Assessment of cow and farm level risk factors associated with Ureaplasma diversum in pasture-based dairy systems - A field study Anais da ABC (AABC)
NASCIMENTO-ROCHA,JOSEFA M.; OLIVEIRA FILHO,BENEDITO D. DE; ARNHOLD,EMANNUEL; PÔRTO,REGIANI N.G.; LIMA,SVETLANA F.; GAMBARINI,MARIA LUCIA.
ABSTRACT Potential risk factors for Ureaplasma diversum in the vaginal mucus of 1,238 dairy cows were included in a multivariate logistic regression model, based on the cow level (i.e., granular vulvovaginitis [+GVV], yearly milk production [4500 kg or more], pregnancy, predominance of Bos taurus [+Bos Taurus], score of corporal condition [at least 2.5], concomitant positivity for Escherichia coli [+E.coli]), and farm level i.e., milking room hygiene (-Milking room), dunghill location, and replacement female). Ureaplasma diversum was present in 41.1% of the samples. Independent risk factors for U. diversum were +GVV (odds ratio [OR], 1.31); +Mycoplasma spp (OR, 5.67); yearly milk production (4500 kg or more) (OR, 1.99); +Bos taurus (OR, 1.68); +E. coli...

Avian mycoplasmosis update Rev. Bras. Ciênc. Avic.
Nascimento,ER; Pereira,VLA; Nascimento,MGF; Barreto,ML.
Avian mycoplasmas occur in a variety of bird species. The most important mycoplasmas for chickens and turkeys are Mycoplasma gallisepticum (MG), M. synoviae (MS), and M. meleagridis. Besides, M. iowe (MI) is an emerging pathogen in turkeys, but of little concern for chickens. Mycoplasmas are bacteria that lack cell wall and belong to the class Mollicutes. Although they have been considered extracellular agents, scientists admit nowadays that some of them are obligatory intracellular microorganisms, whereas all other mycoplasmas are considered facultative intracellular organisms. Their pathogenic mechanism for disease include adherence to host target cells, mediation of apoptosis, innocent bystander damage to host cell due to intimate membrane contact,...

Differences in intracellular localization of corn stunt spiroplasmas in magnesium treated maize. Repositório Alice
NOME, C.; MAGALHAES, P. C.; OLIVEIRA, E.; NOME, S.; LAGUNA, I. G..
Maize plants infected with Spiroplasma kunkelii show symptoms similar to that of plants in a magnesium-deficient soil, and it has been shown that the spiroplasma alters the plants' magnesium absorption. In the current study we compared changes associated to either spiroplasma infection, two soil magnesium levels and their combinations. Plant symptoms were recorded and correlated with transmission electron microscopy observations. Plants grown on a high magnesium treatment showed no macroscopical alterations nor organelle ultrastructural alterations, while plants on a low magnesium treatment showed macroscopical vein yellowing and, ultrastructurally, they had most chloroplasts and mitochondrial membranes altered. Infected plants on a low magnesium treatment...

Differential metabolism of Mycoplasma species as revealed by their genomes Genet. Mol. Biol.
Arraes,Fabricio B.M.; Carvalho,Maria José A. de; Maranhão,Andrea Q.; Brígido,Marcelo M.; Pedrosa,Fábio O.; Felipe,Maria Sueli S..
The annotation and comparative analyses of the genomes of Mycoplasma synoviae and Mycoplasma hyopneumonie, as well as of other Mollicutes (a group of bacteria devoid of a rigid cell wall), has set the grounds for a global understanding of their metabolism and infection mechanisms. According to the annotation data, M. synoviae and M. hyopneumoniae are able to perform glycolytic metabolism, but do not possess the enzymatic machinery for citrate and glyoxylate cycles, gluconeogenesis and the pentose phosphate pathway. Both can synthesize ATP by lactic fermentation, but only M. synoviae can convert acetaldehyde to acetate. Also, our genome analysis revealed that M. synoviae and M. hyopneumoniae are not expected to synthesize polysaccharides, but they can take...

In search of essentiality: Mollicute-specific genes shared by twelve genomes Genet. Mol. Biol.
Souza,Rangel Celso; Almeida,Darcy Fontoura de; Zaha,Arnaldo; Morais,David Anderson de Lima; Vasconcelos,Ana Tereza Ribeiro de.
Mollicutes are cell wall-less bacteria with a genome characterized by its small size. Chromosomal rearrangements help these organisms evade host immune surveillance and hence cause disease. Our goal was to determine genes shared by Mollicutes genomes using the bidirectional best hit methodology. The twelve studied Mollicutes share 210 genes, most of which (> 60%) fall into the following COG categories: translation, ribosomal structure and biogenesis; DNA replication, recombination and repair; nucleotide transport and metabolism and energy production and conversion. Thirty Mollicute-specific genes were identified, 22 of them previously described as essential genes in Mycoplasma genitalium.

Insertion sequences as variability generators in the Mycoplasma hyopneumoniae and M. synoviae genomes Genet. Mol. Biol.
Loreto,Elgion Lúcio Silva; Ortiz,Mauro Freitas; Porto,Jorge Ivan Rebelo.
We have analyzed the sequenced genomes of three strains of Mycoplasma hyopneumoniae and one strain of M. synoviae, and have found three and two different transposable element families, respectively in each species. In M. hyopneumoniae, the Insertion Sequences of the IS4 family is represented by ISMHp1, a putatively active element. The IS3 family is represented by several degenerated sequences. A third element called tMH was found, which shows some characteristics reminiscent of retrotransposons. In M. synoviae, three different possibly active IS4 elements are present (ISMHp1-like; ISMs1 and IS1634-like elements). The IS30 family is represented by the degenerated IS1630-like element. The IS1634-like element is shown to be involved in chromosomal...

Microbial diversity associated with the hydrothermal shrimp Rimicaris exoculata gut and occurrence of a resident microbial community ArchiMer
Durand, Lucile; Zbinden, Magali; Cueff-gauchard, Valerie; Duperron, Sebastien; Roussel, Erwan; Shillito, Bruce; Cambon-bonavita, Marie-anne.
Rimicaris exoculata dominates the megafauna of several Mid-Atlantic Ridge hydrothermal sites. Its gut is full of sulphides and iron-oxide particles and harbours microbial communities. Although a trophic symbiosis has been suggested, their role remains unclear. In vivo starvation experiments in pressurized vessels were performed on shrimps from Rainbow and Trans-Atlantic Geotraverse sites in order to expel the transient gut contents. Microbial communities associated with the gut of starved and reference shrimps were compared using 16S rRNA gene libraries and microscopic observations (light, transmission and scanning electron microscopy and FISH analyses). We show that the gut microbiota of shrimps from both sites included mainly Deferribacteres, Mollicutes,...

Molecular characterization of a phytoplasma associated with a commercial variety of Momordica charantia Scientia Agricola
Munhoz,Elizeu Merizio; Pereira,Thays Benites Camargo; Bedendo,Ivan Paulo.
ABSTRACT: Momordica charantia (bitter melon) presents two distinct types or varieties, known as wild type and commercial type. Plants of the wild type are hosts of a phytoplasma of the group 16SrIII-J, which is associated with a disease known as witches’ broom. However, this disease has not yet been reported in commercial bitter melon. Thus, symptomatic plants of the commercial type were analyzed in order to demonstrate the association between phytoplasmas and disease. In further assays, strains found in symptomatic plants of the commercial type were subjected to analysis of sequences of the secY gene to determine the extent of genetic diversity. Amplification of DNA fragments from genes 16Sr rRNA (1.2Kb) and secY (1.6Kb) revealed association of...

Phytoplasma associated with shoot proliferation in begonia Scientia Agricola
Ribeiro,Luiz Fernando Caldeira; Mello,Ana Paula de Oliveira Amaral; Bedendo,Ivan Paulo; Gioria,Ricardo.
Begonia is a very appreciated genus of ornamental plants, of economic relevancy, having species of flowers and foliage. In commercial croppings, plants exhibiting characteristic symptoms of phytoplasma infection have been observed, such as shoot proliferation, reduced plant, size small leaves and flowers, and phyllody. Leaves were sampled and total DNA was extracted to be used in nested Polymerase Chain Reaction (PCR), in order to detect and identify an expected phytoplasma. The results confirmed consistently the presence of a phytoplasma associated with symptomatic plants through the amplification of a typical genomic fragment of 1.2 kb by using the universal primers R16mF2/mR1 and R16F2n/R2. The use of specific primers R16(III)F2/R1 allowed to identify...
