Resumo: A obtenção de oócitos competentes é um dos fatores que influenciam o sucesso da produção in vitro de embriões em ovinos. Objetivou-se avaliar o efeito de protocolos hormonais na quantidade e qualidade de complexos cumulus-oócitos (CCOs) em ovelhas da raça Santa Inês. Para o número de oócitos GII, houve efeito (P<0,05) da forma de administração, onde aplicações múltiplas resultaram em maior número de oócitos GII do que o tratamento em dose única. Não houve diferença (P>0,05) na taxa de CCOs BCB+ nos diferentes tratamentos: T1(72%), T2 (88%), T3 (80%) ou T4 (76%). ABSTRACT The aim of this work was to evaluate the effect of the Rolipram during the maturation of bovine oocytes and gene expression of embryos produced in vitro. Bovine ovaries were collected in slaughterhouse. The COCs were selected and divided into 5 groups: Control 0 time; Control: IVM for 24 hours; Rolipram treatments with IVM blocking for 24 hours in maturation medium containing (100, 150 and 200µM). After 24 hours all groups were reseated in IVM for another 24 hours. Subsequently COCs were subjected to the same IVM system and fertilized, being checked for cleavage post fertilization and for blastocyst. Effect of Zona Incision by Piezo-Micromanipulator (ZIP) on In Vitro Fertilization in 21 Transgenic Mice Lines OAK

Zona incision using a piezo-micromanipulator (ZIP) has been demonstrated to be effective for in vitro fertilization (IVF) using cryopreserved C57BL/6 spermatozoa. In this study, ZIP oocytes inseminated with frozen-thawed genetically modified C57BL/6J or FVB mice spermatozoa (21 lines) showed fertilization rates of 22-75% and live fetus rates of 8-49%. In 6 of the lines, the fertilization rates for oocytes compared with ZIP (42-75%) were significantly higher than that of nontreated oocytes (0-50%). Using only 90 oocytes for IVF with ZIP, 5 breeding pairs were produced from cryopreserved genetically modified mice spermatozoa. Numerous studies have repeatedly shown that women who smoke experience problems establishing and maintaining pregnancies, and recent work has further demonstrated that the in utero effects of smoke may not be manifested until months or even years after birth. The purpose of this review is to examine the recent literature dealing with the effects of cigarette smoke on the earliest stages of human prenatal development. Studies in this area have included the use of animal models, patients undergoing in vitro fertilization, and embryonic stem cell models. The study was conducted to determine effect of EB or CS supplements in LAMP reaction on greater accuracy of sexing cattle embryos. Bovine ovaries were collected from the slaughterhouse, oocytes were aspirated from follicles then were evaluated morphology. Oocytes were cultured in TCM-199 at 38.5 ºC 24 h. For in vitro fertilization (IVF), oocytes were co-incubated with spermatozoa at 38.5 ºC 24 h. Fertilization rate was evaluated and then the embryos were in vitro cultured until morula stage. Then, embryo samples were frozen and stored at -196 ºC. Frozen embryos (n=35) were randomly extracted for total cellular DNA and amplified by PCR method. DNA samples were stored at -20 ºC.
