Botrytis cinerea is a necrotrophic pathogen causing pre- and post-harvest diseases in at least 235 plant species. It manifests extraordinary genotype and phenotype variation. One of the causes of this variation is transposable elements. Two transposable elements have been discovered in this fungus, the retrotransposon (Boty), and the transposon (Flipper). In this work, two complete (Boty-II-76 and Boty-II-103) and two partial (Boty-II-95 and Boty-II-141) long terminal repeat (LTR) retrotransposons were identified by an in silico genomic sequence analysis. Characterization of Fusarium race 1.2 resistance in melon and mapping of a major QTL for this trait near a fruit netting locus National Institute of Agronomic Research

An Israeli breeding line, BIZ, resistant to all four races of Fusarium oxysporum f.sp. melonis (FOM), has been characterized regarding its response to FOM race 1.2. The infection process of a FOM 1.2 strain that expresses the GFP reporter protein was monitored, suggesting the timing and sites in which fungal progression differs between a resistant and a susceptible genotype. A mapping population derived from the cross PI 414723 and BIZ was used to construct a linkage map with ~90 traits and markers, and QTL analysis of FOM1.2 resistance was initiated. Other traits such as flesh color and rind netting were mapped as well. A major recessive QTL for FOM1.2 resistance was located in linkage group 2, near a major locus controlling net development.

This work aimed to proceed molecular characterization of seven banana accessions (Borneo, Grand Naine, 1304-06, 4249-05, 0337-02, 0323-03 and 4279-06) resistance to the nematode Radopholus similis. These accessions were selected taking in account the reproduction factor (RF) among 26 banana genotypes from a working collection belonging to Embrapa Mandioca e Fruticultura Tropical. The genomic DNA of the seven accessions was extracted, and 36 decamere primers had been used to obtain RAPD markers. The resulting markers were converted into a matrix of binary data. Conversion of an AFLP fragment into one dCAPS marker linked to powdery mildew resistance in melon National Institute of Agronomic Research

Resistance to powdery mildew races 1, 2 and 5 has been evaluated in F2/F3 families from a cross between the resistant line ‘TGR-1551’ and the susceptible Spanish cultivar ‘Bola de oro’. Resistance is controlled by two independent genes, one dominant and one recessive, that both confer resistance to races 1, 2 and 5 of Podosphaera xanthii. Bulked segregant analysis (BSA) and amplified fragment length polymorphism (AFLP) technique have been used for the identification of markers linked to powdery mildew resistance. The AFLP marker E38M82 has been converted into a derived cleaved amplified polymorphic sequences (dCAPS) marker. The marker developed was codominant and the estimated distance with the dominant gene for powdery mildew resistance was 5.7 cM.

Development and characterization of microsatellite loci for Campomanesia xanthocarpa (Myrtaceae) and cross amplification in related species Biological Sciences

Campomanesia xanthocarpa is a native tree, of common occurrence in almost all Brazilian Forest formations, which has its fruits and timber with high commercial value. Using an enriched genomic library we isolated and characterized microsatellite loci for C. xanthocarpa (Myrtaceae), in order to estimate genetic diversity parameters for this and related species. Twenty-eight microsatellite loci were identified and ten of them successfully amplified and showed polymorphism in a sample of 96 individuals, from four natural populations. Development and validation of SSR markers for Coffea arabica L. Repositório Alice

With the objective of developing new SSR markers for Coffea arabica, two enriched genomic libraries with probes (GT)15 and (AGG)10 were constructed. A total of 835 clones were sequenced and 756 presented good quality sequences. Redundant sequences were observed for 113 clones (14.94%). SSRs were found in 287 clones (38%). An estimated size of 417.5Kb of the C. arabica genome was sampled, with an average of one SSR per 1.46Kb. Dinucleotide repeats were more frequent than trinucleotides. Four repeat sequences, (AG/CT)n, (AC/GT)n, (AAG/CTT)n, and (AGG/CCT)n represented 61.1% of the total observed. A total of 96 SSR primers were designed and tested by PCR for two C. arabica genotypes. Flexibility in Cucurbitaceae breeding management with an integrated research software National Institute of Agronomic Research

DORIANE SAS develops and commercializes integrated and modular software for research activity able to manage the breeding data with a high level of flexibility. This software is able to manage complex “experiment cycles” and molecular markers data required by Cucurbitaceae breeding. Thanks to two innovative concepts like "experiment cycle" and "multi level architecture", DORIANE reached this objective building a well organized library adapted for the whole research department and a powerful tool for assisted decision making. Moreover, its client-server architecture facilitates the transmission of information between researchers and laboratories.

The objectives of this work were to investigate the genetic variation in 79 soybean (Glycine max) accessions from different regions of the world, to cluster the accessions based on their similarity, and to test the correlation between the two types of markers used. Simple sequence repeat markers present in genomic (SSR) and in expressed regions (EST-SSR) were used. Thirty SSR primer-pairs were selected (20 genomic and 10 EST-SSR) based on their distribution on the 20 genetic linkage groups of soybean, on their trinucleotide repetition unit and on their polymorphism information content. The objective of this study was to analyze the diversity and discrimination of high-performance Brazilian rice cultivars using microsatellite markers. Twenty-nine rice cultivars belonging to EMBRAPA Arroz e Feijão germplasm bank in Brazil were genotyped by 24 SSR markers to establish their structure and genetic discrimination. It was demonstrated that the analyzed germplasm of rice presents an expressive and significant genetic diversity with low heterogeneity among the cultivars. Genetic structure of populations of Pissodes castaneus (De Geer) (Coleoptera, Curculionidae) using amplified fragment length polymorphism . The objective of this study was to determine the genetic structure of populations of Pissodes castaneus from different areas and on different species of Pinus using the PCR-AFLP technique. Twenty samples were analyzed, representing 19 populations from Brazil and one from Florence, Italy, which is the region of origin of P. castaneus . Genetic variability with mutagenic agents has been employed in plant breeding due to its significant contribution to the improvement of the existing rice genotypes by using genetically diverse parents. This study evaluated the genetic variability and diversity of mutant rice using quantitative traits and inter-simple sequence repeats (ISSR) molecular markers. A field experiment was carried out on M5 generation of 31 developed mutant lines and their parent (control). Morphological traits and 25 ISSR molecular markers were used as tools for determining cultivar identities and genetic diversity.
