A comparative study of various procedures for tryptophanyl peptide bond cleavage by BNPS-skatole [2-(2-nitrophenyl)-3-methyl-3-bromoindolenine] was carried out on native and on reduced and alkylated bovine β-lactoglobulin (BLG). The reaction yield and the composition of the derived products were studied in acetic acid, trifluoroacetic acid (TFA), and ethanol/TFA. For BNPS-skatole removal, extraction by water or ethyl ether was compared with dialysis and gel filtration. The three expected peptides (1–19, 20–61, 62–162) and incomplete cleaved fragments (1–61, 20–162) were separated and characterized by electrophoresis, reverse-phase high-performance liquid chromatography, and mass spectrometry. La couverture des besoins énergétiques alimentaires a été étudiée chez des poissons tropicaux utilisés en aquaculture et connus pour leur meilleure utilisation de l'énergie non-protéique. Ces besoins ont été évalués, et l'utilisation de l'amidon évaluée comme source potentielle d'énergie alimentaire. Les besoins ont été déterminés en fournissant des rations croissantes d'un aliment unique, riche en protéines, ne pouvant avoir d'effet limitant. L'utilisation de l'amidon a été étudiée par calorimétrie indirecte et tests de croissance. La mise au point de la purification des amylases digestives chez 2 tilapias a permis d'étudier leurs caractéristiques et de les comparer entre elles ou à celles du pancréas de porc. The estimation of bacterial DNA synthesis in trophic studies with [3H] thymidine requires quantitative extraction of labeled DNA. To determine the DNA contribution to total macromolecular labeling in a eutrophic ecosystem, we tested three extraction procedures currently used to estimate DNA labeling from thymidine incorportation : enzymatic fractionation, acid-base hydrolysis, and phenol-chloroform extraction. Because labeled macromolecular fractions are generally defined as DNA, RNA, and proteins, we used incorporation of tritiated thymidine, uridine, and leucine to preferentially label DNA, RNA, and proteins.
