Over the last decade, innate immune priming has been evidenced in many invertebrate phyla. If mechanistic models have been proposed, molecular studies aiming to substantiate these models have remained scarce. We reveal here the transcriptional signature associated with immune priming in the oyster Crassostrea gigas. Oysters were fully protected against Ostreid herpesvirus 1 (OsHV-1), a major oyster pathogen, after priming with poly(I·C), which mimics viral double-stranded RNA. Global analysis through RNA sequencing of oyster and viral genes after immune priming and viral infection revealed that poly(I·C) induces a strong antiviral response that impairs OsHV-1 replication. Protection is based on a sustained upregulation of immune genes, notably genes...

An Alternative Inactivant for Rift Valley Fever Virus using Cobra Venom-derived L-Amino Oxidase, which is Related to its Immune Potential BABT
Al-Olayan,Ebtesam M; Mohamed,Aly F; El-Khadraqy,Manal F; Shebl,Rania I; Yehia,HanryM.
ABSTRACT Vaccine improvement depends on the formulation, adjuvant type and inactivant used. The type of formulation may interfere with immunogenicity. The present work aimed to evaluate the inactivation activity and related immune potential of the Cobra venom-derived LAO enzyme compared to the currently used inactivants (BPL and formalin) for both animal and human vaccines. The RVF virus was completely inactivated within 6 hrs, 4 hrs and 2 hrs after treatment with Formalin, LAO and BPL, respectively. The vaccine potency [ED50] was arranged in a descending order from formalin (0.016) to BPL (0.005) and LAO (0.002). The total IgG levels, Neutralizing Index (NI) and Interferon levels were significantly increased compared to those detected after immunization...

Antiviral activity of bovine uterus and placenta induced by Newcastle disease virus Arq. Bras. Med. Vet. Zootec.
Barreto Filho,J.B.; Marques Júnior,A.P.; Golgher,R.R.; Lopes,E.; Carvalho,P.H.A.; Rossi,R.O.D.S..
The antiviral activity profile of the uterus and fetal membranes from bovine placenta, induced by the Newcastle disease virus (NDV) throughout gestation, was investigated. Explants of the endometrium and caruncles were collected from the uterus, and amniochorion, allantochorion and cotyledons, from fetal placenta. Tissue cultures were induced with ~6.0 hemagglutinating units (HU) of NDV. Supernatants were concentrated 20 fold, filtered in 100kDa cut-off membranes and antiviral activity was titrated in MDBK x VSV system. Tissues of the uterus did not exhibit antiviral activity, while allantochorion and amniochorion produced antiviral factors throughout gestation. Antiviral factors were not related with IFN-alpha, gamma, tau or TNF-alpha. The antiviral...

Application of the differential display RT-PCR strategy for the identification of inflammation-related mouse genes BJMBR
Silva,A.M.; Pires,E.G.; Abrantes,E.F.; Ferreira,L.R.P.; Gazzinelli,R.T.; Reis,L.F.L..
The inflammatory response elicited by various stimuli such as microbial products or cytokines is determined by differences in the pattern of cellular gene expression. We have used the differential display RT-PCR (DDRT-PCR) strategy to identify mRNAs that are differentially expressed in various murine cell types stimulated with pro-inflammatory cytokines, microbial products or anti-inflammatory drugs. Mouse embryonic fibroblasts (MEFs) were treated with IFNs, TNF, or sodium salicylate. Also, peritoneal macrophages from C3H/Hej mice were stimulated with T. cruzi-derived GPI-mucin and/or IFN-<FONT FACE="Symbol">g</FONT>. After DDRT-PCR, various cDNA fragments that were differentially represented on the sequencing gel were recovered, cloned and...

Distribution of Ifitm3 in Yellow-Feathered Broilers and Inhibition of Avian Reovirus Multiplication by Ifitm3 Rev. Bras. Ciênc. Avic.
Wang,Q; Yuan,X; Li,C; Deng,H; Wang,C.
ABSTRACT This study was carried to express the interferon-induced transmembrane protein 3 (IFITM3) in vitro and examine its function in inhibition of avian reovirus (ARV) replication. The recombinant prokaryotic vector expressing yellow-feathered broiler IFITM3 was successfully constructed, and the recombinant protein was expressed in competent Escherichia coli BL21 cells. New Zealand white rabbits were immunized with the purified recombinant protein to prepare a polyclonal antibody, with a titer of 1:128,000. Immunohistochemistry, reverse transcription-PCR, and real-time fluorescence quantitative PCR showed that IFITM3 was distributed in the yellow-feathered broiler immune organs, and the expression of IFITM3 in bursa of Fabricius was more than in spleen...

Evaluation of prognostic value of cell adhesion molecules in chronic hepatitis C therapy J. Venom. Anim. Toxins incl. Trop. Dis.
Abdallah,N; Morsi,M; Hamed,N; Abdel Aziz,H.
The most reliable determination of severity and prognosis in chronic viral hepatitis is the histological staging of the disease, which comprises an invasive procedure and is often not well accepted by patients. The search for alternative non-invasive methods is mandatory especially in follow-ups after initial assessment by biopsy. The aim of this study was to evaluate the role of vascular cell adhesion molecule-1 (VCAM-1) and intercellular adhesion molecule-1 (ICAM-1) in patients under interferon alpha therapy whether responsive or non-responsive to therapy. Thirty chronic hepatitis C patients (CHC) under combined therapy of interferon-α with ribavirin, whether responsive or non-responsive, were included in the study as well as ten healthy controls. Serum...

Hepatitis C treatment: shorter and better? BJID
Araújo,Evaldo Stanislau Affonso de; Courtouké,Cláudia; Barone,Antonio Alci.
Herein, we present a synthesis of two publications that evaluate an abbreviated therapeutic approach to treating chronic hepatitis C virus (HCV) infection. Based on those publications, we discuss the use of the early virologic response (EVR) as a tool for the optimized management of patients under treatment, as well as reviewing concepts of HCV viral kinetics. The fourth-week EVR, characterized by HCV RNA dropping to undetectable levels, allows individuals infected with HCV genotype 1 and presenting low baseline viral loads to be treated with the combination of pegylated interferon and ribavirin for 24 weeks, whereas individuals infected with HCV genotypes 2 or 3 can be treated for only 12 weeks. Therefore, by adopting abbreviated treatment regimens...

Hepatitis C virus infection, cryoglobulinemia, and peripheral neuropathy: a case report BJMBR
Vigani,A.G.; Macedo-de-Oliveira,A.; Pavan,M.H.P.; Pedro,M.N.; Gonçales Jr.,F.L..
Hepatitis C virus (HCV) is essentially hepatotropic but its manifestations can extend beyond the liver. It can be associated with autoimmune diseases, such as mixed cryoglobulinemia, membranoproliferative glomerulonephritis, autoimmune thyroiditis, and lymphoproliferative disorders. The mechanisms that trigger these manifestations are not completely understood. We describe a 48-year-old man with chronic HCV infection (circulating HCV RNA and moderate hepatitis as indicated by liver biopsy), cryoglobulinemia, and sensory and motor peripheral neuropathy. The diagnosis of multineuropathy was confirmed by clinical examination and electromyographic tests. A nerve biopsy revealed an inflammatory infiltrate in the perineurial space and signs of demyelination and...

Increased levels of IL-10, IL-12, and IFN- in patients with visceral leishmaniasis BJID
Khoshdel,A.; Alborzi,A.; Rosouli,M.; Taheri,E.; Kiany,S.; Javadian,M.H..
Visceral leishmaniasis remains a serious public health problem in developing countries. Cytokines have a crucial role in the pathogenesis of this disease. We evaluated plasma levels of IL-10, IL-12 and IFN- in 32 patients with active visceral lieshmaniasis, in 29 siblings of the patients and in 23 normal individuals in an endemic area to look for correlations between the clinical outcome of infection and the plasma cytokine levels. An analysis was made with a skin test and a quantitative sandwich enzyme immunoassay. Data were analyzed with the Mann Whitney test and the Kruskal Wallis test. The cytokine levels were significantly higher in the patients than in the control groups. We concluded that normalization of the plasma level of IFN- can serve as a...

Interferon therapy shifts natural killer subsets among egyptian patients with chronic hepatitis C BJID
Fathy,Amal; Eldin,Mohamed Mohy; Metwally,Lobna; Eida,Mohamed; Abdel-Rehim,Marwa; Esmat,Gamal.
Natural killer cells can be divided into five subpopulations based on the relative expression of CD16 and CD56 markers. The majority of natural killer cells are CD56dim, which are considered to be the main cytotoxic effectors. A minority of the natural killer cells are CD56bright, and function as an important source of immune-regulatory cytokines. Shifts of these subsets have been reported in patients with chronic hepatitis C virus infection. We sought to investigate the shift of natural killer subsets among Egyptian patients with chronic HCV and to analyze the influence of interferon therapy on this shift. We applied a flow cytometric analysis of peripheral blood natural killer subsets for 12 interferon-untreated and 12 interferon-treated patients with...

Relationship of cytokines and cytokine signaling to immunodeficiency disorders in the mouse BJMBR
Morawetz,R.A.; Giese,N.A.; Gabriele,L.; Rothman,P.; Horak,I.; Ozato,K.; Morse III,H.C..
The contributions of cytokines to the development and progression of disease in a mouse model of retrovirus-induced immunodeficiency (MAIDS) are controversial. Some studies have indicated an etiologic role for type 2 cytokines, while others have emphasized the importance of type 1 cytokines. We have used mice deficient in expression of IL-4, IL-10, IL-4 and IL-10, IFN-<FONT FACE="Symbol">g</font>, or ICSBP - a transcriptional protein involved in IFN signaling - to examine their contributions to this disorder. Our results demonstrate that expression of type 2 cytokines is an epiphenomenon of infection and that IFN-<FONT FACE="Symbol">g</font> is a driving force in disease progression. In addition, exogenously administered IL-12...

Therapy with interferon-alpha plus ribavirin for membranoproliferative glomerulonephritis induced by hepatitis C virus BJID
Lopes,Edmundo P.A.; Valente,Lucila M.; Silva,A. Eduardo B.; Kirsztajn,Gianna Mastroianni; Cruz,Cibele N.; Ferraz,Maria Lucia G.
We describe a treatment made with interferon-alpha (IFN-alpha) plus ribavirin of two patients with membranoproliferative glomerulonephritis (MPGN) induced by hepatitis C virus (HCV): Case 1 was a 22-year-old woman with leg and facial edema, hypertension and proteinuria, whose liver biopsy revealed chronic active hepatitis; and Case 2 was a 42-year-old man with anasarca, hypertension and proteinuria, whose liver biopsy indicated cirrhosis. Both had anti-HCV, HCV-RNA and cryoglobulins. IFN-alpha (3 million units (MU), 3 times/week) and ribavirin (1 g/day) were administered for 12 months. The drugs were well tolerated by both patients. Serum alanine aminotransferase (ALT) levels normalized and HCV-RNA became negative. Cryoglobulins disappeared and an...

Transcription of interferon stimulated genes in response to porcine rubulavirus infection in vitro BJM
Flores-Ocelotl,María del Rosario; Rosas-Murrieta,Nora Hilda; Vallejo-Ruiz,Verónica; Reyes-Leyva,Julio; Herrera-Camacho,Irma; Santos-López,Gerardo.
Porcine rubulavirus (PoRV) is an emerging virus causing meningo-encephalitis and reproductive failures in pigs. Little is known about the pathogenesis and immune evasion of this virus; therefore research on the mechanisms underlying tissue damage during infection is essential. To explore these mechanisms, the effect of PoRV on the transcription of interferon (IFN) pathway members was analyzed in vitro by semi-quantitative RT-PCR. Ten TCID50 of PoRV stimulated transcription of IFNα, IFNβ, STAT1, STAT2, p48 and OAS genes in neuroblastoma cells, whereas infection with 100 TCID50 did not stimulate transcription levels more than non-infected cells. When the cells were primed with IFNα, infection with 1 TCDI50 of PoRV sufficed to stimulate the transcription of...
