Abstract The purpose of the study was to develop a xenogenic bubaline diaphragm matrix (BDM) for abdominal hernia repair. A fresh diaphragm was decellularized using aqueous sodium dodecyl sulfate (SDS) solutions (0.5-4% w/v) over a period. Acellularity was confirmed histologically and characterized by Masson’s trichrome staining, scanning electron microscopy (SEM), DNA quantification, agarose gel electrophoresis, and Fourier-transform infrared spectroscopy. The BDM was used for clinical abdominal hernia repair in six cattle. Clinical, hemato-biochemical and antioxidant parameters were evaluated to assess biocompatibility of xenogenic BDM. Histologically, the diaphragm treated with 2% SDS for 48 h showed complete acellularity and orderly arranged collagen... |

Studies have proven that the agroforestry systems in the semi-arid region of the State of Ceará, Brazil, induce an increase in soil organic C levels. Notwithstanding, there is no information if this increase also results in qualitative changes in different pools of soil organic matter. The objective of this study was to verify the possible chemical and structural alterations in fulvic and humic acids of a Luvisol in areas adopting agroforestry, traditional intensive cultivation and native forest in a long-term experiment conducted in the semi-arid region of Ceará State, Brazil. The study was conducted in an experimental area of the National Goat Research Center (Embrapa) in Sobral, CE. The following treatments were evaluated: agrosilvopasture (AGP),... |

High resolution proton nuclear magnetic resonance spectroscopy (¹H MRS) can be used to detect biochemical changes in vitro caused by distinct pathologies. It can reveal distinct metabolic profiles of brain tumors although the accurate analysis and classification of different spectra remains a challenge. In this study, the pattern recognition method partial least squares discriminant analysis (PLS-DA) was used to classify 11.7 T ¹H MRS spectra of brain tissue extracts from patients with brain tumors into four classes (high-grade neuroglial, low-grade neuroglial, non-neuroglial, and metastasis) and a group of control brain tissue. PLS-DA revealed 9 metabolites as the most important in group differentiation: γ-aminobutyric acid, acetoacetate, alanine,... |

A simple and rapid method for the determination of nitrate in seawater of high nitrate content (>1 ppm) is proposed. The absorption of the nitrate ion is measured at 210 and 220 mµ. The sample is treated with a measured quantity of hydrochloric acid to eliminate the carbonate and bicarbonate interferences and measured against a nitrate free blank prepared by reduction of the nitrate to ammonia with Raney nickel. The addition of HCl permits also storage of the sample without special precautions and the use of the blank eliminates the most serious of the interferences (bromide) expected in seawater. The interference of organic matter limits however the applicability of the method to concentrations higher than 1 ppm. |

DELVOYE, L., 1992. Endolithic algae in living stony corals: Algal concentrations under influence of depth-dependent light conditions and coral tissue fluorescence in Agaricia agaricites (L) and Meandrina meandrites (L.) (Sclereactinia, Anthozoa). Studies Nat. Hist. Caribbean Region 71, Amsterdam 1992: 24-41. The relation between Scleractinians and their endolithic algae was studied in the depth range of 10 to 35 m in Curaçao. Endolithic algal concentrations are found in the skeleton under the living tissue of stony corals and never in dead parts or in coral rubble. The influences of depth-dependent light conditions and coral tissue fluorescence on endolithic algal concentrations were studied in a non-fluorescent and a fluorescent form of both Agaricia... |

Abstract The potential astringency of eight grape cultivars was evaluated based on polyphenol/protein binding. The parameters such as weights, sizes, total phenols, flavanols and tannins of different grape tissues were determined, and the polyphenol/protein binding was analyzed by SDS-PAGE and spectroscopy. The Cabernet Sauvignon had the highest skin proportion, while the Blue French had the highest seed proportion. The Blue French seeds had the highest total phenolic content, while the Cabernet Sauvignon skins had the highest total phenolic content. SDS-PAGE showed that the seed extracts had a better affinity than skin extracts for saliva proteins, but fluorescence spectra demonstrated that the interactions of skin polyphenols with protein were more... |

Leptospirosis caused by Leptospira interrogans is the most widespread zoonosis and a major public health problem worldwide. Based on light-scattering and absorption, quantification of leptospires using UV-VIS spectroscopy was used as an indirect counting technique by measuring the optical density and comparing this to automated direct counting using a counting chamber in combination with imaging and analyzing software. Two serovars, Bangkok and Copenhagenii, from log-phase growth were used for the establishment of standard curves. They were found to be linear and slightly different in gradient for each serovar. The ease, rapidity, and reliability of these two adapted and optimized counting techniques may provide a useful alternative enumeration technique... |

The capability of near infrared reflectance spectroscopy (NIRS) was evaluated to predict the content of total ash (TA), crude protein (CP), crude fiber (CF), neutral detergent fiber (NDF), acid detergent fiber (ADF) and metabolizable energy (ME); as well as pH and ammonia nitrogen content (N-NH3), in pasture silage, with and without additives. Nine hundred and twenty dried and ground samples of pasture silage, with known chemical composition, were scanned over the visible and NIR region (400 to 2500 nm) at 2 nm intervals. Calibration equations were developed by modified partial least square regression models (MPLS) with different mathematical treatments and light scatter correction as standard normal variation and Detrend (SNV & D) of the spectra.... |

ABSTRACT The actinobacterial strain Streptomyces lavendulocolor VHB-9 was isolated from granite mine soil samples of Khammam district, Telangana state, India. The strain was identified based on detailed microorphological, cultural and phylogenetic analysis. Bioactive guided isolation of the secondary metabolites of the strain was carried out by growing the strain in optimized medium (0.5% lactose, 0.5% peptone, 0.05% K2HPO4, 0.2% CaCO3 with pH adjusted to 7.0). Separation and purification of the active fractions from the crude ethyl acetate extract was carried out by silica gel column chromatography and resulted in the isolation of two active fractions. Structural elucidation of the two (B2 and B3) active compounds was carried out by FT-IR, Mass and NMR... |

Among marine algae species, Alexandrium minutum produces a phycotoxin called paralytic shellfish poisoning (PSP) that is introduced in the food chain through the ingestion of phytoplankton by shellfishs, and later by human consumers. Thus, in situ monitoring of A. minutum proliferation in coastal seawater is of great economical importance for marine resources exploitation. Here, we propose a rapid test for the detection of A. minutum by surface plasmon resonance spectroscopy. First, whole genomic DNA is extracted from the algae. Second, a 677 bp long portion of the 28S ribosomal DNA is amplified by PCR. Third, the PCR product is detected by surface plasmon resonance spectroscopy onto a DNA functionalized gold substrate. |
