The 2010 explosion of the Deepwater Horizon (DWH) oil rig generated the largest marine oil spill in US history with millions of barrels of crude oil released in the Gulf of Mexico (GoM). The eastern oyster, Crassostrea virginica, is an ecologically and economically important species in the northern GoM. Due to its biological characteristics (sessile, filter feeding), juvenile oysters may have been affected. Oyster sex determination is influenced by temperature - First clues in spat during first gonadic differentiation and gametogenesis ArchiMer

The sex-determining system of Crassostrea gigas is still poorly known, especially regarding the potential influence of temperature. In order to address this question, mRNA expressions of actors of the molecular cascade (Cg-DMI, Cg-SoxE, Cg-beta-catenin, Cg-Foxl2/Cg-FoxI2os) and of Oyvlg, a germ cell marker, were investigated by real-time PCR in spat grown at different temperatures (18, 22, 25 and 28 degrees C). In parallel, gonadic differentiation, gametogenesis and sex ratios were assessed by histology at each of these temperatures. Whatever the temperature, Cg-DMI, Cg-SoxE, Cg-beta-catenin and Oyvlg expressions peaked at the same developmental stage, always after Cg-Foxl2/Cg-Foxl2os (around 40-44 dpf for spat grown at 18 degrees C). Early detection of Pacific oyster spat infected with ostreid herpesvirus 1 (OsHV-1) could prevent introduction of OsHV-1-infected individuals into farming areas or onshore rearing facilities, thus reducing the risk of infection of naïve oysters in such production systems. Experiments were conducted on several hundred oyster spat provided by producers in order to examine whether early rearing practices could be considered as potential risk factors for (1) OsHV-1 infection as detected by molecular methods and (2) spat mortality experimentally induced through thermal challenge. Recherche et identification de bactéries pathogènes au sein d'une écloserie nurserie d'huître creuse, Crassostrea gigas, en relation avec la mortalité estivale du naissain ArchiMer

Summer mortality has been observed on oyster spat Crassostrea gigas since 1991 on french coasts. In our study, we deseribed symptoms observed during the mortality, especially adductor muscle weakness and we isolated strains on Zobell medium and TCBS from moribund spat. Strains isolated during a high mortality (60 %) were selected by experimental pathology on spat. Inoculation by bath led us to select a pathogenic strain (TNEMF6) for oyster spat. Phenotypic identification, phylogenetic study on 16S rDNA and quantitative DNA/DNA hybridisation showed that this strain belongs to Vibrio splendidus biovar II. Phenotyping and ribotyping with strains isolated during the study showed that only TNNIII7 and TNMB5 are similar to the pathogenic strain TNEMF6....

Recruitment of the Pacific oyster Crassostrea gigas in a shellfish-exploited Mediterranean lagoon: discovery, driving factors and a favorable environmental window ArchiMer

In the context of increasing demand for environmental recovery, aquatic systems may face the challenge of evolving under oligotrophication. This is the case in Mediterranean lagoons, in particular the shellfish-farmed Thau lagoon in France, where we studied recruitment of the Pacific oyster Crassostrea gigas. Oyster spat and environmental parameters were monitored at several sampling sites for 3 yr (2012 to 2014) using an original method with a temporal overlap deployment of collectors to study pre- and post-settlement processes and to identify the best conditions for recruitment.
