A microarray-based analysis of oocyte quality in the European clam Ruditapes decussatus ArchiMer

A microarray-based analysis was performed with the objective of describing genomic features of oocytes and looking for potential markers of oocyte quality in the economically important European clam, Ruditapes decussatus. Oocytes of 25 females from Ria de Aveiro, Western coast of Portugal (40°42′N; 08°40′W) were selected for this study and oocyte quality was estimated by success of D-larval rate under controlled conditions, which appeared to vary from 0 to 95 %. By genome-wide expression profiling with a DNA microarray, 526 probes appeared differentially expressed between two groups representing the largest and smallest value of D larval rates, named good (represented by a mean D-larval yield of 57 ± 22%) and poor (9 ± 5%) quality oocytes. En 2004 et 2005, une étude menée par le REPAMO (Réseau de Pathologie des Mollusques) a montré que des parasites du genre Perkinsus étaient présentas au sein des principaux domaines français de production de palourdes: La Mer Méditerranée, l'Océan Atlantique et la Manche avec une forte prévalence amis aucun cas de mortalités associées. Measurements of oxygen consumption in the two species Ruditapes decussatus and Ruditapes phi/ippinarum were perfonned using hatchery-reared juveniles, grown under controlled conditions. The temperature range varied from 10 to 30°C. The linear relations between the logarithms of the dry weight and of the oxygen consumption were calculated for each species. Rates were compared in terms of standardized clam tissue weight calculated from the regression of respiration on tissue weight. Such a technique allows to make comparisons between the species by avoiding any interference from the variations in individual weight. Dans le cadre du Réseau zoosanitaires CEE (huître plate) et suivi d'autres espèces, 890 individus provenant des zones 5, 6 et 7 ont été examinés sur l'ensemble de l'année 1995 par différentes techniques : 472 analyses sous coupes histologiques, 328 analyses sur appositions d'organes, 90 analyses sur milieu au thioglycolate.

Correlation between perkinsosis and growth in clams Ruditapes spp. ArchiMer

Perkinsosis is one of the most widespread diseases affecting commercially important species of molluscs globally. We examined the impact of Perkinsus spp. on shell growth at the individual scale in 2 clam species: Ruditapes decussatus from Mundaka Estuary (Spain) and R. philippinarum from Arcachon Bay (France). At Arcachon, 2 contrasting sites in terms of environment and Perkinsus olseni presence were chosen: Arguin (disease-free) and Ile aux Oiseaux (infected site). Données sur la biologie de Ruditapes philippinarum et Ruditapes decussatus en élevage sur la côte morbihannaise ArchiMer

Des essais d'élevage de palourdes en milieu ouvert ont été réalisés sur le littoral morbihannais en 1979 et 1980. R. philippinarum atteint une taille commercialisable au bout de 18 mois à 2 ans suivant le site et la densité au semis. Les taux de survie observés sont supérieurs à 50% pour les essais réalisés en 1979, mais inférieurs pour ceux effectués en 1980. Une mortalité quasi totale de R. decussatus a été observée après 6 mois d'élevage, et la croissance des individus survivants est nettement moins rapide que pour l'espèce précédente. Les résultats montrent que R. philippinarum est mieux adaptée, et ils permettent de proposer un protocole d'élevage pour le secteur considéré.

Ecologie du copépode calanoïde Paracartia grani. Implication dans le cycle de vie du parasite Marteilia refringens dans la lagune de Thau ArchiMer

In this study, the life cycle of Paracartia grani, a calalloid copepod belonging to the Acartiidae family was determined in Thau lagoon. Indeed, the copepod involvement in the life cycle of the parasite Marteilia refringens affecting the bivalve productions is suspected. Mesozooplanktonic community .was monitored twice a month. over two years at a fixed station in the lagoon. Sampling has identified P. grani as the acartiid dominant species in summer. From April to January, the copepod is found in the water column while from February to early April it remains in the sediment as diapausing eggs. Japanese clam (Ruditapes philippinarum) and European clam (Ruditapes decussates) farming based on spat produced in hatcheries has gone beyond the experimental stage on the Atlantic coasts since the research of Latrouite et Claude (1976), Lucas (1977), Flassch (1979), Dréno (1979) and Gras (soon to be published). It has thus become necessary to develop farming procedures reliable enough to be handed over to the professionals in the business.

Elevages de bivalves à partir de naissain d'écloseries dans les claires du bassin de Marennes-Oléron ArchiMer

Des élevages expérimentaux d'huîtres plates Ostrea edulis et de palourdes Ruditapes decussatus et Ruditapes philippinarum ont été réalisés dans les claires à huîtres du bassin de Marennes-Oléron à partir de naissain provenant d'écloserie. Les croissances pondérale et linéraire et le pourcentage de mortalité ont été étudiés. Les premiers résultats obtenus sont encourageants, et, plusieurs ostréiculteurs ont commencé, cette année, avec notre assistance, la culture des palourdes dans leurs claires à huîtres.

The authors study a chlamydial micro-organism parasite of the digestive epithelium of the carpet shell Ruditapes decussatus. The action of this microorganism seems of little import : the intracellular infection is limited to a small number of cells and does not cause any serious lesion in the digestive epithelium.

Using an experimental flowing system, the filtration rate of Mytilus galloprovincialis, Ostrea edulis, Ruditapes decussatus and Ruditapes aureus has been measured by an indirect technique under Mediterranean thermal conditions in coastal open sea. The oxygen consumption and excretion rate of ammonia have been estimated and influence of temperature and salinity tested for M. galloprovincialis . The results are discussed and compared to literature data.

La conchyliculture connaît depuis une trentaine d'années une progression régulière de ses productions à travers le monde. Elle représente une activité économique importante avec plus de 240 000 tonnes de coquillage produit annuellement en France. Cependant le développement de cette activité peut être limité par des mortalités parfois massives dues à des maladies infectieuses. On the contrary to the U. S. where they represent but 4.6% of the total marine production , mollusc, principally bivalves, can no longer be considered as a marginal production. Histological study of a cellular reaction in Ruditpaes decussatus infected by a protozoan ArchiMer

An infection caused by a Perkinsus -like parasite has been observed in Ruditapes decussatus from Portugal (Comps and Chagot, 1987). Following a pathological study carried out on clams affected by mortalities in the Algarve area, this parasite was found again, inducing a reaction process in the host. Histological examination reveals the parasite in the connective tissue of different organs. The adjoining tissues, particularly the epithelia, are not affected.

Induced triploidy in the European clam, Ruditapes decussatus (L.) and performance of triploid larvae ArchiMer

For the first time, effective treatments using cytochalasin B were developed to induce triploidy in the European clam, Ruditapes decussatus (L.). The percentage of triploid embrios was assessed by karyological or image analysis. Two treatments (O.5 or 1 mg of cytochasin B (CB per ml of dimethylsulfoxide in 1 l of sea water) were applied at different times after fertilization (10 to 25 mn), for two different periods (15 and 20 min). Best results were obtained for a CB concentration of 1 mg/l. When treatment was applied 15 min after fertilization for a 20 min period, 94% and 95% of triploid embryos were obtained in two repeated experiments.
