A hemocyanin-derived antimicrobial peptide from the penaeid shrimp adopts an alpha-helical structure that specifically permeabilizes fungal membranes ArchiMer
Petit, Vanessa W.; Rolland, Jean-luc; Blond, Alain; Cazevieille, Chantal; Djediat, Chakib; Peduzzi, Jean; Goulard, Christophe; Bachere, Evelyne; Dupont, Joelle; Destoumieux-garzon, Delphine; Rebuffat, Sylvie.
Background. Hemocyanins are respiratory proteins with multiple functions. In diverse crustaceans hemocyanins can release histidine-rich antimicrobial peptides in response to microbial challenge. In penaeid shrimp, strictly antifungal peptides are released from the C-terminus of hemocyanins. Methods. The three-dimensional structure of the antifungal peptide PvHCt from Litopenaeus vannamei was determined by NMR. Its mechanism of action against the shrimp pathogen Fusarium oxysporum was investigated using immunochemistry, fluorescence and transmission electron microscopy. Results. PvHCt folded into an amphipathic α-helix in membrane-mimicking media and displayed a random conformation in aqueous environment. In contact with F. oxysporum, PvHCt bound...

Asian soybean rust (ASR), caused by the fungus Phakopsora pachyrhizi, is one of the most economically important crop diseases, but is only treatable with fungicides, which are becoming less effective owing to the emergence of fungicide resistance. There are no commercial soybean cultivars with durable resistance to P. pachyrhizi, and although soybean resistance loci have been mapped, no resistance genes have been cloned. We report the cloning of a P. pachyrhizi resistance gene CcRpp1 (Cajanus cajan Resistance against Phakopsora pachyrhizi 1) from pigeonpea (Cajanus cajan) and show that CcRpp1 confers full resistance to P. pachyrhizi in soybean. Our findings show that legume species related to soybean such as pigeonpea, cowpea, common bean and others could...

A procedure to evaluate the efficiency of surface sterilization methods in culture-independent fungal endophyte studies BJM
Burgdorf,R.J.; Laing,M.D.; Morris,C.D.; Jamal-Ally,S.F..
Extraneous DNA interferes with PCR studies of endophytic fungi. A procedure was developed with which to evaluate the removal of extraneous DNA. Wheat (Triticum aestivum) leaves were sprayed with Saccharomyces cerevisiae and then subjected to physical and chemical surface treatments. The fungal ITS1 products were amplified from whole tissue DNA extractions. ANOVA was performed on the DNA bands representing S. cerevisiae on the agarose gel. Band profile comparisons using permutational multivariate ANOVA (PERMANOVA) and non-metric multidimensional scaling (NMDS) were performed on DGGE gel data, and band numbers were compared between treatments. Leaf surfaces were viewed under variable pressure scanning electron microscopy (VPSEM). Yeast band analysis of the...

A test of community assembly rules using foliar endophytes from a tropical forest canopy ArchiMer
Donald, Julian; Roy, Mélanie; Suescun, Uxue; Iribar, Amaia; Manzi, Sophie; Péllissier, Léonie; Gaucher, Philippe; Chave, Jérôme.
1.Community assembly theory assumes that ecological communities are spatially delimited into patches. Within these patches, coexistence results from environmental filtering, competition, and immigration. Truly delineated communities exist in laboratory studies of microbial cultures in Petri dishes, yet empirical tests conducted in continuous environments often use patches defined by convention as opposed to realised boundaries. 2.Here we perform a test of ecological community assembly rules using foliar endophyte communities from a tropical rainforest, where leaves are considered as patches for both fungal and bacterial communities. We determined the diversity of fungal and bacterial endophytes using environmental DNA sequencing of 365 top‐canopy leaves,...

Abiotic effects on the cover and richness of corticolous lichens on Araucaria angustifolia trunks Acta Botanica
Nunes,Lucas; Burle,Giulia; Gumboski,Emerson Luiz; Dechoum,Michele.
ABSTRACT Lichens play a key role in natural ecosystems, as they can function as primary producers, recycle minerals and fix nitrogen. Despite their environmental importance, little is known about lichen ecology in Brazil, and especially about how abiotic factors may influence their spatial distribution. In this study, we aimed to verify how the cover and richness of corticolous lichens on Araucaria angustifolia trunks vary between two different habitats (Forest and Grassland). The photoquadrat sampling method was applied to A. angustifolia trunks. The Coral Point Count software with Excel extensions (CPCe) was used to analyze photographs for lichen cover and richness. Additionally, a redundancy analysis was conducted to estimate how five abiotic and two...

Acremonium kiliense infection in a child with chronic granulomatous disease BJID
Pastorino,Antonio Carlos; Menezes,Ulissis Pádua de; Marques,Heloisa Helena de Souza; Vallada,Marcelo G.; Cappellozi,Vera Lúcia; Carnide,Eugénia Maria Grilo; Jacob,Cristina Miuki Abe.
Infection by unusual microorganisms can be one of the clinical manifestations of primary immunodeficiency (PID). We report on a four-month-old child with pneumonia caused by the fungus Acremonium kiliense as the first clinical manifestation of chronic granulomatous disease. We emphasize the importance of an active search for unusual organisms in immunodeficient patients, and a precise diagnosis and early institution of specific treatment against such microorganisms for the reduction of the morbidity and mortality of these patients.

Addition of Amylase from Aspergillus Awamori to the Diet of Broiler Chickens Rev. Bras. Ciênc. Avic.
Morgado,HS; Cysneiros,CSS; Sousa,CM; Stringhini,JH; Ulhoa,CJ; Silva,AS; Fabino Neto,R; Freitas,PVDX; Oliveira,HP; Batista,LHC.
ABSTRACT Two experiments were performed to evaluate the hematological and blood biochemistry parameters, biometry of digestive organs, enzyme activities, protein content and absolute weight of the pancreas of broilers fed pre-starter and pre-starter diets supplemented or not with amylase from Aspergillus awamori. In total, 120 male Cobb chicks were housed in heated cages in each experiment. A completely randomized experimental design, with two treatments (feed with and without amylase) and six replicates per treatment of 10 birds each was applied. The data were subjected to analysis of variance using the F-test at 5% probability level. The dietary amylase addition did not affect hematological and blood biochemistry parameters and the biometry of the...

Ríos-Yuil,José Manuel; Arenas,Roberto; Fernández,Ramón; Calderón-Ezquerro,María; Rodriguez-Badillo,Raymundo.
INTRODUCTION: An aeromycological study verifies the presence and quantifies the concentration of fungal propagules in the air. It is very important in the hospital setting because of the increasing numbers of immunosuppressed and severely ill patients. The objective of this study was to determine the concentration of fungi in the air of the intensive care unit (ICU) of "Dr. Manuel Gea González" General Hospital. METHODS: This is a descriptive, observational cross-sectional study. Air samples were obtained with a single stage Thermo-Andersen Viable Particle Sampler (Thermo Electron Corporation -Massachusetts, U.S.A.) in a Petri dish with potato dextrose agar for 15 minutes at two different times (morning and afternoon) and heights (1 and 1.5 meters).

An analysis of the spontaneous mutation rate measurement in filamentous fungi Genet. Mol. Biol.
Baracho,Marta S.; Baracho,Ivanhoé R..
Mutations related to gene methG1 of Aspergillus nidulans were analyzed, in order to study a mathematical model for the determination of the mutation rate per nucleus per generation, in filamentous fungi. A replica plating technique was used to inoculate, in a single operation, 26 colonies of the strain, into Petri dishes containing culture medium, and the nine central colonies were analyzed for size and number of conidia in each colony. Using this technique, several central colonies were analyzed with regard to the appearance of mutation, and the number and type of reversions were determined for each colony. The frequencies obtained for each reversion were analyzed, in order to verify if their distribution was in accordance with that of Greenwood and Yule....

An approach to the study of the fungal deterioration of a classical art material: Mastic varnish Electron. J. Biotechnol.
Romero-Noguera,Julio; López-Miras,María del Mar; Martín-Sánchez,Inés; Ramos-López,José Miguel; Bolívar-Galiano,Fernando.
Mastic, one of the best natural varnishes, is frequently used as protective and finishing layer or as component of oleo-resinous media in paintings, both in the past and currently. However, this resin is affected by complex deterioration processes which can change its characteristics and thus the visual aspect of works of art. The alteration processes caused by radiation have been widely studied, but there is a lack of information on the biodeterioration of this natural product. In this paper, fungi from collections as well as from oil paintings of the Fine Arts Museum of Granada (Spain) were inoculated onto slides covered with mastic. The samples, after an incubation period of 15 days, were analysed by gas chromatography-mass spectrometry (GC-MS) to...
