Characterization of variants of Bradyrhizobium elkanii and B. japonicum and symbiotic behaviour in soybeans Ciência Rural
Kober,Marcia de Vargas; Sá,Enilson Luiz Saccol de; Freire,João Ruy Jardim; Giongo,Adriana.
Variation in rhizobia strains isn’t a desirable fact based mainly on the possibility of unexpected results on legume inoculation. In this work, we studied the variability on phenotypic characteristics and genetic stability of rhizobia strains recommended for soybean inoculation. Variants with stable colony morphology were obtained from Bradyrhizobium japonicum strain SEMIA 5080 and from B. elkanii SEMIA 5019. Variants from SEMIA 587 obtained by another author were also used. The variants differed on colony characteristics, nodulation capacity, nitrogen fixation efficiency and competitive ability for nodule formation in two soybean varieties (Jacui 7 and IAS 5). Symbiotic behavior varied according to plant variety. Only the variants 5019 G and 5019 P...

Comparative electrophoretic profile of proteins and esterases in healthy silkworm larvae (Bombyx mori Lineu, 1758) and infected with nucleopolyhedrovirus - doi: 10.4025/actascibiolsci.v35i1.8612 Biological Sciences
Ruiz, Juliana Bueno; Universidade Estadual de Maringá; Ronqui, Ludimilla; Universidade Federal de Rondonia; Oliveira, Jussara Ricardo de; Universidade Paranaense; Ruvolo-Takasusuki, Maria Claudia Colla; Universidade Estadual de Maringá.
Total proteins and esterases from silk gland extracts of Bombyx mori silkworm were characterized and compared with electrophoretic profiles of prepared extracts with silkworm glands infected with nucleopolyhedrovirus (BmNPV). SDS-PAGE (7%) gels was used for the total proteins, and 10% native PAGE for esterases. In the silk glands extracts of healthy silkworms, it was observed seven protein zones with molecular weight varying between 10 kDa (P1) and 60 kDa (P7). In the infected silkworms, a new zone named P8 (90 kDa) was also detected. Esterases activity at 5th instar larvae underwent changes after the infection with BmNPV, since there was a reduction (EST-6 and EST-7) and an increase (EST-8) in the intensity of the regions of esterases activity, and...

Cryopreservation in Coffea canephora Pierre seeds: Slow and fast cooling Ciência e Agrotecnologia
Coelho,Stefânia Vilas Boas; Rosa,Sttela Dellyzete Veiga Franco da; Fantazzini,Tatiana Botelho; Baute,Júlia Lima; Silva,Luciano Coutinho.
ABSTRACT Coffee is one of the main agricultural commodities in the country, and it is important to conservation of plant material for breeding programs. Cryopreservation is a promising alternative for preserving in the long-term the germplasm of species considered recalcitrant. However, studies should be performed to achieve maximum survival of seedlings after immersion in liquid nitrogen. The objective of this work was to find a cryopreservation protocol for storing seeds of Coffea canephora, studying two methods of cryopreservation, slow and fast cooling. Seeds were subjected to drying in silica gel up to the water content of 0.25 g g-1. In the first experiment, dried seeds were subjected to treatments of slow cooling at speeds of -1 ºC min-1,-3 ºC min-1...

Genetic relationships among American species of Prosopis (Leguminosae) based on enzyme markers Genet. Mol. Biol.
Bessega,Cecilia; Saidman,Beatriz Ofelia; Vilardi,Juan César.
In the present work, isoenzyme electrophoresis was used to analyze the variability and phenetic relationships among seven American species of genus Prosopis belonging to three different sections: P. argentina (Monilicarpa), P. glandulosa, P. velutina, P. flexuosa, P. ruscifolia, P. kuntzei (Algarobia), and P. reptans (Strombocarpa). The genetic variability in P. argentina, P. reptans, and P. kuntzei was significantly lower than in the rest of the species analyzed. The species belonging to different sections are highly differentiated, but the relationships retrieved among species belonging to the section Algarobia suggested that the series of this section are not natural groups. P. kuntzei is as differentiated from the remaining species of Algarobia as from...

Identification of Leishmania (Leishmania) chagasi isolated from healthy skin of symptomatic and asymptomatic dogs seropositive for leishmaniasis in the Municipality of Rio de Janeiro, Brazil BJID
Madeira,Maria de Fátima; Schubach,Armando de Oliveira; Schubach,Tânia Maria Pacheco; Leal,Cristianni Antunes; Marzochi,Mauro Célio de Almeida.
Euthanasia of seropositive dogs has been one of the principal measures adopted by the Program for the Control of Leishmaniasis in Brazil for many years. However, its efficacy is currently being questioned. We obtained intact skin samples from 20 Leishmania-reactive dogs from the municipality of Rio de Janeiro that had been referred for euthanasia. The promastigote forms of Leishmania were isolated in culture from 18 of these animals. Fourteen of these isolates were identified as Leishmania (Leishmania) chagasi by isoenzyme electrophoresis; seven of these were from asymptomatic dogs and seven were from symptomatic animals with visceral leishmaniasis. In conclusion, cutaneous parasitism is found in the intact skin of dogs naturally infected with L. (L.)...

Ulisses,Cláudia; Melo-de-Pinna,Gladys Flávia; Willadino,Lilia; Albuquerque,Cynthia Cavalcanti de; Camara,Terezinha Rangel.
The internal morphology of embryos from immature and mature fruits of Heliconia bihai (L.) L. cv. Lobster Claw Two was examined. Embryos were inoculated into MS media (full MS and ½ MS) and GA3 (0, 2.5 and 5 mg L-1) with either sucrose or glucose. These plantlets were then replicated and transferred to MS medium (full MS or ½ MS) with 0 or 2.5 mg L-1 BAP and their multiplication was evaluated 30 and 45 days after inoculation. The genetic variability of the multiplied plants was estimated using isoenzyme analyses. The internal morphology of the mature embryos revealed their tissues to be in more advanced stages of differentiation than immature embryos. In the conversion phase, 85% of the inoculated embryos developed into plants in the ½ MS medium with...

Isoenzymatic polymorphisms in urban populations of Drosophila willistoni RChHN
SAAVEDRA,CARMEN C. R.; NAPP,MARLY; REGULY,MARIA LUIZA; VALENTE,VERA LUCIA S.
Natural populations of Drosophila willistoni collected both in urban sites and in the wild, as control, were analysed with respect to their isoenzyme polymorphisms in six loci of enzyme systems, through horizontal electrophoresis. The first pool of populations was collected and analysed during the years 1987 and 1988, and the other during 1992 and 1993, in comparable seasons. The level of enzymatic polymorphism first detected was smaller in comparison to that observed four years later, suggesting an increase of variability along the time elapsed since the first sampling of this species, in the city of Porto Alegre. Our findings are suggestive of an increasing adjustment of the species to this new environment

Molecular characterisation of radish cultivars Rev. Ciênc. Agron.
Cruz,Sara Michelly; Nery,Marcela Carlota; Von Pinho,Édila Vilela de Resende; Laia,Marcelo Luiz de.
The radish has been gaining importance in the Brazilian market due to its use as a green manure and cover crop, and also due to its potential for the production of biodiesel, however there are only two registered cultivars, which are morphologically very similar. Determination of genetic purity is a prerequisite in marketing seeds as it ensures uniformity of the crop and is important in breeding programs. The aim of this work therefore was to evaluate the similarity and genotype patterns which permit differentiation of the commercial radish cultivars IPR 116 and CATI AL-1000. In order to do this, isoenzyme electrophoretic patterns were analysed in dry seeds, soaked seeds, seedling leaves and young leaves; the isoenzymes used being: superoxide dismutase,...

Molecular weight estimation of esterase isoenzymes in closely related Drosophila Species (Diptera: Drosophilidae) in non-denaturing polyacrylamide gel electrophoresis BABT
Mateus,Rogério Pincela; Cabral,Hamilton; Bonilla-Rodriguez,Gustavo Orlando; Ceron,Carlos Roberto.
A method that allows the measure of molecular weight of two well-known and closely related esterases from Drosophila mojavensis and its sibling species, D. arizonae, is here described, using native polyacrylamide gel electrophoresis at several concentrations, applying Fergunson´s principles. These enzymes, namely EST-4 and EST-5, presented molecular weight values between 81 and 91 kDa. In spite of their distinct expression pattern through the insect's life cycle, they showed properties of isoenzymes codified by distinct structural genes, supporting the hypothesis of a rather recent gene duplication event that generated both in D. mojavensis and D. arizonae, as well as in other species of repleta group. The method is simple and adequate to be applied to...

On the origin and diversification of Venezuelan freshwater fishes: the genus Gephyrocharax (Ostariophysi: Characidae) a case study Neotropical Ichthyology
Bonilla-Rivero,Ana; López-Rojas,Héctor.
We conducted a phylogeographic analysis of the genus Gephyrocharaxin Venezuela to evaluate geomorphologic evidence for the formation of the country's main watersheds and to establish a biogeographical hypothesis of possible diversification mechanisms of the Neotropical freshwater fish fauna. We assayed eight enzyme systems and general proteins to estimate genetic variability (H, P), intraspecific structuring in several Gephyrocharax valencia and G. venezuelae populations (FIS, FIT, and FST), and a phylogenetic approach for the three species of Gephyrocharax in Venezuela, using Corynopoma riisei as the external group. Fourteen presumptive lociindicate that populations of the three species of Gephyrocharaxanalyzed show a clear genetic inter-specific...

Panstrongylus megistus is an important Chagas Disease vector and is said to be one of the species that might replace Triatoma infestans as the main vector of that disease in Brazil. The different degrees of P. megistus domiciliation in Brazil and its epidemiological relevance draw forth the need for the development of genetic studies that make it possible to analyze and understand the interchange of individual and gene fluxes among different populations. Thus, the present work aimed at studying the genetic variability of P. megistus in the State of Paraná - south of Brazil- and at comparing it with populations of the same species from five other states in Brazil (SP, MG, SC, RS, SE). In order to attain the proposed objective, 25 populations were studied...

Physiological quality of coffee seeds produced under different levels of solar radiation and maturation stages Rev. bras. sementes
Baliza,Danielle Pereira; Caixeta,Franciele; Pinho,Édila Vilela de Resende Von; Cunha,Rodrigo Luz da; Martins,Denize Carvalho; Rosa,Sttela Dellyzete Veiga Franco da.
In Brazil, although the coffee plantations are predominantly grown under full sunlight, the use of agroforestry systems can lead to socioeconomic advantages, thus providing a favorable environment to the crop by promoting its sustainability as well as environmental preservation. However, there is a lack of information on physiological quality of the coffee seeds produced under different levels of solar radiation. Within this context, the objective of this study was to evaluate the influence of different levels of solar radiation and maturation stages on the physiological quality of coffee (Coffea arabica L.) seeds, cv. Acaiá Cerrado MG-1474. Three levels of solar radiation (plants grown under full sunlight; under plastic screens of 35% shading; and under...

Structural and regulatory differences in amylase isoenzymes in germinating Brazilian barley cultivars Genet. Mol. Biol.
Georg-Kraemer,J.E.; Cavalli,S.S..
The amylase electrophoretic patterns of 10 Brazilian brewing-barley varieties with different malting grades and diastatic power were analyzed during the 7-day germination which occurs during the malting process. Intra and inter-variety genetic variability was observed at both the structural and regulatory level. In the first few days after germination all varieties showed a few active loci, all of them with low activity. In subsequent days, new loci became active and those already detected since early germination showed increased activity. All varieties showed a continuous increase in amylase synthesis until the 3rd and/or 4th day after germination. Some varieties maintained high amylase activity until the last day of germination, while others showed a...
