A method for measuring the settling velocity distribution of large biotic particles Inra

A simple method for measuring the settling velocity (V s) distribution of pollen and spores 30–100 μm in diameter is detailed and evaluated. The method is called the ‘settling tower' and consists in taking sequential pictures of particles falling under gravity in calm air. The scene is illuminated by a cold light source, while a camera takes 15 pictures per second. Between 20,000 and 100,000 images are analysed to obtain the distribution of V s for a given set of particles. The method was validated using two standard particles with mean diameters of 68 and 108 μm, respectively, as well as Lycopodium spores, with a mean diameter of 35 μm. Fungal pathogens are among the most virulent bioagressors of protected crops. For sustainable plant production and to protect the crop against these airborne organisms one must determine their origin, i.e. whether they come from outside the greenhouse or are produced inside it.We considered Botrytis cinerea spore concentration as a particular physical species which is transported by air in the same way as heat, CO2, water vapour or any tracer gas. Effects of fluctuating moisture and temperature regimes on the persistence of quiescent conidia of Isaria fumosorosea Inra

Conidia of Isaria fumosorosea were submitted to three regimes of temperature and moisture to simulate microclimatic conditions which prevail in temperate (43% RH and 28 degrees C to 98% RH and 15 degrees C), subtropical (75% RH and 35 degrees C to 98% RH and 25 degrees C), and arid areas (13% RH and 40 degrees C to 33% RH and 15 degrees C) with daily fluctuating cycles. Germination, conidial viability, and virulence to Spodoptera frugiperda larvae were less affected after 20 days exposure under temperate cycling conditions than under arid and subtropical conditions. Effects of wetting on the persistence of quiescent conidia of Isaria fumosorosea Inra

Quiescent conidia of Isaria fumosorosea were submitted to various wetting-drying cycles under different regimes of temperature and air humidity. Germination and viability of conidia collected on cultures freshly host-passed (P2) were not affected at 25 degrees C during five cycles at increasing wet phase duration (2-12 h per daily cycle) under any moisture conditions (13-86% RH). Infectivity levels remained stable, but mortality was slightly postponed. In vitro-cultured inocula (P5) were significantly affected after only one cycle at higher air humidity (75 and 86% RH) and temperature (35 and 40 degrees C). --BUTS DE L'ETUDE :Etudier les effets du nombre d’abeilles, de la présence de QMP (phéromone mandibulaire de la reine) et de la résistance naturelle au Varroa sur la résistance de l’abeille Apis mellifera au parasite Nosema ceranae.--METHODES & TECHNIQUES :Expériences en laboratoire dans des cagettes avec les différents facteurs. Analyse statistique avec le logiciel XLStat.--RESULTATS :Le Nosema augmente la mortalité dans les groupes de 30 et 100 abeilles avec ou sans QMP. Le pouvoir germinatif des urédospores de M. Larici-populina et de M. Allii-populina conservées au laboratoire à + 4 °C décroit rapidement et,comme en pépinière, tend à s’annuler après ou 8 mois. Si l’humidité relative est faible (34 p. 100) la conservation est portée à un an et les sporées germent à plus de 50 p. 100 durant les huit premiers mois.

Etude expérimentale de la libération des conidies d’Erysiphe graminis DC. f. sp. hordei sous l’effet du vent Inra

Des expériences effectuées sur des pustules isolées d’Erysiphe graminis f. sp. hordei, placées dans une petite soufflerie ont montré que la libération des spores est passive. Toute présence d’eau bloque la libération. Le seuil minimal de vent dépend de la longueur des chaînes de conidies de la pustule et donc du nombre de spores présentes ; aussi des périodes suffisamment longues sans vent où la sporulation est continue amènent un abaissement de ce seuil. La courbe reliant le nombre de spores libérées à la vitesse du vent a une forme en S plus ou moins redressé suivant la densité initiale de spores. Les spores arrachées à de fortes vitesses du vent sont immatures et germent mal. L’humidité relative de l’air environnant la pustule n’exerce aucune...
