Characterization of the nucellus‑specific dehydrin MdoDHN11 demonstrates its involvement in the tolerance to water deficit. Repositório Alice
FALAVIGNA, V. da S.; MALABARBA, J.; SILVEIRA, C. P.; BUFFON, V.; MARIATH, J. E. de A.; PASQUALI, G.; MARGIS-PINHEIRO, M.; REVERS, L. F..
Abstract Key message MdoDHN11 acts in the nucellus layer to protect the embryo and the endosperm from limited water availability during apple seed development.AbstractDehydrins (DHNs) are protective proteins related to several plant developmental responses that involve dehy-dration such as seed desiccation and abiotic stresses. In apple (Malus × domestica Borkh.), the seed-specific MdoDHN11was suggested to play important roles against dehydration during seed development. However, this hypothesis has not yet been evaluated. Within this context, several experiments were performed to functionally characterize MdoDHN11. In situ hybridization analysis during apple seed development showed that MdoDHN11 expression is confined to a maternal tissue called nucellus,...

Cloning and expression of embryogenesis-regulating genes in Araucaria angustifolia (Bert.) O. Kuntze (Brazilian Pine) Genet. Mol. Biol.
Schlögl,Paulo Sérgio; Santos,André Luis Wendt dos; Vieira,Leila do Nascimento; Floh,Eny Iochevet Segal; Guerra,Miguel Pedro.
Angiosperm and gymnosperm plants evolved from a common ancestor about 300 million years ago. Apart from morphological and structural differences in embryogenesis and seed origin, a set of embryogenesis-regulating genes and the molecular mechanisms involved in embryo development seem to have been conserved alike in both taxa. Few studies have covered molecular aspects of embryogenesis in the Brazilian pine, the only economically important native conifer in Brazil. Thus eight embryogenesis-regulating genes, viz.,ARGONAUTE 1, CUP-SHAPED COTYLEDON 1, WUSCHEL-related WOX, S-LOCUS LECTIN PROTEIN KINASE, SCARECROW-like, VICILIN 7S, LEAFY COTYLEDON 1, and REVERSIBLE GLYCOSYLATED POLYPEPTIDE 1, were analyzed through semiquantitative RT-PCR during embryo development...

Establishment of post-harvest early-developmental categories for viability maintenance of Araucaria angustifolia seeds Acta Botanica
Araldi,Cristhyane Garcia; Coelho,Cileide Maria Medeiros.
Araucaria angustifolia seeds are recalcitrant, and their metabolism remains high during storage. This research aimed to describe the initiation of germination in A. angustifolia seeds during storage in order to standardize the assessment of physiological quality and to promote seed conservation. Seeds were collected from two populations and stored for 270 days in the natural laboratory environment and cold chamber. Seeds were classified according to four early developmental stages: I - mature seeds; II - seeds with elongation along the embryonic axis; III - beginning of root protrusion; IV - advanced germination stage, with seedling shoots. After categorization, physical and physiological quality was assessed. In freshly collected seeds, only category I...

Relation between Seed Appearance and Phenolic Maturity: A Case Study Using Grapes cv. Carménère Chilean J. Agric. Res.
Fredes,Claudio; Von Bennewitz,Eduardo; Holzapfel,Eduardo; Saavedra,Felipe.
Sensory evaluation of grapes (Vitis vinifera L.) plays a key role in determining the harvest time in grapevine varieties. The harvest time of cv. Carménère is one of the latest of Chile. During the season 2007-2008, the evolution of the appearance of ‘Carménère’ seeds was evaluated as a harvest criterion, comparing it with the chemical and phenolic ripening. The samples were obtained from an organic vineyard located in Curicó Valley, Chile. Starting at 16 ºBrix, 100 seed berries samples were collected weekly from medium vigor vines in order to register photographically the ventral and dorsal sides of each seed. In addition to the seed tannins percentage, the extractable anthocyanins, total anthocyanins and total polyphenols index, as well as the titratable...

Relationship between gibberellic acid and growth parameters in developing seed and pod of pigeon pea Braz. J. Plant Physiol.
Chudasama,Rita S.; Thaker,Vrinda S..
Changes in endogenous gibberellic acid (GA) levels were determined in developing seeds and pods of pigeon pea (Cajanus cajan). Antibodies against GA3 were raised in rabbits and indirect ELISA developed to estimate GA level. Two varieties, black seeded variety (V1) and B.D.N2 (V2), were selected on the basis of their seed index value. The pod length and number of seeds per pod were significantly different for the two varieties. Dry weight, water content (WC), rate of water accumulation, rate of dry matter accumulation (DMA), and GA content in pod and seed were all larger in V1 than in V2. In both varieties, higher GA content was observed during cell elongation and DMA phases, respective to their varietal differences in growth traits. Relationships between...

Transcriptome analyses of the Dof-like gene family in grapevine reveal its involvement in berry, flower and seed development. Repositório Alice
SILVA, D. C. da; FALAVIGNA, V. da S.; FASOLI, M.; BUFFON, V.; PORTO, D. D.; PAPPAS JÚNIOR, G. J.; PEZZOTTI, M.; PASQUALI, G.; REVERS, L. F..
The Dof (DNA-binding with one finger) protein family spans a group of plant transcription factors involved in the regulation of several functions, such as plant responses to stress, hormones and light, phytochrome signaling and seed germination. Here we describe the Dof-like gene family in grapevine (Vitis vinifera L.), which consists of 25 genes coding for Dof. An extensive in silico characterization of the VviDofL gene family was performed. Additionally, the expression of the entire gene family was assessed in 54 grapevine tissues and organs using an integrated approach with microarray (cv Corvina) and real-time PCR (cv Pinot Noir) analyses. The phylogenetic analysis comparing grapevine sequences with those of Arabidopsis, tomato, poplar and already...

Variations in sugars and cyclitols during development and maturation of seeds of brazilwood (Caesalpinia echinata Lam., Leguminosae) Braz. J. Plant Physiol.
Borges,Igor F.; Barbedo,Claudio J.; Richter,Andreas A.; Figueiredo-Ribeiro,Rita de Cássia L..
Seeds of Caesalpinia echinata are tolerant to desiccation, keeping their viability up to 0.08 g H2O g-1 DM. When stored under laboratory conditions they lose their viability within one month, while under low temperatures germination ability is maintained during two years of storage. Here, we report on the variations of soluble carbohydrates, cyclitols and cyclitol galactosides in seeds of C. echinata during development and maturation, aiming for a better understanding of the involvement of these compounds in the maturation process and seed quality. Individual flowers were tagged on the day of anthesis and pods were collected directly from the branches from 32 to 59 d after anthesis (DAA). Seeds were also collected after shedding. The physiological maturity...

Study of sunn hemp seed development and maturation was studied at the Department of Plant Science, Faculty of Natural Resources, Prince of Songkla University, Hat Yai campus, Hat Yai, Songkhla, during March-June, 2008. The blooming flowers were tagged to indicate the date of flowering. Pods at 10, 13, 16, 19, 22, 25, 28, 31, 34, 37, 40, 43 and 46 days after flowering were harvested to investigate pod color, seed color and seed quality. The results showed that the sunn hemp had 6 developmental stages of pod color which were green, yellow-green, light grey-green, dark greygreen, grey-brown and grey-brown-black at 10-16, 19-28, 31, 34, 37-40 and 43-46 days after flowering, respectively. The sunn hemp seeds were capable of germination at approximately 25 days...
