Babesia bovis: Effects of cysteine protease inhibitors on in vitro growth OAK
Okubo, Kazuhiro; Yokoyama, Naoaki; Govind, Yadav; Alhassan, Andy; Igarashi, Ikuo.
In the present study, we examined the effects of four kinds of cysteine protease inhibitors (E64, E64d, leupeptin, and ALLN) on the in vitro asexual growth of Babesia bovis. Of these, only the lipophilic inhibitors, E64d and ALLN, were found to effectively inhibit the growth of B. bovis. In further experiments, E64d, but not ALLN, significantly suppressed the parasite’s invasion of host erythrocytes, while both chemicals, especially ALLN, inhibited the parasite’s replication within the infected erythrocytes. These data suggested the presence of cysteine protease(s) derived from B. bovis, in which the protease(s) would play important roles in the erythrocyte invasion and/or replication processes of the parasite.

Effect of actinomycin D on simian rotavirus (SA11) replication in cell culture BJMBR
Stefanelli,C.C.; Castilho,J.G.; Botelho,M.V.J.; Linhares,R.E.C.; Nozawa,C.M..
Rotaviruses are the major cause of viral diarrhea in humans and animals. Actinomycin D (Act D) is an antibiotic that intercalates DNA and therefore inhibits DNA-dependent transcription. The current study was carried out to assess the influence of Act D on the replication of simian rotavirus (SA11) in cell culture. Virus-infected MA-104 cell cultures were studied in the presence of Act D at concentrations of 1.25 and 2.5 µg/ml. Treatment of rotavirus-infected cells with 2.5 µg/ml Act D 48 h post-infection reduced the cytoplasmic metachromasia after staining with acridine orange by 25%. Viral RNA labeled with ³H-uridine in the presence of the drug was separated by polyacrylamide gel electrophoresis. Viral RNA replication was not affected by Act D, but...

Leishmania amazonensis promastigotes present two distinct modes of nucleus and kinetoplast segregation during cell cycle. Repositório Alice
SILVA, M. S. da; MONTEIRO, J. P.; NUNES, V. S.; VASCONCELOS, E. J.; PEREZ, A. M.; FREITAS JÚNIOR, L. de H.; ELIAS, M. C..
Abstract: Here, we show the morphological events associated with organelle segregation and their timing in the cell cycle of a reference strain of Leishmania (L.) amazonensis promastigotes, the main causative agent of Tegumentary leishmaniasis in the Americas. We show evidences that during the cell cycle, L. amazonensis promastigotes present two distinct modes of nucleus and kinetoplast segregation, which occur in different temporal order in different proportions of cells. We used DAPI-staining and EdU-labeling to monitor the segregation of DNA-containing organelles and DNA replication in wild-type parasites. The emergence of a new flagellum was observed using a specific monoclonal antibody. The results show that L. amazonensis cell cycle division is...

Limited replication of yellow fever 17DD and 17D-Dengue recombinant viruses in rhesus monkeys Anais da ABC (AABC)
Trindade,Gisela F.; Marchevsky,Renato S.; Fillipis,Ana M.B. de; Nogueira,Rita M.R.; Bonaldo,Myrna C.; Acero,Pedro C.; Caride,Elena; Freire,Marcos S.; Galler,Ricardo.
For the development of safe live attenuated flavivirus vaccines one of the main properties to be established is viral replication. We have used real-time reverse transcriptase-polymerase chain reaction and virus titration by plaque assay to determine the replication of yellow fever 17DD virus (YFV 17DD) and recombinant yellow fever 17D viruses expressing envelope proteins of dengue virus serotypes 2 and 4 (17D-DENV-2 and 17D-DENV-4). Serum samples from rhesus monkeys inoculated with YFV 17DD and 17D-DENV chimeras by intracerebral or subcutaneous route were used to determine and compare the viremia induced by these viruses. Viral load quantification in samples from monkeys inoculated by either route with YFV 17DD virus suggested a restricted capability of...
