Anti-leptospirosis agglutinins in Brazilian capybaras (hydrochoerus hydrochaeris) J. Venom. Anim. Toxins incl. Trop. Dis.
Langoni,Helio; Kuribara,Ivone Yumi; Correa,Ana Paula Ferreira Lopes; Ullmann,Leila Sabrina; Sánchez,Gabriela Pacheco; Lucheis,Simone Baldini.
Abstract Background The interest in commercial use of wild animals is increasing, especially regarding raising of capybaras. Although this wild species is potentially lucrative for the production of meat, oil and leather, it is suggested as a probable reservoir of leptospires. Methods Due to the economic importance of this species and the lack of studies concerning leptospirosis, the presence of anti-leptospirosis agglutinins was assayed in 55 serum samples of capybaras (Hydrochoerus hydrochaeris) from commercial and experimental breeding flocks located in São Paulo state, Paraná state, and Rio Grande do Sul state, Brazil. Samples were obtained through cephalic or femoral venipunction (5 to 10 mL). Microscopic agglutination test was used according to...

Bilateral facial palsy associated with leptospirosis BJID
Silva,Andressa Alves da; Ducroquet,Marcelo; Pedrozo Junior,Jeff Chandler.
Leptospirosis is a zoonosis of worldwide occurrence caused by the spirochete Leptospira interrogans. It is an acute feverish disease with a broad clinical spectrum and follows a characteristic biphasic course. Bilateral facial palsy is a rare clinical condition and the differential diagnosis of its causes is extensive. The objective of this exploratory study, presented as a case report, is to describe the occurrence of bilateral facial palsy as an unusual manifestation of leptospirosis. This suggestion should not be overlooked when analyzing the causes for bilateral facial palsy, and should be considered with other possible differential diagnoses, some of which are potentially fatal.

Characterization of a virulent Leptospira interrogans strain isolated from an abandoned swimming pool BJM
Forster,Karine M.; Hartwig,Daiane D.; Seixas,Fabiana K.; McBride,Alan J.A.; Monte,Leonardo G.; Recuero,Ana Lúcia C.; Brod,Claudiomar S.; Hartleben,Cláudia P.; Amaral,Marta; Dellagostin,Odir A..
Pathogenic Leptospira spp. are the etiological agents of leptospirosis, an important disease of both humans and animals. In urban settings, L. interrogans serovars are the predominant cause of disease in humans. The purpose of this study was to characterize a novel Leptospira isolate recovered from an abandoned swimming pool. Molecular characterization through sequencing of the rpoB gene revealed 100% identity with L. interrogans and variable-number tandem-repeat (VNTR) analysis resulted in a banding pattern identical to L. interrogans serogroup Icterohaemorrhagiae, serovar Copenhageni or Icterohaemorrhagiae. The virulence of the strain was determined in a hamster model of lethal leptospirosis. The lethal dose 50% (LD50) was calculated to be two...

Cytokine and antibody production during murine leptospirosis J. Venom. Anim. Toxins incl. Trop. Dis.
Marinho,M.; Silva,C.; Lima,V. M. F.; Peiró,J. R.; Perri,S. H. V..
The aim of the present study was to investigate the kinetics of humoral and cellular responses during leptospirosis. We observed that the presence of tumor necrosis factor-alpha (TNF-alpha) and interleukin-6 (IL-6) was associated with antibody production and bacterial recovery, and the compromising of both TNF-alpha and IL-6 in the immunopathogenesis of leptospirosis during an experimental infection of BALB/c mice inoculated with Leptospira interrogans serovar Canicola was verified. Results showed higher levels of TNF-alpha and IL-6 in the initial phase of infection, in which the greatest bacterial clearance was observed. However, when the bacterial recovery was compared with the kinetics of the production of antibodies, the results revealed a kinetics...

HEINEMANN,Marcos Bryan; GARCIA,José Fernando; NUNES,Cáris Maroni; HIGA,Zenaide Maria Moraes; VASCONCELLOS,Silvio Arruda; Richtzenhain,Leonardo José.
Due to the high importance of the venereal transmission of bovine leptospirosis, this study aimed to test the ability of PCR to detect Leptospira interrogans serovar hardjo DNA in experimentally contaminated bovine semen. Employing primers directed to the 16S rRNA gene, 10 bacteria/ml of semen could be detected by PCR. Results achieved in this work show that PCR can have a great potential to detect Leptospira spp. in insemination centers.

Miotto,Bruno Alonso; Hora,Aline Santana da; Taniwaki,Sueli Akemi; Brandão,Paulo Eduardo; Heinemann,Marcos Bryan; Hagiwara,Mitika Kuribayashi.
Abstract A modified TaqMan real-time polymerase chain reaction targeting a 138 bp fragment within the lipl32 gene was developed to identify exclusively pathogenic Leptospira spp. in dog urine samples. Thirty-five samples from dogs with suspected clinical leptospirosis and 116 samples from apparently healthy dogs were tested for presence of leptospiral DNA using the TaqMan-based assay. The results were compared with those from a well-established conventional PCR targeting the 16S RNA encoding gene associated with nucleotide sequencing analysis. The overall agreement between the assays was 94.8% (confidence interval [CI] 95% 88-100%). The newly developed assay presented 91.6% (CI 95% 71.5-98.5%) relative sensitivity (22[+] lipl32 PCR/24[+] 16S RNA and...

ECKSTEIN, C.; LOPES, L. B.; MOUSTACAS, V. S.; RODRIGUES, R. O.; CASTRO, B. G. de; SANTOS, R. L..
Leptospirosis has a worldwide distribution with high social and economic impact. It is caused by a variety of Leptospira spp. serovars, particularly in tropical regions. Various species of animals can be affected by Leptospira spp. including cattle, horses, sheep, goats and swine, and can act as a reservoir for human infection. In Brazil, sheep leptospi¬rosis has been diagnosed in a variety of States, however, little is known about sheep flocks of Mato Grosso. Considering the marked growth of sheep industry in Mato Grosso State in past recent years, this study aimed to evaluate the frequency of serologically positive sheep for Leptospira spp. serovars in the State of Mato Grosso (Brazil).

Abstract In swine and bovines, leptospirosis prevention and control is carried out via vaccination of susceptible animals using bacterins. However, the efficiency of leptospirosis vaccines has been questioned. This work aimed to investigate the potency of five leptospirosis vaccines sold commercially in Brazil, challenging the animals with one autochthonous strain of Leptospira, Canicola serovar, denoted LO4, isolated from swine. The standard protocol was followed, and renal carriers of Leptospira were identified among the surviving animals by culture and PCR. Of the five vaccines tested, only two proved effective. None of the surviving animals was positive by culture; however, one animal was positive by PCR. Three of the five vaccines sold commercially in...
