Continuous enzymes and fungal metabolites production in solid state fermentation using a counter-current reactor IRD

This work presents the continuous production of fungal biomass and enzymes by solid state fermentation (SSF) in a counter-current reactor adapted for this purpose. Pre-germinated conidia of #Aspergillus niger$ were used as an inoculum and sugarcane bagasse, embedded with a nutritive solution, was the solid support. The solids residence time distribution (RTD) was carried out by feeding one impulse of blue-coloured humidified bagasse and this RTD was fixed at 20 hours. This study demonstrated that the values of the measured parameters (pH, moisture, biomass, glucoamylase production) were similar to those reported for batch SSF using the same solid support and micro-organism. Contribution à l'étude physiologique et enzymologique d'une nouvelle souche de Lactobacillus plantarum amylolytique isolée du manioc fermenté IRD

Afin de maîtriser l'étape de fermentation intervenant dans la majorité des procédés traditionnels de transformations des racines de manioc, les caractéristiques de croissance et de production d'acide lactique d'un certain nombre de bactéries lactiques ont été examinées. Une importante biomasse algale s'échoue, chaque année, de novembre à juin, en certains endroits de la côte du Sénégal. Cette biomasse, qui n'est pas utilisée, contribue à la pollution des plages. C'est pour cela que les auteurs ont tenté, en utilisant des procédés de biotechnologie de valoriser cette biomasse par la filière biogaz-compost , la transformant en deux sous-produits : un biogaz (combustible) et un compost à usage agronomique. Kinetics of the solid state fermentation of raw cassava flour by Rhizopus formosa 28422 IRD

The strain #Rhizopus formosa$ 28422 was selected from the stock of ten strains from genera #Rhizopus$, for their capacity to attack raw cassava starch by solid substrate fermentation and showed the highest growth in this substrate. The optimal substrate composition, estimated by surface response design experiments, was 10% cassava bagasse, 10% soybean flour and 80% cassava flour. Optimal fermentation conditions were temperature, 32°C, moisture, 64%, initial pH, 6.5 and inoculum rate, 10exp6 spores/g DM. Maintenance of heat and water balances as a scale-up criterion for the production of ethanol by Schwanniomyces castellii in a solid state fermentation system IRD

The scale-up of column fermenters by 6 to 410 gravimetric scale factors, from 10 g moist substrate size, has been achieved efficiently by maintaining heat and water balances in the media. The data on the patterns of ethanol production, biomass formation, the concentration of different carbohydrates, dry matter and pH values against time were of equal magnitude in 10- and 60-g size column fermenters. The reactors of 60-, 370- and 4100-g sizes also compared well in respect of O2 consumption, CO2 evolution and the specific growth rates in aerobic and anaerobic fermentations phases. Molecular techniques applied to fungal strain upgradation capability related to SSF cultures IRD

Mutant strains derived from #A. niger$ C28B25 and belonging to series AW99 [producing maximum pectinase level in submerged fermentation (SmF)] and series AW96 [producing maximum pectinase level in solid state fermentation (SSF)] were compared concerning pectinase properties and thermal stability. Pectinases produced by AW96 exhibited increased thermal stability compared to AW99. Results support the idea that not only the quantity but also the quality of the enzymes can be modified when SSF or SmF techniques are used. The use of differential display polymerase chain reaction may enable the isolation of solid state fermentation-specific genes. (Résumé d'auteur)

On-line automated monitoring and control systems for CO2 and O2 in aerobic and anaerobic solid-state fermentations IRD

Two systems for monitoring and control of gases from solid-state cultures have been developed. The first involves on-line automated monitoring of CO2 and O2 concentration in exhaust gases from eight fermenters or eight gas sampling ports in a large fermenter. It proved to be efficient in obtaining information on the physiological state and respiration rate of the culture in a real-time process. Furthermore, the specific growth rate (mù) can be estimated reliably by gas measurements in aerobic cultures. The second system is for automated control of exit gases from aerobic solid-state fermentations. Production of a Lactobacillus plantarum Starter with linamarase and amylase activities for cassava fermentation IRD

#Lactobacillus plantarum$ strain A6 isolated from cassava, cultured on cellobiose MRS medium showed a growth rate of 0.41 h-1, a biomass yield of 0.22 g g-1, and produced simultaneously an intracellular linamarase (76.4 U g-1 of biomass) and an extracellular amylase (36 U ml-1). The synthesis of both enzymes was repressed by glucose. The use of such a strain as a cassava fermentation starter for gari production had the following influences: a change from a heterofermentative pattern observed in natural fermentation to a homofermentation, a lower final pH, a faster pH decline rate and a grater production of lactic acid (50 g kg-1 DM).
