Calreticulin (CRT), a Ca2+-binding storage protein and chaperone in the endoplasmic reticulum, modulates cell adhesiveness and integrin-dependent Ca2+ signaling. However, the role of CRT during implantation remains poorly understood. In the present study, we characterized the expression of CRT mRNA and the protein in mouse endometria from pregnancy DI to D7. Real-Time PCR and in situ hybridization results showed that the levels of CRT mRNA in the endometria of pregnant mice were significantly higher than those of non-pregnant mice (P<0.05), and increased gradually from pregnancy DI to D4, reaching the máximum level on D4, followed by a plateau from D4 to D7. Using immunofluorescence histochemistry and western blot, changes of CRT expression in the...

Co-culture of Mouse Two-cell Embryos on Human Endometrial Stromal Cells in Proliferative and Secretory Phases International Journal of Morphology
Fakheri,Taravat; Nankali,Anisodowleh; Khazaei,Mozafar; Ghanbari,Ali; Abedini,Mehrandokht; Saeidiborojeni,Hamidreza.
There were no significant differences in the distribution of embryos reaching to 2- cells, 4- cells, morula or blastocysts culturing on human endometrial stromal cells (Secretory or proliferative phases). The percent of morula in stage A (without fragmentation), stage B (<25% fragmentation), stage C (25-50% fragmentation) and stage D (>50% fragmentation) and did not showed significant differences between two coculture groups. Thus, the phase that the endometrial stromal cells were in thereby did not affect on the quality of embryos.

Cycle modulation of insulin-like growth factor-binding protein 1 in human endometrium BJMBR
Corleta,H.; Capp,E.; Strowitzki,T..
Endometrium is one of the fastest growing human tissues. Sex hormones, estrogen and progesterone, in interaction with several growth factors, control its growth and differentiation. Insulin-like growth factor 1 (IGF-1) interacts with cell surface receptors and also with specific soluble binding proteins. IGF-binding proteins (IGF-BP) have been shown to modulate IGF-1 action. Of six known isoforms, IGF-BP-1 has been characterized as a marker produced by endometrial stromal cells in the late secretory phase and in the decidua. In the current study, IGF-1-BP concentration and affinity in the proliferative and secretory phase of the menstrual cycle were measured. Endometrial samples were from patients of reproductive age with regular menstrual cycles and...

Distribution of versican and hyaluronan in the mouse uterus during decidualization BJMBR
San Martin,S.; Soto-Suazo,M.; Zorn,T.M.T..
Preparation for embryo implantation requires extensive adaptation of the uterine microenvironment. This process consists of cell proliferation and cell differentiation resulting in the transformation of endometrial fibroblasts into a new type of cell called decidual cell. In the present study, we followed the space-time distribution of versican and hyaluronan (HA) in different tissues of the uterus before and after embryo implantation. Fragments of mouse uteri obtained on the fourth, fifth, sixth and seventh days of pregnancy were fixed in Methacarn, embedded in Paraplast and cut into 5-µm thick sections. HA was detected using a biotinylated fragment of the proteoglycan aggrecan, which binds to this glycosaminoglycan with high affinity and specificity....

Expression of uterine lipocalin 2 and its receptor during early- to mid-pregnancy period in mares OAK
Haneda, Shingo; Nagaoka, Kentaro; Nambo, Yasuo; Kikuchi, Masato; Nakano, Yasuko; Li, Junyou; Matsui, Motozumi; Miyake, Yo-ichi; Imakawa, Kazuhiko.
From previous cDNA subtraction studies analyzing gene expression in equine endometrium, high lipocalin 2 (LCN2) mRNA expression was found in the gravid endometrium. In the uterus, LCN2 may transport hydrophobic molecules and siderophores with iron, or may form a complex with another protein, however, the expression of uterine LCN2 beyond day 20 of equine pregnancy and its receptor has not been characterized. To study the expression and potential roles of uterine LCN2 from pre-implantation to mid-gestation period, stage-specific endometrial samples were obtained from day 13 (day 0 = ovulation) cyclic and days 13, 19, 25, and 60 to 131 pregnant mares. Expression of LCN2 mRNA increased in day 19 gravid endometrium and was abundant from day 60 onward. The...

Histochemical Study of Early Embryo Implantation in Rats International Journal of Morphology
Spósito,Danilo Rodrigues; Santos Jr,Arnaldo Rodrigues.
Embryo implantation is the process that results in attachment of the conceptus to the uterine wall. In this histochemical study, we investigated the early stage of embryo implantation in rats by morphological analysis and by the detection of total proteins and glycosaminoglycans using hematoxylin-eosin, toluidine blue at pH 4.0 (TB), and Xylidine Ponceau at pH 2.5 (XP). In non-pregnant females, the uterine layers could be clearly distinguished and showed the normal histology of the organ. In pregnant females, an increase in the number of cells and a reduction in the interstitial space were observed in the endometrium close to the implantation sites. The blastocyst was partially inserted in the endometrium, with the observation of the inner cells mass...

ULTRASTRUCTURAL ANALYSIS OF PINEALECTOMY AND LACK OF LIGHT INFLUENCE OVER COLLAGEN IN THE ENDOMETRIUM OF RATS International Journal of Morphology
Pinto de Medeiros,Juliana; Wanderley Teixeira,Valéria; Aguiar Coelho Teixeira,Álvaro; Baratella Evêncio,Liriane; Evêncio Neto,Joaquim.
Some studies have shown that pinealectomized female rats present a large concentration of collagen fibers in the endometrium. The lack of light results in morphologic alterations in the uterine lamina propria and in the muscular layer, reducing thus the uterine weight. Therefore, this work aimed to analyze ultra-structurally the influence of melatonin in the remodeling of the collagen in the endometrium, in the estrus phase, in pinealectomized adult rats or submitted to lack of light. The experimental groups: I- sham-pinealectomized maintained in a clear/dark cycle 12/12 hours, during three months; II- pinealectomized rats maintained in a clear/dark cycle 12/12 hours, for three months; III- normal rats maintained in clear/dark cycle 12/12 hours for three...
