Using genotypes adapted to different regions is one of the main ways to increase Brazilian bean yield. The aim of the present study was to assess the genotypic performance of Carioca beans through mixed models. Fourteen Carioca bean genotypes were assessed in four locations in Pernambuco State (Arcoverde, Caruaru, Belém de São Francisco and São João counties) in 2015. The experiments followed a completely randomized block design, with three repetitions. Genetic parameters were estimated according to the REML/BLUP methodology, whereas genotype selection was based on the harmonic mean of relative performance of genetic values method (MHPRVG). The mean genotype heritability had moderate magnitude, high selective accuracy, besides allowing selection of...

Dynamics of 45S rDNA sites in the cell cycle: fragile sites and chromosomal stability in Lolium and Festuca. Repositório Alice
ROCHA, L. C.; SILVA, G. A.; BUSTAMANTE, F. O.; SILVEIRA, R. A. D.; MITTELMANN, A.; TECHIO, V. H..
Abstract Analyses carried out with fluorescence in situ hybridization (FISH) in C-metaphases of the Lolium-Festuca complex have shown the occurrence of spontaneous fragile sites (FSs) in 45S rDNA regions. FSs are expressed as gaps but they do not result in breaks or chromosomal fragments in these species. These gaps have high DNA condensation observed as thin chromatin fibers that connect the apparent segments of the fragile chromosome, allowing for genomic stability. Assessing the behavior of these regions in the cell cycle of Lolium and Festuca species may lead to a better understanding of the dynamics that preserve stability during cell division. Furthermore, it is interesting to track the dynamics of chromosomes bearing 45S rDNA sites in the cell cycle...

Evaluation of cassava plants generated by somatic embryogenesis at different stages of development using molecular markers Electron. J. Biotechnol.
Osorio,Mayra; Gámez,Elizabeth; Molina,Sandy; Infante,Diógenes.
Background: Cassava (Manihot esculenta Crantz) is a crop that is high in carbohydrates in the roots and in protein in the leaves, important for both human consumption and animal feed, and also has a significant industrial use for its starches. In this study we evaluated the genetic variability with molecular markers in different stages in micropropagated plants from somatic embryos of Venezuelan native clone 56. Results: Three markers were used: ISTR, AFLP and SSR, finding that ISTR showed the highest polymorphism among individuals tested. With AFLP a high similarity between the evaluated individuals was observed and with SSR total monomorphism was seen. Using cluster analysis it was found that individuals from an embryo labeled as fasciated at the...

Giménez,C; de García,E; Haddad,O.
Evaluation of clonal micropropagation and phenotype stability of elite somaclones are critical steps for development of new varieties. In the present work somaclon variant CIEN BTA-03 (resistant to Black Sigatoka), obtained through in vitro process from cultivar Williams (susceptible to Black Sigatoka), was micropropagated via apical shoot culture for five multiplication cycles in 0.5 mg/l of benzyl-aminopurine (BA). To verify the genetic stability of the progeny of this elite material, random amplified polymorphic DNA (RAPD) markers were used. A total of 5,292 monomorphic bands were obtained from the amplification of fifty six DNA samples (extracted from vitroplants randomly selected) with 10 different primer combinations. Non-polymorphic RAPD bands were...

Genetic stability in rice micropropagation Biocell
Medina,R; Faloci,M; Marassi,M.A.; Mroginski,L.A..
An efficient clonal propagation procedure for six rice varieties cultivated in Argentina was developed by using shoot tip cultures, and the genetic stability of the micropropagated plants was verified by isozyme analysis. One week old seedlings obtained on MS medium were sectioned and subcultured on MS medium (0.75% agar) supplemented with different combination and concentrations of cytokinins (BAP and KIN) and auxins (2,4-D and NAA). After four weeks of culture, multiple shoots were obtained. The best response was observed on MS supplemented with BAP 5 mg l-1. Shoot clumps were multiplied in MS liquid medium containing BAP 5 mg l-1. Profuse rooting was obtained after transfer to MS medium lacking growth regulators and with sucrose 8% (w/v). Complete...

Rescue of a non-viable accession and rapd analysis of recovered plants of Arachis retusa. Repositório Alice
GAGLIARDI, R.F.; PACHECO, G.P.; OLIVEIRA, C.A.; CARNEIRO, L.A.; VALLS, J.F.M.; VIEIRA, M.L.C.; MANSUR, E..
In vitro regeneration of Arachis retusa was examined for the purpose of germplasm renewal and conservation. Random amplified polymorphic DNA (RAPD) fingerprinting was used to evaluate the genetic stability of plants derived from embryo axes and apical segments. Ten arbitrary decamer primers were screened and five of them were selected. Ninety genomic regions were evaluated, with an average of 18 loci per clone. All amplified segments were monomorphic. The results indicate that recovered plants are genetically stable at the assessed genomic regions and that both regeneration processes are suitable for in vitro germplasm preservation of Arachis species.

Stability and adaptability of sweet orange using mixed models. Repositório Alice
CAPISTRANO, M. da C.; ANDRADE NETO, R. de C.; SANTOS, V. B. dos; LESSA, L. S.; RESENDE, M. D. V. de; GURGEL, F. de L..
Evaluations with sweet oranges are usually performed in several harvests and places to verify the differential behavior according to the environmental variations. This makes the selection by traditional statistical methodologies more and more difficult since they have limitations in the case of data unbalanced, common in citrus during the experimental phase due to the possibility of plot loss over the years. The objective of this work was to estimate the temporal stability and adaptability of sweet orange genotypes cultivated in eight crops under the edaphoclimatic conditions of Rio Branco, Acre. The experimental design used was in randomized blocks containing 55 genotypes and three replications. Four agronomic characteristics were evaluated during eight...
