Biological characteristics of European hake (Merluccius merluccius) sperm ArchiMer

Very little is known on European hake's reproductive biology and especially on biological characteristics of its sperm despite a growing interest in its aquaculture potential. This study reports, for the first time some hake sperm characteristics. After activation, the swimming phase lasts 3 min (8 min when activated with 50% sea water (SW) but lower initial velocity). The initial flagellar beat frequency (BF), velocity and percentage of motile cells decreased after 100 s. After 2 days at 4 degrees C, the mean Adenylate Energy Charge (AEC) level was 0.71. Sperm stored at 4 degrees C still showed motility after 9 days for 2 individuals. When cryopreserved, the motility recovery index of thawed spermatozoa ranged from 11.8% to 29.6%.

Cinética de espermatozoides criopreservados de bovinos após sexagem por citometria de fluxo. Repositório Alice

Resumo - O objetivo deste estudo foi comparar a cinética de sêmen bovino criopreservado não sexado, sexado X e sexado Y antes e depois da seleção espermática por gradiente de Percoll. Amostras criopreservadas de sêmen não sexado (grupo NS) e sexado X (grupo SX) e Y (grupo SY) por citometria de fluxo, de quatro touros, foram avaliadas quanto à motilidade e à cinética espermática com o "computer-assisted semen analysis" (CASA) e o restante da amostra de cada grupo foi submetido à seleção espermática em gradiente de Percoll (45:60%). Após a seleção, foram realizadas as mesmas avaliações que antes da passagem pelo Percoll. A motilidade do grupo NS foi superior à dos grupos SX e SY e não foi observada diferença entre os grupos SX e SY nos parâmetros de cinética...

Colesterol carregado pela ciclodextrina sobre a criopreservação dos espermatozoides de garanhões da raça Nordestina. Repositório Alice

Nesta pesquisa avaliou-se o efeito do colesterol sobre o sêmen de garanhões da raça Nordestina sobre a qualidade espermática. Vinte ejaculados de dois aranhões foram diluídos com BotuSemen e colesterol carreado pela ciclodextrina (CCC) adicionado no sêmen: controle, 0,75mg de CCC e 1,0mg de CCC/120x106 sptz/mL, e incubado a 26°C/15min. O sêmen foi diluído 1:5 (v/v) com diluente Lactose-gema de ovo e resfriado a 5°C/2h, envasado em palhetas de 0,5mL, e acondicionado sob vapor de nitrogênio líquido, e depois imersos. As amostras foram descongeladas (37 °C/30s) e avaliadas. As variáveis foram avaliadas com ANOVA e teste de Tukey (P<0,05). Artificial insemination mastery in red drum, Sciaenops ocellatus, requires a first experimental step wOOng at getting basic biological data. Regarding males, a relation between optical density of the solution (OD) measured by spectrophotometry and sperm concentration (SC) has been established : OD O,161*SC + 0,119. Mean sperm concentration measured is 4,36.10 10 spz.mL-1. This study shows an influence of the solution of dilution on spermatozoa motility and its evolution during short term preservation. Cyclic nucleotide phosphodiesterase inhibition increases tyrosine phosphorylation and hyper motility in normal and pathological human spermatozoa Biocell

Our objective was to determine the effect of phosphodiesterase (PDE) inhibition on: 1) tyrosine phosphorylation of human spermatozoa at the tail level; and 2) sperm motion parameters and hyperactivated motility. The study was conducted with normozoospermic and asthenozoospermic samples incubated under in vitro capacitating conditions. The main outcome measures were computer-assisted sperm motion analysis and fluorescent immunodetection of phosphotyrosine-containing proteins. Pentoxifylline (PTX) was used as PDE inhibitor because of its wide use in the clinic. This study evaluated the semen quality of Santa Ines sheep fed diets containing different levels of whole cottonseed (WCS). Twenty sheep were distributed into four groups that were given diets with 0; 20; 30; 40% of WCS. For this study the semen was collected by electroejaculation, and a monthly andrological examination was performed during nine months. The analyzed variables were: scrotal circumference (SC), appearance and color of the ejaculate, progressive motility (PM), vigor, mass movement (MM), total sperm concentration (spzX109), sperm morphology (SM) and blood hematocrit. In the Pacific oyster, spermatozoa are characterized by a remarkably long movement phase (i.e., over 24 h) sustained by a capacity to maintain intracellular ATP level. To gain information on oxidative phosphorylation (OXPHOS) functionality during the motility phase of Pacific oyster spermatozoa, we studied 1) changes in spermatozoal mitochondrial activity, that is, mitochondrial membrane potential (MMP), and intracellular ATP content in relation to motion parameters and 2) the involvement of OXPHOS for spermatozoal movement using carbonyl cyanide m-chlorophenyl hydrazone (CCCP). Motility characteristics of spermatozoa in cod (Gadus morhua) and hake (Merluccius merluccius) ArchiMer

As in many marine fish, motility is triggered by osmotic pressure (OP) in cod and hake spermatozoa and motility ranges several minutes with a high velocity (above 50 mu m/s) period restricted to 100 s. A decrease in the flagellar beat frequency (BF) is partly responsible of this briefness. Detailed observations of the flagella during the motility period showed: 1) sea water provokes OP damages (blebs) which impair the correct wave propagation 2) waves become restricted to the proximal flagellum and tip becomes devoid of wave 3) the wave amplitude decreases. The combination of these factors drastically limits to the earliest period of motility the ability for spermatozoa to efficiently progress towards egg for fertilization.

Sperm features in turbot (Scophthalmus maximus): a comparison with other freshwater and marine fish species ArchiMer

This paper presents some features of turbot sperm and compares them with data reported in the literature on other freshwater and marine fish. In captivity, turbot spermiation period lasts 6 months. When compared to other fish species, turbot males are characterized by low values of maximum gonadosomatic index (0.6-0.8%) and are poor sperm producers in terms of volume (0.2-2.2 ml), concentration (0.7-11.0 x 10(9) spermatozoa/ml) and total sperm number released per stripping (0.2-12.0 x 10(9) spermatozoa). Compared to most freshwater species, the duration of cell movement is long (1-17 min). The spermatozoon of turbot can be considered to be of a primitive type. Sperm Motility and Viability in West African Dwarf Rams Treated with Euphorbia hirta International Journal of Morphology

Four adult sexually matured and clinically healthy West African Dwarf (WAD) rams aged between 24 and 30 months were used for the study. The rams were first used as control and later as experimental animals upon being orally dosed with Euphorbia hirta extract at 400mg/kg body weight for 14 days. Semen samples were collected from the rams a day after the administration of the plant extra and seven days after. The objective of the study was to investigate the effect of Euphorbia hirta on the semen picture of WAD rams. Sperm motility in European hake, Merluccius merluccius, and characterization of its spermatozoa concentration and volume, spermatocrit, osmolality and pH ArchiMer

Due to stock declines there is a growing interest in farming of European hake Merluccius merluccius. Thus, knowledge of its sperm biology is of importance not only for purposes of broodstock management, but also for the development of sperm preservation techniques. Hake sperm were collected from mature males caught during the summer-early autumn waters off western Norway and during the winter-early spring in the Bay of Biscay (France). Sperm quality characteristics were assessed after storage at 4 degrees C for 25 +/- 14 h.
