The objective of this study was to evaluate the flow of nitrogenous compounds, protein degradability, rumen degradation of total carbohydrate and organic matter and microbial efficiency in heifers subjected to diets containing corn silage, sugarcane or Tifton. For this purpose, the 2,6-diaminopimelic acid (DAPA) technique was adopted and analytical procedures for amino acids by HPLC were adapted. The objectives of this work were to carry out a comparative chemical study and to evaluate the antinociceptive and anti-inflammatory activities of ethanol extracts (EtOHE) and vanilic acid (VA) from cultivated and wild Amburana cearensis A.C. Smith (Fabaceae), an endangered species used in Northeast Brazil for the treatment of asthma. A comparison between enzyme immunoassay and HPLC for ochratoxin A detection in green, roasted and instant coffee BABT

An indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) for ochratoxin A (OTA) detection in green, roasted and instant coffees was developed using anti-OTA monoclonal antibody. Immunological reagents prepared were OTA-BSA (4.76 mg/mL), anti-OTA.7 MAb (2x10³-fold dilution) and HRP-anti IgG (10³-fold dilution). The detection limit was 3.73 ng OTA/g and correlation coefficients (r) between this immunoassay and high performance liquid chromatography were 0.98 for green coffee, 0.98 for roasted and 0.86 for instant. OTA levels detected by ic-ELISA were higher than by HPLC, with ELISA/HPLC ratio of 0.66 - 1.46 (green coffee), 0.96 - 1.11 (roasted) and 0.93 - 1.82 (instant). Abstract The consumption of yerba mate beverages is related to several health benefits. These desirable properties are mainly due to the bioactive compounds. However, the levels of those compounds are directly affected by factors such as geographical origin. This study aimed to use chemometrics to evaluate the antioxidant compounds of yerba mate (consumed as chimarrão) marketed in southern Brazil. Com objetivo de estudar absorcao, distribuicao e metabolismo de 6-benziladenina (BA) em hyacinthus amethystina cv. Albus (Jacinto pirenaico), utilizaram-se explantes de escamas de bulbos cultivados em meio MS, suplementado com 4,90 M de AIB e 22,19 M de BA. Os explantes foram cultivados em duas posicoes quanto a sua polaridade: dorsal e ventral. Os explantes foram cultivados em presenca de 8-(14C)-BA com atividade especifica de 2,0 GBq/mmol., adicionado ao meio em tal proporcao que cada recipiente de cultivo tivesse uma atividade de 940KBq. ABSTRACT The acaricidal activity of crude ethanolic extract and fractions from the leaves of Morus nigra (Moraceae) was carried out on female cattle ticks Rhipicephalus microplus, using the adult immersion test. The mortality and fertility of females exposed to different concentrations of hexane, chloroform and ethyl acetate fractions, as well as ethanolic extract of M. nigra with concentrations of 5, 10 and 25mg/ml were evaluated using three treatment groups, two control groups and triplicate tests. Accumulation and extractability of grape skin tannins and anthocyanins at different advanced physiological stages Inra

Quantitative and qualitative modifications of tannins and anthocyanins in grape skin were investigated at different dates of harvest, from berries sorted on the basis of their density. Free anthocyanins accumulated until 170 g/L of sugars in pulp before undergoing a slight decrease. Changes in anthocyanin composition were observed with increasing sugar levels in the pulp that reflected structural differences between classes of anthocyanins. The proportion of methoxylated anthocyanins continued to increase in the skin as sugar accumulated while the proportion of coumaroylated anthocyanins initially increased (up to 200 g/L of sugars in the pulp) and then rapidly decreased. Nonylphenol ethoxylates (NPEOs), which are widely used for industrial and domestic purposes, exert adverse effects on wildlife after being used and discharged into the environment. However, their ultimate biodegradability and biodegradation pathway remains unclear. In this study, the aerobic degradability of nonylphenol ethoxylates (NPEOs) by the acclimated microorganisms in active sludge was examined using shaking-flask tests. The degradation of benzene rings in NPEOs was determined using UV spectroscopy and high performance liquid chromatography (HPLC). Milk products such as cheeses may be contaminated by aflatoxin M1 when dairy cattle have consumed feeds contaminated with aflatoxin B1. Samples of "Minas" cheeses (fresh, canastra and standard) were collected by the Inspection Service in the Mercado Central in Belo Horizonte city, Minas Gerais - Brazil. A purified extract was obtained by extraction with dichloromethane followed by a washing with n-hexane and immunoaffinity column purification. The quantification of aflatoxin M1 was done by high performance liquid chromatography (HPLC) using a fluorescence detector. Recoveries were about 75%. A serotonergic pathway is apparently involved in parasite-host interactions. Previous studies conducted in our laboratory showed increased rates in oxygen consumption and alterations in body posture in the crab Hemigrapsus crenulatus parasitized by the acanthocephalan, Profilicollis antarcticus. Such changes may be related to the functions described for biogenic amines in crustaceans. During the infective stage the acanthocephalans live freely in the hemocelomic cavity, suggesting that the possible alteration induced by biogenic amines may be related to their neurohormonal function in crustaceans. Análise e identificação das antocianinas de couve rôxa (Brassica spp.) IPB - Escola Superior Agrária

A couve rôxa é uma espécie comestível com uma composição em antocianinas rica e complexa. Sabe-se que contem antocianinas poliglicosiladas, aciladas com um ou mesmo dois grupos acilo aromáticos, nomeadamente com ácidos coumárico e caféico. Este padrão de acilação é particularmente interessante, porque confere estabilidade às antocianinas e aumenta o seu potencial de utilização em aplicações famacêuticas, alimentares e cosméticas. Este estudo tem por objectivo determinar a estrutura das antocianinas de variedades de couve rôxa cultivadas em Portugal, com vista ao seu aproveitamento industrial.

Análise e identificação das antocianins do mirtilo da Madeira (Vaccinium padifolium) e do mirtilo dos Açores (Vaccinium cillyndraceum) IPB - Escola Superior Agrária

Analisaram-se as antocianinas dos mirtilos da Madeira e dos Açores. Os pigmentos foram isolados por cromatografia líquida preparativa bi-dimensional (exclusão molecular vs fase reversa), e identificados por RMN (1H, CAPT, COSY, HSQC, HMBC). Em V. padifolium isolou-se e identificou-se a malvidina 3-rhamnósido, um pigmento muito raro, elevando para 26 o total de antocianinas já descritas nesta espécie (3 tri-glicósidos, 6 di-glicósidos e 17 monoglicósidos, das quais 5 são pigmentos novos. Há ainda alguns pigmentos que não puderam ser identiifcados, ou porque foram isolados em baixíssimas quantidades, ou porque se degradaram. Anthocyanin stability: a chemometrical approach IPB - Escola Superior Agrária

The stability of cyanidin 3-glucoside towards five different factors (pH, anthocyanin concentration, sodium chloride concentration, ascorbic acid concentration, Oxygen) was studied during 60 days storage at room temperature using a 2^4-1 reduced factorial design. The influence of each individual parameter on anthocyanin stability was found to vary as a function of time. Sodium chloride and ascorbic acid were, respectively, the variables most contributing to and against anthocyanin stability. Results suggest there may be some degree of interaction between anthocyanin/ascorbic acid, and between pH/Oxygen, affecting anthocyanin degradation.

Anthocyanins from pomegranate (Punica granatum L.) IPB - Escola Superior Agrária

The anthocyanin contents in the fruits from three cultivars of Punica granatum L. (asseria, mollar and negral) was studied. Six anthocyanins were isolated from the red seeds of pomegranate fruits using a combination of Sephadex LH-20 column chromatography and reverse-phase semi-preparative HPLC. On the basis of chromatographic and spectroscopic techniques, they were identified as the 3-monoglucosides and 3,5-diglucosides of delphinidin, cyanidin and pelargonidin. Availability of analytical standards is a critical aspect in developing methods for quantitative analysis of anthocyanins. Antidiabetic activity of aqueous extract of Sigesbeckia orientalis (St. Paul’s Wort) in alloxan-induced diabetes model BJPS

The current study evaluated antidiabetic and antihyperlipidemic activities of aqueous extract of Sigesbeckia orientalis L. (St. Paul’s Wort) (AESO) in an alloxan-induced diabetic rat model. Following OECD guidelines safe doses of AESO were assessed in rats for the main study. Serum/bood glucose, α-amylase, and lipids levels and histopathological evaluations were conducted to assess antidiabetic and associated antihyperlipidemic efficacies of AESO. AESO was found to be safe up to the dose of 2000 mg/kg. Significant (p < 0.05) reduction in glucose and lipids (total cholesterol, triglycerides, low-density lipoproteins) levels was observed in AESO treatment groups. Antifungal activity of different neem leaf extracts and the nimonol against some important human pathogens BJM

This study was conducted to evaluate the effect of aqueous, ethanolic and ethyl acetate extracts from neem leaves on growth of some human pathogens (Aspergillus flavus, Aspergillus fumigatus, Aspergillus niger, Aspergillus terreus, Candida albicans and Microsporum gypseum) in vitro. Different concentrations (5, 10, 15 and 20%) prepared from these extracts inhibited the growth of the test pathogens and the effect gradually increased with concentration. The 20% ethyl acetate extract gave the strongest inhibition compared with the activity obtained by the same concentration of the other extracts.
