A carbon-14-glucose assay to compare microbial activity between rhizosphere samples Inra

The present work investigated to what extent the utilisation of 14C-glucose by soil microorganisms could be used to compare microbial activity between rhizosphere samples. A very low amount of 14C-glucose was added to the soil to be tested. After incubation for 6 days at a constant temperature in the dark, we determined the mineralised fraction (14CO2) and the 14C of the microbial biomass rendered extractable after fumigation with chloroform vapour (14CFE). In unplanted soils, we observed that the 14CO2/14CFE ratio increased when microbial growth was stimulated by addition of unlabelled glucose (20-500 µg C g-1 soil) compared to the control (no unlabelled glucose added). Accelerated mineralisation of atrazine in maize rhizosphere soil Inra

The mineralisation rate of atrazine measured in soil pre-treated with this herbicide, was significantly higher in the maize rhizosphere than in bulk soil. Maize rhizosphere was also shown to significantly increase microbial biomass C as compared with bulk soil. Ribosomal intergenic spacer analysis conducted on nucleic acids extracted directly from soil samples revealed that the structure of microbial communities observed in the rhizosphere was slightly different from that of bulk soil. The quantification of the relative amount of the gene atzC, which encodes an enzyme involved in atrazine mineralisation, was carried out on soil nucleic acids by using quantitative-competitive PCR assays. Amitrole degradation in vineyard soils in relation to pedo-climatic conditions Inra

The fate of [14C]-amitrole herbicide was studied in eight soils having different capacities for amitrole mineralisation. Laboratory incubations were run combining different experimental conditions: temperature (4, 28 and 50°C), soil moisture (50, 100 and 150% of soil water holding capacity) and microbial activity (sterile and non-sterile conditions). During incubation, samples of the soils were periodically extracted with 0.5 M NH4OH and extracts were analysed by HPLC. The lengths of time needed for 50% dissipation of amitrole (DT50) in soils ranged from less than 1 day to more than 70 days. Carbon decomposition of the topsoils and soil fractions under forest and pasture in the western Brazilian Amazon basin, Rondonia Inra

The topsoils of two sites, comprising natural forest and 4- and 20-year-old pastures, respectively, were selected in Rondônia to evaluate the changes of soil organic matter due to pasture establishment. These changes were evaluated by measuring the proportions of the C and N associated with clay and silt fractions, and by the C decomposition (CD) rate of the whole topsoils and their size fractions. The topsoils studied had large proportions of C and N associated with fine fractions, especially with clay fractions. Characterization of organic matter in topsoils under rain forest and pasture in the eastern Brazilian Amazon basin Inra

In topsoils under forest and 7-, 12- and 17-year-old pastures, organic matter was characterized by analysing C and N distribution in particle-size fractions, the C decomposition rates of soil and particle-size fractions and by employing density-fractionation of macro-organic matter (>150 μm). The C and N associated with clay fractions increased with increasing age of pasture. The weight (%) of macro-organic matter and its heavy fractions (>1.37 g cm–3) also increased with increasing age of pasture. Copper binding capacity of root exudates of cultivated plants and associated weeds Inra

Cu binding to root exudates of two cultivated plants, wheat (Triticum aestivum) and rape (Brassica napus), and two weeds associated with wheat, dog daisy (Matricaria inodora) and cornflower (Centaurea cyanus), was studied in vitro under hydroponic and sterile conditions. Nutrient solutions were prepared with or without P. A MetPLATE microbiological test was used to assess the metal complexing capacity of root exudates. In the P-deficient solutions, no exudation was observed for any of the four plants; consequently, no Cu binding occurred. Factors influencing carbon decomposition of topsoils from the Brazilian Amazon Basin Inra

The influence of texture and local climate, especially precipitation, on C decomposition was evaluated in topsoils under forest and pastures at different sites located in the western, central, and eastern Brazilian Amazon Basin. Independent of the geographical location and vegetation cover, C decomposition was lower in topsoils of fine texture with a sand/clay ratio of less than 1, compared to topsoils of coarse a texture with a sand/clay ratio ranging from 2 to 8. In the former, the low sand/clay ratio appears to reduce C decomposition. In the latter, besides the effect of the high sand/clay ratio, the local climate, especially the annual precipitation, seems to play an important role in controlling C decomposition.

How increasing availabilities of carbon and nitrogen affect atrazine behaviour in soils Inra

The effect of increasing amounts of glucose and mineral N on the behaviour of atrazine was studied in two soils. One had been exposed to atrazine under field conditions (adapted soil), the other had not (non-adapted soil), resulting, respectively, in an accelerated degradation of atrazine in the adapted soil and in a slow degradation of the herbicide in the non-adapted soil. The dissipation of 14C-atrazine via degradation and formation of non-extractable "bound" residues was followed during laboratory incubations in soils supplemented or not with increasing amounts of glucose and mineral N. Maize root mucilage decreases adsorption of phosphate on goethite Inra

This work was carried out to determine the influence of root exudates on phosphorus solubility in the presence of a strong phosphate sorbant. Root mucilage was collected from maize grown under field conditions. Polygalacturonic acid (PGA) was also used as a model of exudates. Adsorption of phosphate by synthetic goethite was measured in the absence of either (1) root mucilage collected from maize in the field, or (2) PGA. The ranking order of the adsorption on goethite was phosphate at low concentration >PGA >mucilage >phosphate at high concentration. L’étude microstructurale d’un sol calcaire issu de l’altération de marnes révèle la présence de nombreuses micro-structures calcaires se développant au niveau des discontinuités fissurales. Il s’agit de vestiges de racines de graminées, constitués de cellules calcifiées d’environ 100 μm de diamètre, indépendantes les unes des autres mais ayant conservé leur arrangement tissulaire. Soil surface crusts in rangelands and their role in runoff and erosion have long been recognized in tropical Australia. However, these crusts are much less documented than the crusts developed in southern and central arid and semi-arid Australia. Short-terme changes in the spatial distribution of microorganisms in soil aggregates as affected by glucose addition Inra

We studied the location of microorganisms in soil aggregates by low temperature scanning electron microscopy and by confocal laser microscopy (direct counts of bacteria) on dissected aggregates. We compared a sandy and a clayey soil and studied the effect of adding glucose on the spatial distribution of microorganisms. In the sandy soil, pores with diameters between 6 µm and 30 µm were the most abundant. In the clayey soil, the pores were <0.2 µm in diameter. In the sandy soil, microorganisms were present both on the surface and inside aggregates, whereas they were concentrated on the surface of clayey soil aggregates. After dissolution of fertiliser granules, a high nitrogen concentration is recovered in the immediate vicinity of granules, which may enhance damaging processes like nitrite accumulation or ammonia volatilisation. Based on the diffusion equations of Cranck, the granule-soil microsite was modelled to obtain the actual fertilised surface plot and the effective rate of N application on this surface.
