A marine bacterial adhesion microplate test using the DAPI fluorescent dye: a new method to screen antifouling agents ArchiMer

Aims: To develop a method to screen antifouling agents against marine bacterial adhesion as a sensitive, rapid, quantitative microplate fluorescent test. Methods and Results: Our experimental method is based on a natural biofilm formed by mono-incubation of the marine bacterium Pseudoalteromonas sp. D41 in sterile natural sea water in a 96 wells polystyrene microplate. The 4'6-diamidino-2-phenylindole (DAPI) dye was used to quantify adhered bacteria in each well. Total measured fluorescence in wells was correlated with the amount of bacteria showing a detection limit of one bacterium per 5 µm2 and quantifying 2 x 107 to 2 x 108 bacteria adhered per cm2. Abstract: The accumulation of exopolysaccharides (EPS) produced by microorganisms occurs in the presence of excess substrate and limiting conditions of elements that are essential to growth, such as nitrogen, phosphorus, sulfur, and magnesium. The presence of EPS produced by bacterial cells contributes to slime colonies formation in solid medium and increased viscosity in liquid medium. This paper proposes an alternative method for screening EPS-producing lactic acid bacteria using solid medium-containing discs of filter paper that are saturated with active cultures. This research aimed to evaluate the potential of several native white rot fungi (WRF) isolated from subtropical environments of Misiones (Argentina) to produce different ligninolytic enzymes. Coriolus versicolor f. antarcticus BAFC 266, Pycnoporus sanguineus BAFC 2126 and Phlebia brevispora BAFC 633 showed the highest phenoloxidase activity. Ganoderma applanatum strain E, P. sanguineus BAFC 2126 and P. brevispora BAFC 633 revealed marked laccase and peroxidase activity. C. versicolor f. antarcticus, G. applanatum (strain A) and Trametes villosa, gave high positive reactions with 2,6-dimethoxyphenol oxidation at the lowest tested pH. Competing Screening Rules AgEcon

Various studies show that agricultural cooperatives behave differently than their investor-owned counterparts. One explanation may be that the internal decision making process differs in these two governance structures. A model is developed to explore how endogenous screening rules affect efficient organizational choices and industrial structures. It is shown that screening level choice may outweigh architecture choice and that screening rules are strategic substitutes. Conditions are derived under which cooperatives are efficient organizational forms. It is also shown that competition may increase the attractiveness of investor-owned firms and circumstances are determined in which cooperatives and investor owned firms coexist in equilibrium.

The leafminer, Liriomyza sativae Blanchard, is distributed around the world and is an important pest of vegetables and ornamentals. Given the resistance potential of the leafminer to current insecticides, the use of resistant plant cultivars and parasitoids could be effective integrated pest management (IPM) strategies against it. Cucumber (Cucumis sativus L.) is a preferred host for this insect. Seventeen cultivars of cucumber have been evaluated to study resistance mechanisms to L. sativae. All cucumber cultivars were evaluated in screening tests in greenhouse with indices such as the number of leafminer stings, the number of larval mines, the proportion of larval mines to leafminer stings, and the rate of injury. Genetic variability among wheat (Triticum aestivum L.) germplasm for resistance to spot blotch disease JARTS

Spot blotch caused by Bipolaris sorokiniana (Sacc.) Shoem. is the most devastating disease limiting wheat productivity in warm and humid environments. One hundred and fifty wheat genotypes were evaluated under field conditions in 2013 and 2014 in six different locations in Zambia. The genotypes showed different levels of resistance to spot blotch. Genotypes 19HRWSN6 (Kenya Heroe), 19HRWSN7 (Prontia Federal) and 19HRWSN15 (BRBT2/METSO) were resistant lines across environments. Marine bacteria are a rich source of bioactive metabolites. However, the microbial diversity of marine ecosystem still needs to be explored. The aim of this study was to isolate and characterize bacteria with antimicrobial activities from various marine coastal environment of New Caledonia. We obtained 493 marine isolates from various environments and samples of which 63 (12.8%) presented an antibacterial activity against a panel of reference pathogenic strains (Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli and Enterococcus faecalis). Debido al alto grado de contaminación por plaguicidas de productos agrícolas y al alto costo que implica su detección se consideró importante optimizar el método de escrutinio de inhibición de colinesterasas humanas en presencia de extractos vegetales. Se demostró que las suspensiones de glóbulos rojos y plasma son estables al menos por tres meses si se mantienen entre 4 y 8 °C. El reactivo de color es estable por al menos dos años y medio en refrigeración y los sustratos de ambas colinesterasas son estables congelados al menos por dos años.
