A conservative region of the mercuric reductase gene (merA) as a molecular marker of bacterial mercury resistance BJM

The most common bacterial mercury resistance mechanism is based on the reduction of Hg(II) to Hg0, which is dependent of the mercuric reductase enzyme (MerA) activity. The use of a 431 bp fragment of a conservative region of the mercuric reductase (merA) gene was applied as a molecular marker of this mechanism, allowing the identification of mercury resistant bacterial strains.

The decapod Grapsus grapsus is commonly found on oceanic islands of the Pacific and Atlantic coasts of the Americas. In this study, a simple, quick and reliable method for detecting its larvae in plankton samples is described, which makes it ideal for large-scale studies of larval dispersal patterns in the species.

Ochratoxin A (OTA) is one of the most abundant food-contaminating mycotoxins. Its occurrence in several agricultural commodities has been considered a worldwide problem. This toxin is mainly produced by Aspergillus species. OTA has nephrotoxic, immunosuppressive, and carcinogenic effects and consequently the contamination with this toxin represents a high risk for human and animal health. Assessment of genetic diversity in the Russian olive (Elaeagnus angustifolia) based on ISSR genetic markers Rev. Ciênc. Agron.

Elaeagnus is a Eurasian tree with 77 species worldwide. In this study, ISSR markers were used to establish the level of genetic relationships and polymorphism across nine genotypes of Elaeagnus angustifolia collected from 9 different regions of West Azarbaijan province. The ISSR analysis with 11 anchored primers also generated 116 scorable loci, of which 92 were polymorphic (79.3%). The estimated Jaccard similarity coefficient ranged from 0.44 to 0.76 for the ISSR markers. Cluster analysis was carried out, based on the Unweighted Pair Group Method with Arithmetic Averages (UPGMA) and the dendrogram drawn with the help of the NTSYSpc 2.02 software. The association between G316A growth hormone polymorphism and quantitative traits was investigated in an F2 population of pigs. Association analyses were performed using a statistical model that included genotype, sex, batch and sex by genotype interaction as fixed effects and sire as random effect. MUC1 is a heavily glycosylated mammalian transmembrane protein expressed by mucosal secretory tissues for both protection against microbial infection and lubrication. An important characteristic of MUC1 is its variable number of tandem repeats (VNTR) containing several sites for O-glycosylation. The heart fatty acid-binding protein (HFABP) gene was sequenced in parental animals of a F2 crossing of boars of the Brazilian native Piau breed with commercial sows (Landrace x Large White Pietrain). Primers used for PCR were designed to amplify four exons of the gene. The PCR products were sequenced and compared with the GenBank sequences. Differences between the generated sequences and the GenBank sequences were observed for both genetic groups. A total of 246 F2 animals were genotyped using the Hinf I restriction enzyme. Two genotypes were identified, 198 being animals HH and 48 Hh. The greenbug Schizaphis graminum (Rondani) is one of the most important cereals pests in the world. Within populations of this species, several biotypes, which are clones that share same virulence relationships with cultivated plants, can be distinguished. Molecular markers have been used to genetically characterize insect populations because they are fast and cost effective. Characterization of Fusarium race 1.2 resistance in melon and mapping of a major QTL for this trait near a fruit netting locus National Institute of Agronomic Research

An Israeli breeding line, BIZ, resistant to all four races of Fusarium oxysporum f.sp. melonis (FOM), has been characterized regarding its response to FOM race 1.2. The infection process of a FOM 1.2 strain that expresses the GFP reporter protein was monitored, suggesting the timing and sites in which fungal progression differs between a resistant and a susceptible genotype. A mapping population derived from the cross PI 414723 and BIZ was used to construct a linkage map with ~90 traits and markers, and QTL analysis of FOM1.2 resistance was initiated. Other traits such as flesh color and rind netting were mapped as well. A major recessive QTL for FOM1.2 resistance was located in linkage group 2, near a major locus controlling net development.

Indica and japonica are two main subspecies of Asian cultivated rice (Oryza sativa L.) that differ clearly in morphological and agronomic traits, in physiological and biochemical characteristics and in their genomic structure. However, the proteins and genes responsible for these differences remain poorly characterized. In this study, proteomic tools, including two-dimensional electrophoresis and mass spectrometry, were used to globally identify proteins that differed between two sequenced rice varieties (93-11 and Nipponbare). This work aimed to proceed molecular characterization of seven banana accessions (Borneo, Grand Naine, 1304-06, 4249-05, 0337-02, 0323-03 and 4279-06) resistance to the nematode Radopholus similis. These accessions were selected taking in account the reproduction factor (RF) among 26 banana genotypes from a working collection belonging to Embrapa Mandioca e Fruticultura Tropical. The genomic DNA of the seven accessions was extracted, and 36 decamere primers had been used to obtain RAPD markers. The resulting markers were converted into a matrix of binary data. Conversion of an AFLP fragment into one dCAPS marker linked to powdery mildew resistance in melon National Institute of Agronomic Research

Resistance to powdery mildew races 1, 2 and 5 has been evaluated in F2/F3 families from a cross between the resistant line ‘TGR-1551’ and the susceptible Spanish cultivar ‘Bola de oro’. Resistance is controlled by two independent genes, one dominant and one recessive, that both confer resistance to races 1, 2 and 5 of Podosphaera xanthii. Bulked segregant analysis (BSA) and amplified fragment length polymorphism (AFLP) technique have been used for the identification of markers linked to powdery mildew resistance. The AFLP marker E38M82 has been converted into a derived cleaved amplified polymorphic sequences (dCAPS) marker. The marker developed was codominant and the estimated distance with the dominant gene for powdery mildew resistance was 5.7 cM.
