Bacterioplankton features and its relations with doc characteristics and other limnological variables in Paraná river floodplain environments (PR/MS-Brazil) BJM
Teixeira,Mariana Carolina; Santana,Natália Fernanda; Azevedo,Júlio César Rodrigues de; Pagioro,Thomaz Aurélio.
Since the introduction of the Microbial Loop concept, many studies aimed to explain the role of bacterioplankton and dissolved organic carbon (DOC) in aquatic ecosystems. Paraná River floodplain system is a very complex environment where these subjects were little explored. The aim of this work was to characterize bacterial community in terms of density, biomass and biovolume in some water bodies of this floodplain and to verify its temporal variation and its relation with some limnological variables, including some indicators of DOC quality, obtained through Ultraviolet-visible (UV-VIS) and fluorescence spectroscopic analysis. Bacterial density, biomass and biovolume are similar to those from other freshwater environments and both density and biomass were...

Chemical and structural characterization of soil humic substances under agroforestry and conventional systems Rev. Bras. Ciênc. Solo
Moraes,Gislane M. de; Xavier,Francisco Alisson da Silva; Mendonça,Eduardo de Sá; Araújo Filho,João Ambrósio de; Oliveira,Teógenes Senna de.
Studies have proven that the agroforestry systems in the semi-arid region of the State of Ceará, Brazil, induce an increase in soil organic C levels. Notwithstanding, there is no information if this increase also results in qualitative changes in different pools of soil organic matter. The objective of this study was to verify the possible chemical and structural alterations in fulvic and humic acids of a Luvisol in areas adopting agroforestry, traditional intensive cultivation and native forest in a long-term experiment conducted in the semi-arid region of Ceará State, Brazil. The study was conducted in an experimental area of the National Goat Research Center (Embrapa) in Sobral, CE. The following treatments were evaluated: agrosilvopasture (AGP),...

Classification of brain tumor extracts by high resolution ¹H MRS using partial least squares discriminant analysis BJMBR
Faria,A.V.; Macedo Jr.,F.C.; Marsaioli,A.J.; Ferreira,M.M.C.; Cendes,F..
High resolution proton nuclear magnetic resonance spectroscopy (¹H MRS) can be used to detect biochemical changes in vitro caused by distinct pathologies. It can reveal distinct metabolic profiles of brain tumors although the accurate analysis and classification of different spectra remains a challenge. In this study, the pattern recognition method partial least squares discriminant analysis (PLS-DA) was used to classify 11.7 T ¹H MRS spectra of brain tissue extracts from patients with brain tumors into four classes (high-grade neuroglial, low-grade neuroglial, non-neuroglial, and metastasis) and a group of control brain tissue. PLS-DA revealed 9 metabolites as the most important in group differentiation: γ-aminobutyric acid, acetoacetate, alanine,...

Determination of nitrate ion in marine biotopes with high nitrate content by ultraviolet spectrophotometry OMA
Mertens, J.; Massart, D.L..
A simple and rapid method for the determination of nitrate in seawater of high nitrate content (>1 ppm) is proposed. The absorption of the nitrate ion is measured at 210 and 220 mµ. The sample is treated with a measured quantity of hydrochloric acid to eliminate the carbonate and bicarbonate interferences and measured against a nitrate free blank prepared by reduction of the nitrate to ammonia with Raney nickel. The addition of HCl permits also storage of the sample without special precautions and the use of the blank eliminates the most serious of the interferences (bromide) expected in seawater. The interference of organic matter limits however the applicability of the method to concentrations higher than 1 ppm.

Leptospirosis research: fast, easy and reliable enumeration of mobile leptospires Biol. Res.
SCHREIER,STEFAN; TRIAMPO,WANNAPONG; DOUNGCHAWEE,GALAYANEE; TRIAMPO,DARAPOND; CHADSUTHI,SUDARAT.
Leptospirosis caused by Leptospira interrogans is the most widespread zoonosis and a major public health problem worldwide. Based on light-scattering and absorption, quantification of leptospires using UV-VIS spectroscopy was used as an indirect counting technique by measuring the optical density and comparing this to automated direct counting using a counting chamber in combination with imaging and analyzing software. Two serovars, Bangkok and Copenhagenii, from log-phase growth were used for the establishment of standard curves. They were found to be linear and slightly different in gradient for each serovar. The ease, rapidity, and reliability of these two adapted and optimized counting techniques may provide a useful alternative enumeration technique...

Morphological Interpretation of Reflectance Spectrum (MIRS) using libraries looking towards soil classification Scientia Agricola
Demattê,José Alexandre Melo; Bellinaso,Henrique; Romero,Danilo Jefferson; Fongaro,Caio Troula.
The search for tools to perform soil surveying faster and cheaper has led to the development of technological innovations such as remote sensing (RS) and the so-called spectral libraries in recent years. However, there are no studies which collate all the RS background to demonstrate how to use this technology for soil classification. The present study aims to describe a simple method of how to classify soils by the morphology of spectra associated with a quantitative view (400-2,500 nm). For this, we constructed three spectral libraries: (i) one for quantitative model performance; (ii) a second to function as the spectral patterns; and (iii) a third to serve as a validation stage. All samples had their chemical and granulometric attributes determined by...

SENSOR SYSTEMS FOR MAPPING SOIL FERTILITY ATTRIBUTES: CHALLENGES, ADVANCES, AND PERSPECTIVES IN BRAZILIAN TROPICAL SOILS REA
Molin,José Paulo; Tavares,Tiago Rodrigues.
ABSTRACT Soil fertility attributes have different scales and forms of spatial and temporal variations in agricultural fields. Adequate spatiotemporal characterization of these attributes is fundamental to the successful development of strategies for variable rate application of fertilizers, enabling the classic benefits of precision agriculture (PA). Studies on Brazilian soil have shown that at least 1 sample ha-1 is required for the reliable mapping of key fertility attributes. However, this sampling density is difficult owing to the operational challenges of sample collection and the cost of laboratory analyses. Given this limitation, soil sensors have emerged as a practical and complementary technique for obtaining information on soil attributes, at...

Toward Detection of Harmful Algae Blooms by in situ Surface Plasmon Resonance Spectroscopy ArchiMer
Laurent, Sebastien; Colas, Florent; Hamelin, Muriel; Crassous, Marie-pierre; Antoine, Elisabeth; Lehaitre, Michel; Compere, Chantal.
Among marine algae species, Alexandrium minutum produces a phycotoxin called paralytic shellfish poisoning (PSP) that is introduced in the food chain through the ingestion of phytoplankton by shellfishs, and later by human consumers. Thus, in situ monitoring of A. minutum proliferation in coastal seawater is of great economical importance for marine resources exploitation. Here, we propose a rapid test for the detection of A. minutum by surface plasmon resonance spectroscopy. First, whole genomic DNA is extracted from the algae. Second, a 677 bp long portion of the 28S ribosomal DNA is amplified by PCR. Third, the PCR product is detected by surface plasmon resonance spectroscopy onto a DNA functionalized gold substrate.

Usefulness of near infrared reflectance (NIR) spectroscopy and chemometrics to discriminate between fishmeal, meat meal and soya meal samples Ciencia e Investigación Agraria
Cozzolino,Daniel; Restaino,Ernesto; La Manna,Alejandro; Fernandez,Enrique; Fassio,Alberto.
Near infrared reflectance (NIR) spectroscopy was used in combination with chemometrics to discriminate between fishmeal, meat meal and soya meal samples. Samples were obtained from commercial feed miles and scanned in the NIR region (1100 - 2500 nm) in a monochromatic instrument in reflectance mode. Principal component analysis (PCA) and linear discriminant analysis were used to classify samples based on their NIR spectra. Full cross-validation was used in the development of classification models. Partial least squares-discriminant analysis (PLS-DA) correctly classified 85.7% of the fishmeal samples and 100% of the meat meal and soya meal samples. These results demonstrate the usefulness of NIR spectra combined with chemometrics as an objective and rapid...

Utilizing visible and near infrared spectroscopy based on multi-class support vector machines classification to characterize olive oil adulteration CIGR Journal
Ghasemi-Varnamkhasti, Mahdi; Amini-Pozveh, Samaneh; Mireei, Seyed Ahmad; Mishra, Puneet; Ghosh, Satyabrata; Ghanbarian, Davoud; Izadi, Zahra.
Rapid and non-destructive adulteration detection is of particular importance to oil industries. This paper presents an application of visible and near-infrared spectroscopy (VNIR) for detection of adulteration levels in olive oil. Sunflower oil was used as an adulterant to olive oil and adulteration samples with different levels ranging from 0 to 40% were prepared and used for the experiments. The spectra were first considered in the range of 500-900 nm and then smoothened and normalized to reduce the light scattering effects. Principal component analysis (PCA) was performed on the spectra to have a primary data visualization and feature extraction. The extracted PCA scores were used to calculate the Mahalanobis distances of the adulterated samples from...

VIS/NIR spectral signature for the Identification of Peanut Contamination of Powder Foods CIGR Journal
Ghosh, Satyabrata; Domínguez, Teresa R. Cuadrado; Diezma, Belén; Lleó, Lourdes; Barreiro, Pilar; Lacarra, Teresa García; Roger, Jean-Michel.
Visible-Near Infrared reflectance spectra are proposed for the characterization IRMM 481 peanuts in comparison to powder food materials: wheat flour, milk and cocoa. Multidimensional analysis of spectra of powder samples shows a specific NIR band centred at 1200 nm that identifies peanut compared to the rest of food ingredients, regardless compaction level and temperature. Spectral range 400-1000 nm is not robust for identification of blanched peanut. The visible range has shown to be reliable for the identification of pre-treatment and processing of unknown commercial peanut samples. A spectral index is proposed based on the combination of three wavelengths around 1200 nm that is 100% robust against pre-treatment (raw or blanched) and roasting (various...
