L’écosystème digestif est sous l’influence de facteurs abiotiques et biotiques qui déterminent son équilibre et par conséquent influencent la santé digestive de l'hôte. Chez le lapin en croissance, un déséquilibre de l’écosystème caecal est associé aux entéropathies, responsables de mortalités importantes en élevage. La compréhension des interactions biotope – biocénose digestive permettra la mise en oeuvre de stratégies pour garantir l'équilibre de cet écosystème. Enterococcus faecalis is an important nosocomial pathogen associated with high morbidity and mortality for patients who are immunocompromised or who have severe underlying diseases. The E. faecalis genome encodes numerous surface-exposed proteins that may be involved in virulence. This work describes the characterization of the first internalin-like protein in E. faecalis, ElrA, belonging to the recently identified WxL family of surface proteins. ElrA contains an N-terminal signal peptide for export, a leucine-rich repeat domain that may interact with host cells, and a C-terminal WxL domain that interacts with the peptidoglycan. De nombreuses études menées chez l’homme ont montré que l’obésité est associée à un état inflammatoire chronique caractérisé par une augmentation de la sécrétion de nombreuses molécules dont la leptine et des cytokines inflammatoires comme le TNF-α et l’IL-6 (Clement et al., 2004). PPARalpha transcriptionally induces AhR expression in Caco-2, but represses AhR pro-inflammatory effects Inra

In this work we demonstrate that Caco-2 cell treatment with WY-14643 (a potent PPARα agonist) causes an increase in AhR expression. Luciferase assays and directed mutagenesis experiments showed that induction mainly occurred at transcriptional level and involved a PPRE site located within the AhR promoter. These results were further confirmed by the use of PPARα knockout mice in which AhR induction by WY14643 was abrogated. In addition to CYP1 regulation, AhR has been described as being involved in inflammation, so we also studied the effect of AhR regulation by PPARα on the expression of some inflammation target genes. 3-Methylcholanthrene (a potent AhR agonist) increased the expression (mRNA) of the major inflammatory targets IL-1β and MMP9.
