A naturally occurring recombinant isolate of Lettuce mosaic virus Inra

LMV-Common and LMV-Most are two seed-borne types of Lettuce mosaic virus (LMV), genus Potyvirus. LMV-Most, but not LMV-Common, overcomes the resistance afforded to lettuce by two recessive genes, mo1 1 and mo1 2. An RT-PCR-based assay thought to be specific for LMV-Most also amplified LMV-Tn2, previously typified as LMV-Common. The sequence of selected regions along the genome indicated that LMV-Tn2 is a natural recombinant between LMV-Most and LMV-Common isolates, with a putative recombination site located within the P3 coding region. This is the first evidence of a naturally occurring LMV recombinant isolate.

A new lineage sheds light on the evolutionary history of Potato virus Y Inra

Potato virus Y (PVY) is one of the rare plant viruses for which some biological traits (host range and symptomatology) are highly correlated with phylogeny, allowing the reconstruction of the evolutionary history of these traits. In this article, a new lineage of PVY isolates from Chile is described, showing unique genomic and biological properties. This lineage was found to be the sister group of all other PVY isolates and helped in the reconstruction of the ancestral traits and evolutionary history of PVY, suggesting that veinal necrosis in tobacco is an ancestral state and that adaptation to pepper (Capsicum spp.) and potato (Solanum tuberosum) has been modified several times during PVY history

A novel natural mutation in HC-Pro responsible for mild symptomatology of Zucchini yellow mosaic virus (ZYMV, Potyvirus) in cucurbits Inra

A natural mild isolate of Zucchini yellow mosaic virus was found to contain a mutation in the helper component (HC-Pro) within a conserved motif, “CDNQLD”, located 12 residues downstream from the “FRNK” motif involved in symptom severity. Introducing the mutation in an infectious cDNA clone of ZYMV resulted in an almost complete absence of symptoms, although viral accumulation was only partially reduced. The FRNK(X)12CDNQLD sequence might be part of a larger motif that is conserved in potyviruses and plays a role in symptomatology and/or silencing inhibition

Molecular cloning of recessive resistance genes to potyviruses in a large range of host species identified the eukaryotic translation initiation factor 4E (eIF4E) as an essential determinant in the outcome of potyvirus infection. Resistance results from a few amino acid changes in the eIF4E protein encoded by the recessive resistance allele that disrupt the direct interaction with the potyviral protein VPg. In plants, several loci encode two protein subfamilies, eIF4E and eIF(iso)4E. While most eIF4E-mediated resistance to potyviruses depends on mutations in a single eIF4E protein, simultaneous mutations in eIF4E (corresponding to the pvr2 locus) and eIF(iso)4E (corresponding to the pvr6 locus) are required to prevent pepper veinal mottle virus (PVMV)...

Amélioration et valorisation de l'igname : rapport d'exécution pour la période du 01.07.92 au 31.08.93 IRD

Cette seconde étude a été centrée sur la connaissance de la culture #in vitro$ de l'Igname, la sanitation des vitroplants par culture de méristème, par thermothérapie et par chimiothérapie. Cette étude a également comporté l'étude de la diversité biologique du virus de la Mosaïque de l'Igname (YMV), la mise au point d'un kit de diagnostic, sa connaissance particulière de virus au Burkina Faso (distribution géographique, épidémiologie, diversité), en Guadeloupe et en Guyane. L'indexation de la collection de vitroplants a permis de diagnostiquer en dehors du YMV, un virus de type Potexvirus et de soupçonner la présence d'un Cucumovirus et d'un autre Potyvirus. Biological and serological diversity of a potyvirus infecting maize in the Mediterranean area Inra

Le potyvirus dominant sur maïs dans la moitié sud de la France et la partie nord de l’Espagne et de l’Italie est le virus de la mosaïque nanisante du maïs souche A (MDMV-A). Ce virus est présent principalement en production de maïs doux et de maïs semence dans les zones ou le sorgho dAlep (Sorghum halepense L) est abondant. Notre travail avait pour but de mettre au point une méthode de diagnostic et d’amorcer l’étude de la biodiversité de ce pathogène. Une collection de plus de 100 isolats a été constituée. Biological characterization and complete nucleotide sequence of a Tunisian isolate of Moroccan watermelon mosaic virus Inra

During a survey conducted in October 2005, cucurbit leaf samples showing virus-like symptoms were collected from the major cucurbit-growing areas in Tunisia. DAS-ELISA showed the presence of Moroccan watermelon mosaic virus (MWMV, Potyvirus), detected for the first time in Tunisia, in samples from the region of Cap Bon (Northern Tunisia). MWMV isolate TN05-76 (MWMV-Tn) was characterized biologically and its full-length genome sequence was established. MWMV-Tn was found to have biological properties similar to those reported for the MWMV type strain from Morocco. The alkaloids contained in Colchicum autumnale seeds are used in numerous medicines. Good quality seeds are difficult to obtain from this undomesticated plant. Therefore, a research program was set up aiming to cultivate C. autumnale in order to improve alkaloid contents and seed yields. In this context, a collection was established in 1999 by transplanting corms from twelve different locations in Eastern France. However, serious symptoms of necrosis and decay have appeared in this collection since 2001. Characterization of a new potyvirus infecting pepper crops in Ecuador Inra

Sequencing 2,951 nucleotides of the 3′ proximal region of the genome of a potyvirus isolate collected from Capsicum pubescens (rocoto) pepper in Ecuador revealed that this was the first representative of a new species tentatively named Ecuadorian rocoto virus (ERV). Phylogeny reconstruction showed that this isolate clustered with potato virus V (PVV), Peru tomato virus and wild potato mosaic virus into a monophyletic group, and was closest to PVV. The isolate was shown to be infectious in tobacco, tomato and, contrary to PVV, in pepper. The pvr2 1 , pvr2 2 , and Pvr4 genes present in many pepper cultivars conferred resistance toward this isolate and could help control ERV

Characterization of peanut mottle virus in Cote d'Ivoire IRD

Un virus a été récemment isolé de l'Arachide dans la région septentrionale de la Côte d'Ivoire, pays où il n'avait pas encore été diagnostiqué. Il provoque, sur les feuilles, des marbrures, des dépressions internervaires ainsi que des taches très typées vert clair et vert foncé. Ce virus a été caractérisé ; il s'agit d'une souche du virus de la Marbrure de l'Arachide (Peanut Mottle Virus), virus membre du groupe des Potyvirus ; les études ont porté sur la gamme des plantes hôtes, le mode de transmission, la morphologie des particules, les propriétés physico-chimiques et sérologiques, en comparaison avec d'autres virus de l'Arachide. Contribution of host plant resistance and geographic distance to the structure of Potato virus Y (PVY) populations in pepper in northern Tunisia Inra

The genetic structure of Potato virus Y (PVY) populations was investigated in naturally-infected pepper (Capsicum annuum) fields, collected at eight different localities in northern Tunisia, where 23% of the sampled plants were homozygous for the pvr21 recessive resistance allele, while the other plants carried the dominant susceptibility allele pvr2+. En Guadeloupe, le virus 1 de la mosaïque de la pastèque, Papaya Ringspot Virus-Watermelon (PRSV-W)= Watermelon Mosaic Virus 1 (WMV-1), est très répandu dans les cultures de courgette où il provoque des symptômes de mosaïque avec cloques et des déformations filiformes très marquées sur le feuillage et également des déformations de fruits. Depuis 1982, on a constaté qu’un autre faciès maladif était aussi épidémique dans les champs de courgette. Determination of the best conditions for the regeneration of microplants and the elimination of YMV from excised meristems of yam nodal cuttings (Dioscorea spp.) IRD

Les effets du milieu de culture, du conditionnement et de la dimension des méristèmes ont été étudiés à différents temps de croissance des méristèmes et des microplants, dans l'objectif d'éliminer le virus de la Mosaïque de l'Igname. Deux espèces de #Dioscorea$ infectées ont été étudiées : #D. cayenensis-rotundata$ et #D. praehensilis$. 76 % de la première espèce et 20% de la seconde se sont révélés indemnes de virus à l'issu du traitement. Distribution géographique du virus de la mosaïque de l'igname au Burkina Faso IRD

L'Igname est sensible à une maladie provoquée par le virus de la Mosaïque de l'Igname. Afin de prévenir l'extension de la maladie par des échanges de matériel végétal, une étude de sa répartition géographique a été effectuée au Burkina Faso dans trois zones de culture. La situation écologique de la région Centre est particulièrement intéressante avec près de 100% des plantes atteintes ; la variété principalement cultivée, Igname de Pilimpikou, est spécifique de cette région. En outre la détection à l'aide d'anticorps monoclonaux indique une diversité biologique liée à la distribution géographique. (Résumé d'auteur)

dN/dS-based methods detect positive selection linked to trade-offs between fitness traits in the coat protein of Potato virus Y Inra

The dN/dS ratio between nonsynonymous and synonymous substitution rates is used extensively to identify codon positions involved in adaptation. However, the accuracy of this approach is questioned and few studies have attempted to validate experimentally its predictions. Using the coat protein (CP) of Potato virus Y (PVY), we identified several positively-selected codon positions with the softwares PAML and HyPhy. These positions differed between PVY clades.
