Bacillus megaterium: an endophytic bacteria from callus of Ilex paraguariensis with growth promotion activities. Repositório Alice

Endophytic microorganisms live inside healthy plants and their isolation and identification may favor biotechnological applications. We isolated endophytic bacteria from Ilex paraguariensis callus cultivated in vitro for five months. Sequencing of 16S rRNA region to identify two isolates as Bacillus megaterium was used. The presence of bacterial cells in the intercellular spaces of callus cultures was detected by ultra structural analyses. The isolates were also evaluated for indole acetic acid (IAA) production and their potential in plant growth promotion of Phaseolus vulgaris L. Bacterial biodiversity from an anaerobic up flow bioreactor with ANAMMOX activity inoculated with swine sludge BABT

The present study aimed to describe the bacterial community present at an anaerobic up flow bioreactor with ANAMMOX activity, inoculated with the sludge from the anaerobic pond of a swine slurry treatment system. The description was based on the molecular DNA techniques using primers for amplification of complete 16S rRNA gene and also new primers to amplify smaller fragments from 16S rRNA. During the bioreactor operation time, the bacterial community changed significantly, increasing the nitrogen removal efficiency, reaching after 500 days a removal rate of 94%. Borrelia burgdorferi sensu lato in humans in a rural area of Paraná State, Brazil BJM

This study describes the detection of Borrelia garinii and Borrelia burgdorferi sensu stricto (s.s.) in Brazilian individuals using PCR and DNA sequencing. Our results suggest that these species are emerging pathogens in this country, and additional studies are necessary to determine the epidemiological characteristics of this disease in Brazil.

Cloning and sequencing of c-DNA encoding N-methyl-D-aspartate receptor subunit-1 in the hypothalamus of male sheep R. Bras. Zootec.

The objective of the present study was to report the molecular cloning and determination of the sequences N-methyl-D-aspartate subunit-1 receptor (NMDAR-1) in the hypothalamus of sheep. The re-amplified DNA template for NMDA -1 from the hypothalamus of male sheep was cloned using pBluescript-Sk-plasmids (pBSK, 2958 bp). Purified plasmids containing the NMDA receptor c-DNA were sequenced using the dye-terminator chemistry at the University of Nebraska-Lincoln, Nebraska, USA. Avian Infectious Laryngotracheitis, caused by Infectious Laryngotracheitis Virus (ILTV), has been reported for decades in Brazilian laying and broiler flocks. More recently, outbreaks have occurred in São Paulo State. This study reports the application of PCR and DNA sequencing targeted to the p32 gene of ILTV using laying chicken samples from Bastos, São Paulo, Brazil. Three out of four field samples were positive by PCR. DNA sequencing of two samples evidenced homology of the amplified fragments with the p32 gene of ILTV. The results definitely confirmed the presence of ILTV in the birds during the outbreak. To explore the microbial diversity at deep sea vents, molecular techniques based on the analysis of the gene coding the 16S ribosomal RNA permitted to highlight a large archaeal and bacterial diversity. We hereby present the complete sequence and annotation of pRG930cm, a spectinomycin/streptomycin/chloramphenicol-resistant cosmid vector. pRG930cm (17,256 bp; GenBank Accession No.: FM174471) has a broad host range, and is stably maintained by a number of Gram-negative bacteria including Pseudomonas spp, Escherichia coli, Agrobacterium tumefaciens and Azorhizobium caulinodans ORS571. pRG930cm is already widely used and its sequence will aid efficient construction and analysis of cosmid libraries.

Genetic and taxonomic relationships of five species of Rallidae (Aves: Gruiformes) based on mitochondrial cytochrome oxidase subunit I sequences Animal Sciences

This is the first study to detection the genetic relationship between Porphyrio alleni and four Rallidae species: Fulica atra, Gallinulla angulata, Gallinulla chloropus and Porphyrio porphyrio. Mitochondrial cytochrome oxidase subunit I (COI) sequences were used as an effective marker in this study. DNA of Rallidae species were extracted, amplified using polymerase chain reaction (PCR) and then sequenced. The results obtained from information based on COI sequences revealed that Gallinulla angulata and Gallinulla chloropus fall into two separate clades and they are not monophyletic. Genetic characterization of dengue virus type 3 isolates in the State of Rio de Janeiro, 2001 BJMBR

The genetic characterization of dengue virus type 3 (DEN-3) strains isolated from autochthonous cases in the State of Rio de Janeiro, Brazil, in 2001 is presented. Restriction site-specific (RSS)-PCR performed on 22 strains classified the Brazilian DEN-3 viruses as subtype C, a subtype that contains viruses from Sri Lanka, India, Africa and recent isolates from Central America. Nucleic acid sequencing (positions 278 to 2550) of one DEN-3 strain confirmed the origin of these strains, since genotype III - classified by sequencing - and RSS-PCR subtype C are correlated. This genetic subtype has been associated with hemorrhagic dengue epidemics and the information provided here could be useful to implement appropriate prevention and control measures.

The aim of this work was to characterize rhizobia isolated from the root nodules of cowpea (Vigna unguiculata) plants cultivated in Amazon soils samples by means of ARDRA (Amplified rDNA Restriction Analysis) and sequencing analysis, to know their phylogenetic relationships. The 16S rRNA gene of rhizobia was amplified by PCR (polymerase chain reaction) using universal primers Y1 and Y3. Identification of a mutation in the spike protein cleavage site in Brazilian strains of wild-type bovine coronavirus BJM

The spike (S) protein of coronaviruses, a type I membrane glycoprotein, is primarily responsible for entry into susceptible cells by binding with specific receptors on cells and mediating subsequent virus-cell fusion. The bovine coronavirus (BCoV) S protein is cleaved into two subunits, the N-terminal S1 and the C-terminal S2. The proteolytic cleavage site of S protein is highly conserved among BCoV strains and is located between amino acids 763 and 768 (KRRSRR). Isolation of endophytic bacteria from arboreal species of the Amazon and identification by sequencing of the 16S rRNA encoding gene Genet. Mol. Biol.

Endophytic bacteria from three arboreal species native to the Amazon (Carapa guianenses, Ceiba pentandra, and Swietenia macrophylla), were isolated and identified, through partial sequencing of the 16S rRNA encoding gene. From these, 16 isolates were obtained, although, when compared to sequences deposited in GenBank, only seven had produced identifiable fragments. Bacillus, Pantoea and two non-culturable samples were identified. Results obtained through sequence analysis revealed low genetic diversity across the isolates, even when analyzing different species and plant structures. This is the first report concerning the isolation and identification of endophytic bacteria in these plant species.
