Association of BoLA-DRB3 genotype with somatic cell count in milk of Polish Holstein cattle R. Bras. Zootec.
Oprzadek,Jolanta M.; Brzozowska,Anna M.; Urtnowski,Piotr; Rutkowska,Karolina; Lukaszewicz,Marek.
ABSTRACT The objective of this study was to evaluate the suitability of the BoLA-DRB3 gene polymorphism to define the phenotypic value of somatic cell count of 808 Polish Holstein cows. The cows were fathered by 190 sires. The PCR-RFLP alleles were identified with the BstYI, HaeIII, and RsaI restriction enzymes. For statistical analysis, 17 alleles whose frequencies in the herds were equal to or higher than 2% were selected. Somatic cell count was analyzed based either on the lactation averages or on test-day yields. Significant relationship between the occurrence of the BoLA-DRB3 gene alleles and somatic cell count were stated. In case of estimating the effects for all lactations together, the effect of substitution was observed for 11 alleles. The...

Current Contribution of Four Biotechnologies to New Zealand's Primary Sector AgEcon
Kaye-Blake, William; Saunders, Caroline M.; Emanuelsson, Martin.
Over the last several decades, changes to production technologies have significantly increased output from the primary sector. The contribution of improved genetics has been variously estimated, and the impacts of genetic engineering have received much attention. The present research estimated the economic impact of four modern biotechnologies (other than genetic engineering) on the primary sector. Information from interviews with key informants was incorporated into a cost-benefit analysis to estimate these impacts. The net contribution was estimated at $266 million per year, with impacts spread unevenly through the sector and across biotechnologies.

Development of two sequence-specific PCR markers linked to the le gene that reduces pod shattering in narrow-leafed Lupin (Lupinus angustifolius L.) Genet. Mol. Biol.
Boersma,Jeffrey G.; Buirchell,Bevan J.; Sivasithamparam,Krishnapillai; Yang,Huaan.
Wild types of narrow-leaf lupin (Lupinus angustifolius L.) have seed pods that shatter upon maturity, leading to the loss of their seeds before or during the harvest process. Two recessive genes have been incorporated into domesticated cultivars of this species to maximize harvest-ability of the produce. One of these genes is called lentus (le). Two microsatellite - anchored fragment length polymorphism (MFLP) candidate markers were identified as closely linked to the le gene in a recombinant inbred line (RIL) population derived from a domesticated x wild type cross. The candidate MFLP markers were isolated from the gel, re-amplified by PCR, cloned and sequenced. The MFLP polymorphisms were converted into sequence-specific PCR-based markers. Linkage...

Identification of microsatellites linked to Lr47 Electron. J. Biotechnol.
Vanzetti,Leonardo Sebastián; Brevis,Juan Carlos; Dubcovsky,Jorge; Helguera,Marcelo.
Leaf rust resistance gene Lr47 is located within a interstitial segment of Triticum speltoides Taush. 7S chromosome translocated to the short arm of chromosome 7A of bread wheat. This gene is resistant against currently predominant races of leaf rust from Argentina. The objectives of this study were to identify microsatellites linked to this source of resistance as a potential tool to introgress this source of resistance. Isogenic lines with and without Lr47 developed from 10 cultivars/breeding lines as well as 10 microsatellites previously mapped in 7AS chromosome were used in this study. Microsatellite gwm 60 was the only marker that co-segregated completely linked to Lr47. These data indicate that gwm 60 could be a valuable marker to introgress Lr47 in...

Identification of molecular markers on bovine chromosome 18 associated to calving interval in a Brangus-Ibagé cattle herd Ciência Rural
Weimer,Tania de Azevedo; Steigleder,Clara Sabina; Machado,Márcia Silveira; Almeida,Sabrina Esteves Matos; Oliveira,João Francisco Coelho; Moraes,José Carlos Ferrugem; Henkes,Luis Ernani.
In the detection phase of a bovine marker assisted selection program, this paper investigated the genetic variability of three microsatellites on the chromosome 18 (BTA 18). The possible associations between genotypes or alleles of these markers versus weight at first calving and a lifetime calving interval (as indicators of reproductive performance) were evaluated in a beef cattle herd (5/8 Aberdeen Angus x 3/8 Nelore). Eleven alleles were detected in TGLA227 and ILSTS002 and three in BMS3004, the most frequent being TGLA227*79, ILSTS002*133, ILSTS002*135 and BMS3004*129. Polymorphic information content ranged from 0.41 to 0.84, while heterozygosity ranged from 49% to 86%, with an average value of 77%. The association analyses performed between genotype...

Molecular identification and mapping of a novel stripe rust resistance gene in wheat resistance line CH5389 Ciência Rural
Zhan,Haixian; Guo,Huijuan; Qiao,Linyi; Mao,Liping; Zhang,Shuosheng.
ABSTRACT: Stripe rust, caused by Puccinia striiformis is one of the most destructive diseases of wheat worldwide. CH5389 is a wheat-Thinopyrum intermedium derived line conferring stripe rust resistance. Genetic analyses of seedlings of F2 populations and F2:3 families developed by crossing CH5389 and susceptible common wheat revealed that stripe rust resistance in CH5389 was controlled by a single dominant gene that was designated YrCH5389. Eight SSR and EST-PCR polymorphic markers on chromosome 3AL were identified in F2 population of CH5389/Taichung29. The YrCH5389 was flanked by EST marker BE405348 and SSR marker Xwmc388 on chromosome 3AL with genetic distances of 2.2 and 4.6 cM, respectively. Comparative genomic analysis demonstrated that the...

Occurrence of the rust resistance gene Lr37 from Aegilops ventricosa in Argentine cultivars of wheat Electron. J. Biotechnol.
Bulos,Mariano; Echarte,Mariel; Sala,Carlos.
Leaf rust of wheat (Triticum aestivum L.) caused by the fungus Puccinia triticina (formerly P. recondita f. sp. tritici), is one of the most important foliar diseases of this crop. Lr37 rust resistance gene, which confers resistance in wheat against leaf rust, was introgressed into cultivated wheat from Aegilops ventricosa Tausch. Rust races with virulence to Lr37 have been identified in different countries, but it still provides resistance to a wide range of races and is useful in combination with other resistance genes. There are no reports about the presence, frequency and origin of Lr37 in Argentinean wheat cultivars. In this work, we analyzed 88 registered Argentinean wheat cultivars developed by different breeding companies and institutions during...

QTLs for seedling traits under salinity stress in hexaploid wheat Ciência Rural
Ren,Yongzhe; Xu,Yanhua; Teng,Wan; Li,Bin; Lin,Tongbao.
ABSTRACT: Soil salinity limits agricultural production and is a major obstacle for increasing crop yield. Common wheat is one of the most important crops with allohexaploid characteristic and a highly complex genome. QTL mapping is a useful way to identify genes for quantitative traits such as salinity tolerance in hexaploid wheat. In the present study, a hydroponic trial was carried out to identify quantitative trait loci (QTLs) associated with salinity tolerance of wheat under 150mM NaCl concentration using a recombinant inbred line population (Xiaoyan 54×Jing 411). Values of wheat seedling traits including maximum root length (MRL), root dry weight (RDW), shoot dry weight (SDW), total dry weight (TDW) and the ratio of TDW of wheat plants between salt...

Bahng Taen is a non-glutinous, non-photoperiod sensitive, short duration and high yielding rice variety. This variety showed resistance against brown planthopper (BPH) in the Eastern region, but the resistance has decreased drastically. Therefore, marker-assisted backcrossing was applied to improve BPH resistance by introgression of a Bph3 resistant gene from PTB33 and Rathu Heenati into Bahng Taen. Two SSR markers, RM 170 for Bahng Taen with PTB33 crossing and RM 190 for Bahng Taen with Rathu Heenati crossing, were used in combination with plant type selection to obtain the target plants with Bph3 gene. Twenty six and thirty heterozygous BC4F1 plants were developed from the cross of Bahng Taen with PTB33 and Bahng Taen with Rathu Heenati, respectively....

การพัฒนาพันธุ์ข้าวปทุมธานี 1 ให้ต้านทานเพลี้ยกระโดดสีน้ำตาล โดยใช้โมเลกุลเครื่องหมาย Thai Agricultural
Jate Kotcharerk; Phamorn Pattavatang; Suradet Palawisut; Jirapong Jairin.
Main purpose of this research was to increase the ability of PathumThani 1 rice to resist brown planthopper. The major tool used to accomplish this was marker assisted selection technology targeting on the gene Bph 3. The target gene harbored in the donor; UBN03078-101-342-4-19 (KDML105 x RathuHeenatiintrogression lines), should be transferred into the new generations of rice breeding program performed in this research. We aimed to develop the PathumThani 1 which still maintains its eating and cooking properties but hopefully increases BPH resistance. F1 generations of the crossing between PathumThani 1 and the donor were selected by targeting theRM 586, RM8072 andRM8074molecular markerof the Bph3 then back-crossed to generate BC1F1-BC4F2. Selection of...
