ABSTRACT: The absolute quantification determines the absolute amount of a targeted nucleic acid expressed as a copy number or concentration. The knowledge of virus concentrations in commercial crops possesses high relevance to ensure a reliable diagnosis. The objective of this study was to perform an absolute quantification of five viruses in infected grapevines (Vitis spp.). Different known amounts of the standard sample (cloned viral cDNA or in vitro transcribed viral RNA) were quantified by TaqMan RT-qPCR. Based on these data, standard curves were generated plotting Ct values (threshold cycle) against the log of the standard sample amount. En este estudio se determinaron las relaciones filogenéticas y los niveles de variación de aislamientos de PVX obtenidos en tejidos foliares de plantas de Solanum tuberosum subsp. andigena var. Diacol-Capiro y S. phureja var. Criolla Colombia en Antioquia, utilizando métodos de secuenciación de nueva generación (NGS) y de Sanger. Inicialmente, se detectó el PVX mediante DAS-ELISA (Agdia-PSA10000), confirmándose su presencia en ocho de las muestras por Inmunocaptura-RT-PCR en tiempo real (IC-RT-qPCR). Los resultados de las pruebas serológicas indicaron la infección de PVX en 14,7 % y 13,3 % de las muestras de Diacol-Capiro y Criolla Colombia, respectivamente. RESUMEN Las enfermedades virales son uno de los principales problemas fitopatológicos de la papa. Con el fin de determinar los virus más prevalentes en cultivos de papa var. Diacol Capiro en el oriente Antioqueño (Colombia), se evaluó mediante RT-qPCR la presencia de diez virus de ARN (PVY, PVA, PVV, TaLMV, PVS, PLRV, PYVV, PVX, ToRSV y PMTV) en 36 muestras de tejido foliar. Los resultados indicaron la ocurrencia de cinco de los diez virus evaluados, con niveles de prevalencia de 88,9 %, 75 %, 75 %, 41,7 % y 25 % para PVY, PVX, PYVV, PLRV y PVS, respectivamente. Las virosis son uno de los problemas fitosanitarios más limitantes para la producción de papa, siendo los virus Potato leafroll virus (PLRV), Potato virus Y (PVY), Potato virus S (PVS) y Potato yellow vein virus (PYVV) los que generan mayores pérdidas económicas en la región andina. ABSTRACT This study aimed to evaluate the effects of priming on seed germination under salt stress and gene expression in seeds and seedlings of P. angulata L. After priming for 10 days, seed germination was tested in plastic trays containing 15 ml of water (0 dS m-1 - control) or 15 ml of NaCl solution (2, 4, 6, 8, 10, 12, 14 and 16 dS m-1). ABSTRACT Gene expression of CDKN1A, CDKN1B, and TP53, and immunostaining of p21, p27 and p53 were evaluated to verify the role of these cell cycle inhibitors in canine prostates with proliferative inflammatory atrophy-PIA and prostatic carcinoma-PC. Seventy samples, 15 normal, 30PIA and 25PC. Regarding number of p27 and p53 labeled cells, difference between normal and PIA and PC was observed, as well as between PIA and PC for p53.
