, 2, 4, and 8 hours at 37°C.
