ABSTRACT Fungi of Pleurotus genus have attracted a great interest due to their medicinal properties such as anti-inflammatory, antimicrobial and antitumor. These properties are attributed mainly to polysaccharides synthesized by Pleurotus. This work aimed to study the mycelial growth of P. ostreatus in submerged culture, evaluating the influence of the initial concentration of substrate (20 and 40 g/L of glucose) and the pH (4 and 6) on kinetic parameters of production of biomass. The effectiveness of different doses (10, 30 and 50 mg/kg) of a mycelium polysaccharide fraction extracted from P. ostreatus in reducing Sarcoma 180 development in mice was also verified. In the range of this study, maximum concentration of mycelial biomass (about 12.8 g/L) was...

Aflatoxin detoxification by manganese peroxidase purified from Pleurotus ostreatus BJM
Yehia,Ramy Sayed.
Manganese peroxidase (MnP) was produced from white rot edible mushroom Pleurotus ostreatus on the culture filtrate. The enzyme was purified to homogeneity using (NH4)2SO4 precipitation, DEAE-Sepharose and Sephadex G-100 column chromatography. The final enzyme activity achieved 81UmL-1, specific activity 78 U mg-1 with purification fold of 130 and recovery 1.2% of the crude enzyme. SDS-PAGE indicated that the pure enzyme have a molecular mass of approximately 42 kDa. The optimum pH was between 4-5 and the optimum temperature was 25 ºC. The pure MnP activity was enhanced by Mn2+,Cu2+,Ca2+ and K+ and inhibited by Hg+2 and Cd+2.H2O2 at 5 mM enhanced MnP activity while at 10 mM inhibited it significantly. The MnP-cDNA encoding gene was sequenced and determined...

Apati,Giannini Pasiznick; Furlan,Sandra Aparecida; Laurindo,João Borges.
Dehydration and rehydration processes of Pleurotus ostreatus fruiting bodies were investigated in this work. Mushroom samples were dehydrated at 40, 50 and 60 ºC, using drying air with relative humidity of 75 %. The rehydration was investigated at different temperatures of immersion water (25, 55 and 85 ºC) and different immersion times (30, 75 and 120 minutes). The best rehydration occurred for the samples dried at 40 ºC. The rehydration could be done in water at room temperature, during 30 minutes. Water sorption isotherms of samples were determined at 30, 40 and 50 ºC. Both GAB and BET models satisfactorily represented the experimental data of moisture sorption of dried mushrooms.

ALCÂNTARA, M. C.; LIMA, E. A. de; GOES, B. C.; HELM, C. V.; AUER, C. G.; ZEFERINO, I..

Efficiency of treatments for controlling Trichoderma spp during spawning in cultivation of lignicolous mushrooms BJM
Colavolpe,María Belén; Mejía,Santiago Jaramillo; Albertó,Edgardo.
Trichoderma spp is the cause of the green mold disease in mushroom cultivation production. Many disinfection treatments are commonly applied to lignocellulose substrates to prevent contamination. Mushroom growers are usually worried about the contaminations that may occur after these treatments during handling or spawning. The aim of this paper is to estimate the growth of the green mold Trichoderma sp on lignocellulose substrates after different disinfection treatments to know which of them is more effective to avoid contamination during spawning phase. Three different treatments were assayed: sterilization (121 ºC), immersion in hot water (60 and 80 ºC), and immersion in alkalinized water. Wheat straw, wheat seeds and Eucalyptus or Populus sawdust were...

Mycelial growth of strains of Pleurotus ostreatus developed on agar and its correlation with the productivity in pilot production farm BJM
Sastre-Ahuatzi,M.; Téllez-Téllez,M.; Díaz-Godínez,G.; Montiel-González,A.M.; Díaz,R.; Sánchez,C..
Radial growth rate, intracellular laccases and proteases activities, and protein content were evaluated in five strains of Pleurotus ostreatus, grown on starch-based and glucose-based agar media containing different concentrations of the glucose analogue 2-deoxyglucose (2-DG). Productivity of the strains in pilot scale cultivation was also determined. The mycelium of four strains had approximately between 0.6- to 3-fold higher protein content when grown on glucose medium containing 0.01 g/L of 2-DG than when grown on glucose medium. The radial growth rate and intracellular laccases activity of some strains showed a positive and a negative correlation with the productivity, respectively. These results suggest that the strains with high radial growth rate...

MKHIZE,Senzosenkosi Surprise; CLOETE,Jacques; BASSON,Albertus Kotze; ZHARARE,Godfrey Elijah.
Abstract Improving the performance of mushroom in terms of high production and fast growth rate is essential in mushroom cultivation. In the present study the performance of Pleurotus ostreatus was evaluated using varying levels of wheat bran (WB) and maize flour (MF). The results indicated that Pleurotus ostreatus was highly influenced by different levels of supplementation, with 8% WB, 18% WB and 2% MF having higher contamination rate. The low levels of supplementation gave significantly better mycelial growth rate (MGR) and shorter colonisation period as observed that the control had highest MGR whereby 20% MF had lowest MGR. The pinning time (TP) was shortest at the first flush with minimum of 3 days (12% MF). The higher levels of supplementation...

Romero Arenas, Omar.

Recycling of the biomass waste defatted almond meal as a novel nutritional supplementation for cultivated edible mushrooms Agronomy
Pardo-Giménez, Arturo; Carrasco, Jaime; Roncero, José María María; Álvarez-Ortí, Manuel; Cunha Zied, Diego; Pardo-González, Jose Emilio.
Defatted almond meal (DAM) is an useless biomass waste obtained after oil extraction. The substrate designed for mushroom cultivation is achieved through a controlled composting process from agricultural by-products (chicken manure and wheat straw). This work shows the potential of DAM as efficient compost supplement for culturing the species Agaricus bisporus (J.E. Lange) Imbach and Pleurotus ostreatus (Jacq.) P. Kumm. Supplementation during A. bisporus cultivation results in larger mushrooms with a firmer texture and higher dry matter and protein contents in comparing with the non-supplemented substrate. In P. ostreatus, supplementation at a dosage of 15 g kg-1 provided a yield improvement up to 31.8%, compared to the control without supplement. The...

Use of remazol blue dyed avicel for the determination of cellulolytic activity in basidiomycetes Rev. Microbiol.
Correia,Marcos José; Pereira Junior,José Antônio de Sousa; Santos,Jefferson Cunha dos; Cavalcanti,Maria Auxiliadora de Queiroz.
A modified method for direct determination of cellulolytic activity using Avicel colored with Remazol Brilliant Blue R (RBBR) in Agar test tubes is discussed. Refinements were introduced in a simple method for quantitation of cellulase activity, based on the release of dye from Avicel-RBBR medium by the enzymatic hydrolysis. Modifications in Avicel-dye preparation were enhanced and a spectrophotometer for direct OD measurement in agar test tubes used. The use of a spectrophotometer improved the precision of the collected data, since absorbance measurements could be done at the maximum wavelenght for RBBR (595 nm).
